    Notes: Summary The fluorescent spectra of whole broth, cell-free broth and resuspended cells were compared during the fermentation ofCandida utilis growing in ethanol for the purposes of identifying the location of monitorial fluorophores in cellular systems. Four cellular fluorophores, tryptophan, pyridoxine, NADH and riboflavin were examined. The results indicated that the fluorescence signal of tryptophan and NADH came mainly from intracellular fluorophores, and the fluorescence signal of pyridoxine and riboflavin mainly came from extracellular fluorophores. The contributions of intracellular and extracellular fluorophores to culture fluorescence signals were found to change during the fermentation.

    Notes: Summary Mixing times and gas hold-ups in a 250L bioreactor containing a phosphate buffer or an active fermentation were determined on-line using fluorometric (MEFS and NADH) probes as functions of agitation and aeration rates. Both mixing time and hold-up of a fermentaion can be determined using a MEFS probe. Hold-up may also be measured with a presently available commercial NADH probe.

    Notes: Summary The kinetic and fluorometric behavior ofPseudomonas putida (ATCC 17484) growing on phenol as the carbon limiting substrate was investigated. A model was developed to describe the behavior under dynamic conditions of flow or substrate concentration “shift-up”. Parameters contained in the model were estimated. It was observed that the model described the kinetic behavior reasonably well. Factors which affected the fluorescence output of the whole broth culture were also studied.
