    Description / Table of Contents: Résumé La synthèse et la sécrétion de la glycoprotéine matricielle de la dentine sont étudiées par radioautographie des odontoblastes et de la dentine d'incisives de rats. Ces derniers reçoivent, par voie intraveineuse, du3H-fucose. A des intervalles de temps variés, ils sont sacrifiés par perfusion intra-cardiaque de glutaraldéhyde. Les radioautographies de specimens démineralisés sont préparées pour la microscopie optique et électronique. Des grains d'argent se situent au niveau de l'appareil de Golgi 5–10 minutes après injection, mais sont absents dans les autres régions. A 35 minutes, les grains sont visibles à la base du prolongement odontoblastique et dans les ramifications voisines, en association avec des granules allongés, limités par une membrane: quelques grains se localisent dans la matrice prédentinaire adjacente. A 4 heures, on observe une réaction de la jonction dentine-prédentine, où un matériel granulaire dense est associé aux fibres de collagène. Ces résultats confirment que l'odontoblaste synthétise une glycoprotéine contenant du fucose, et indiquent que le3H-fucose est ajouté a la glycoprotéine en voie de formation dans l'appareil de Golgi: cette glycoprotéine migre ensuite dans le prolongement odontoblastique, puis est déchargée dans la matrice. Une certaine quantité de glycoprotéine s'accumule dans la région où la prédentine se transforme en dentine. Ce matériel granulaire observé à ce niveau, pourrait indiquer un dépôt de glycoprotéine. Comme cette région coincide avec le front de minéralisation, le dépôt de glycoprotéine marquée pourrait coincider avec le début de cette minéralisation.
    Abstract: Zusammenfassung Synthese und Sekretion des Glucoproteins der Dentinmatrix wurden anhand von Autoradiographien der Odontoblasten und des zugehörigen Dentins an Rattenschneidezähnen untersucht. Zu diesem Zweck erhielten die Ratten3H-Fucose i.v. An verschiedenen Zeitpunkten danach wurden sie durch intrakardiale Perfusion mit Glutaraldehyd getötet. Autoradiographien von entkalkten Proben wurden anschließend für Licht-und EM-Autoradiographie präpariert. 5–10 min nach Injektion ist der Golgi-Apparat mit Silberkörnern bedeckt; die übrigen Regionen der Odontoblasten und des umgebenden Prädentins und Dentins sind jedoch frei davon. Nach 35 min werden die Körner auch über der Basis der Odontoblastenfortsätze und ihrer nächsten Verzweigungen beobachtet; sie sind verbunden mit membranbegrenzten, dort lokalisierten, länglichen Granula. Einige wenige Körner befinden sich über der angrenzenden Prädentinmatrix. Nach 4 Std zeigt sich über der Matrix eine Reaktion auf der Dentinseite der Prädentin-Dentin-Berührungszone, wo ein elektronenundurchlässiges, granuläres Material mit kollagenen Fasern verbunden vorliegt. Mit diesen Beobachtungen kann belegt werden, daß die Odontoblasten fucosehaltiges Glucoprotein synthetisieren und sie weisen darauf hin, daß sich3H-Fucose mit dem sich bildenden Glucoprotein im Golgi-Apparat verbindet. Dieses Glucoprotein wandert später zu den Odontoblastenfortsätzen und wird dann an die Matrix abgegeben. Ein großer Teil des Glucoproteins sammelt sich an jener Stelle an, wo Prädentin zu Dentin umgewandelt wird. Beim hier beobachteten, granulären Material kann es sich um den morphologischen Beweis für das abgelagerte Glucoprotein handeln. Da die Verkalkungsgrenze sich an dieser Stelle befindet, kann die Ablagerung von markiertem Glucoprotein mit der beginnenden Verkalkung in Zusammenhang gebracht werden.
    Notes: Abstract The synthesis and secretion of the matrix glycoprotein of dentin was investigated by radioautography of the odontoblasts and associated dentin in the rat incisor. To this end, rats received3H-fucose intravenously. At various times thereafter, they were sacrificed by intracardiac perfusion with glutaraldehyde. Demineralized specimens were then prepared for light and electron microscope autoradiography. Silver grains overlie the Golgi apparatus 5–10 min after injection, but are absent over other regions of the odontoblasts and adjacent predentin and dentin. By 35 min, grains are observed over the base of the odontoblastic processes and their nearby branches, in association with membrane-limited elongated granules located therein; a few grains are located over the adjacent predentin matrix. At 4h, a reaction is seen over the matrix on the dentin side of the predentin-dentin junction where an electron-dense granular material is associated with collagen fibers. These findings provide evidence that odontoblasts synthesize fucose-containing glycoprotein, and indicate that3H-fucose is added to forming glycoprotein in the Golgi apparatus; this glycoprotein later migrates to the odontoblastic processes and is then discharged into the matrix. Much of the glycoprotein accumulates at the site where predentin transforms into dentin. The granular material observed at this location may be morphological evidence of the glycoprotein being deposited. Since this is also the site of the mineralization front, the deposition of labeled glycoprotein may be related to the onset of mineralization.
