    Notes: Summary In Rivulus marmoratus development of retinal pigmented epithelium (RPE) parallels that of retinal photoreceptors. Although structurally functional by mid-incubation the full complex structure is not achieved even when the yolk-sac is absorbed (3-days post-hatched). Melanogenesis is evident at 0.2 incubation with premelanosomes present up to three days after hatching. The distribution of junctional complexes, basal membrane foldings and coatedpits throughout development is noted. Myeloid bodies, already present at mid-incubation, appear initially as single lenticular-shaped structures which later may form whorls, or coalesce around oil droplets, glycogen clusters or phagosomes thereby giving rise to myeloid patterns characteristic of a mature RPE. The functional significance of these changes is discussed.

    Notes: Abstract Intramolecular excimer formation of 1,3-di(1-pyrenyl)propane (Py-3-Py) and fluorescence polarization of 1,6-diphenyl-1,3,5-hexatriene (DPH) were used to evaluate the effect of ethanol on the rate and range of the lateral mobility and the range of the rotational mobility of bulk bilayer structures of the plasma membrane vesicles (ATCC-PMV) isolated from cultured hybridoma cells (ATCC TIB 216). In a concentration-dependent manner, ethanol increased the excimer to monomer fluorescence intensity ratio (I′/I) of Py-3-Py in the ATCC-PMV and decreased the anisotropy (r), limiting anisotropy (r∞) and order parameter (S) of DPH in the ATCC-PMV. This indicates that ethanol increased both the lateral and rotational mobility of the probes in the ATCC-PMV. Selective quenching of DPH by trinitrophenyl groups was utilized to examine the range of transbilayer asymmetric rotational diffusion of the ATCC-PMV. The anisotropy (r), limiting anisotropy (r ∞) and order parameter (S) of DPH in the inner monolayer were 0.024, 0.032, and 0.069, respectively, greater than calculated for the outer monolayer of the ATCC-PMV. Selective quenching of DPH by trinitrophenyl groups was also used to examine the transbilayer asymmetric effects of ethanol on the range of the rotational mobility of the ATCC-PMV. Ethanol had a greater increasing effect on the range of the rotational mobility of the outer monolayer as compared to the inner monolayer of the ATCC-PMV. It has been proven that ethanol exhibits a selective rather than nonselective fluidizing effect within the transbilayer domains of the ATCC-PMV.

To study K+ channels in the basolateral membrane of chloride-secreting epithelia, rat tracheal epithelial monolayers were cultured on permeable filters and mounted into an Ussing chamber system. The mucosal membrane was permeabilized with nystatin (180 μg/ml) in the symmetrical high K+ (145 mm) Ringer solution. During measurement of the macroscopic K+ conductance properties of the basolateral membrane under a transepithelial voltage clamp, we detected at least two types of K+ currents: one is an inwardly rectifying K+ current and the other is a slowly activating outwardly rectifying K+ current. The inwardly rectifying K+ current is inhibited by Ba2+. The slowly activating K+ current was potentiated by cAMP and inhibited by clofilium, phorbol 12-myristae 13-acetate (PMA) and lowering temperature. This is consistent with the biophysical characteristics of I SK channel. RT-PCR analysis revealed the presence of I SK cDNA in the rat trachea epithelia. Although 0.1 mm Ba2+ only had minimal affect on short-circuit current (I sc) induced by cAMP in intact epithelia, 0.1 mm clofilium strongly inhibited it. These results indicate that I SK might be important for maintaining cAMP-induced chloride secretion in the rat trachea epithelia.

    Notes: Summary The effect of isotretinoin on experimentally induced comedones was studied by means of histoplanimetry and scanning electron microscopy (SEM). To induce comedone formation, insoluble cutting oil was applied to the ventral surface of the ears of rabbits for 2 weeks. After comedones formed, isotretinoin was fed to the rabbits by a feeding tube, daily, for 4 weeks. A low dose (2.0 mg/kg) and a high dose (20.0 mg/kg body weight) were selected for two different groups. Soybean oil served as the vehicle. Histoplanimetrically, the high-dose group showed a significant decrease in size of comedones when compared to the control group and the low-dose group [P〈0.005], and in the high-dose group, the follicular lumen contained a small number of loose, non-adherent, horny cells. Upon SEM examination, the comedones were seen to have a ‘chrysanthemum’ appearance before treatment. After treatment, the high-and low-dose groups both showed evidence of the elimination of comedones on three-dimensional analysis.

    Notes: Summary Tuberculomas of the brain are rare in the United States. Although CAT (computerized axial tomography) scanning cannot distinguish these infections from other intracranial mass lesions, it provides a non-invasive technique with which to follow the course of presumptive lesions during medical therapy, and with which to search for clinically unsuspected infections. The necessity for and timing of surgery can be considered more easily with this technique. Two cases are presented with their CAT scans, including one example of the disappearance of a presumptive tuberculoma, during appropriate medical therapy.

    Notes: We report molecular dynamics simulations of LiCl, NaBr, and CsBr in supercritical water in order to explain the experimental observations of the limiting conductances as a function of the density of water at supercritical state points. As was the case in our previous work on NaCl in supercritical water [Lee et al., Chem. Phys. Lett. 293, 289 (1998)], we find that the experimental trends in the limiting conductances as a function of water density are reproduced in our simulations—a clear change of slope from the assumed linear dependence of limiting conductances of LiCl, NaBr, and CsCl on the water density. We also found that the effect of the number of hydration water molecules around ions dominates in the higher-density region while the interaction strength between the ions and the hydration water molecules (as measured by the potential energy per hydration water molecule) dominates in the lower-density region. In the case of Cs+ and Br−, however, the latter factor in the lower-density region is not as dominant as in the cases of Na+ and Cl− since a clear difference between the potential energy per hydration water molecule at densities above and below 0.45 g/cm3 was not clearly observed in the cases. In the case of Li+, the interaction between the ions and the hydration water in the lower-density region is almost a nonfactor since the potential energy per hydration water molecule is monotonically decreased with decreasing water density, which is consistent with the linear increase of the limiting conductance for the Li+ ion with decreasing water density. © 2000 American Institute of Physics.
