    Notes: Abstract Plant cells of tobacco (Nicotiana tabacum L.) were grown for several generations in suspension cultures. Cells were immobilized in continuous bioreactors in calcium alginate (Ca Alg) beads or in poly-L-lysine (PLL) encapsulated calcium alginatehydrogels. In each case, the cells were fed continuously a modified Linsmaier-Skoog plant cell culture medium. The bioreactor effluent was analyzed for total phenolic compounds. The net specific productivity of phenolics was calculated on a daily basis for several test runs. For comparison, productivity in suspension cultures was monitored. Productivity of suspended cells declined to zero within 9 d; both immobilized and encapsulated cells remained productive for 16 d following inoculation. Specific productivity by encapsulated cells was higher than that by immobilized cells; in both types similar rates of decline in productivity occurred.

    Notes: Summary Fed-batch fermentations of Acidothermus cellulolyticus utilizing mixtures of cellulose and sugars were investigated for potential improvements in cellulase enzyme production. In these fermentations, we combined cellulose from several sources with various simple sugars at selected concentrations. The best source of cellulose for cellulase production was found to be ball-milled Solka Floc at 15 g/l. Fed-batch fermentations with cellobiose and Solka Floc increased cell mass only slightly, but succeeded in significantly enhancing cellulase synthesis compared to batch conditions. Maximum cellulase activities obtained from fermentations initiated with 2.5 g cellobiose/l and 15 g Solka Floc/l were 0.187 units (U)/ml, achieved by continuous feeding to maintain 〈0.1 g cellobiose/l, and 0.215 U/ml using the same initial medium when 2.5 g cellobiose/l was step-fed after the sugar was nearly consumed. In batch, dual-substrate systems consisting of simple sugars with Solka Floc, substrate inhibition was evident in terms of specific growth rates, specific productivity values, and maximum enzyme yields. Limiting concentrations of glucose or sucrose at 5 g/l, and cellobiose at 2.5 g/l, in the presence of Solka Floc, yielded cellulase activities of 0.134, 0.159, and 0.164 U/ml, respectively.

    Notes: Abstract Physiological concentrations of ethylene in the growth medium of autotropic suspension culture Chenopodium rubrum L. cells reduced the activity of cell wall bound invertase by 25 – 47%, compared to controls. Northern blot analysis using homologous probe binding to total RNA preparations revealed that reduction in specific activity was paralleled by repression of the corresponding gene.

    Notes: The effect of different concentrations and combinations of oxygen, carbon dioxide, and ethylene on cell growth and taxol production in suspension cultures of Taxus cuspidata was investigated using several factorial design experiments. Low head space oxygen concentration (10% v/v) promoted early production oftaxol. High carbon dioxide concentration (10% v/v) inhibited taxol production. The most effective gas mixture composition in terms of taxol production was 10% (v/v) oxygen, 0.5% (v/v) carbon dioxide, and 5 ppm ethylene. Cultures grown underambient concentration of oxygen had a delayed uptake of glucose and fructose compared to cultures grown under 10% (v/v) oxygen. Average calcium uptake rates into the cultured cells decreased and average phosphate uptake rates increased as ethylene was increased from 0 to 10 ppm. These results may indicate that gas composition alters partitioning of nutrients, which in turn affects secondary metabolite production. © 1995 John Wiley & Sons, Inc.

    Notes: Pressed and wilted samples of sweet sorghum [Sorghum bicolor (L.) Moench var. Rio] were ensiled for periods up to 155 days. A kinetic study of the biochemical changes which occurred during ensiling showed that in wilted sorghum ensilage invert sugars and mannitol levels collectively were maintained at 65% of the original ferment able sugar content of the sorghum. The acidic environment produced by ensiling also served as a pretreatment that resulted in enhanced yields of reducing sugar when the sorghum was contacted with cellulolytic enzymes. The quantity of sugar obtained from enzymatic hydrolysis more than compensated for carbohydrate used by organisms during the ensiling process. Both Saccharomyces uvarum and Clostridium acetobutylicum were able to ferment a medium constituted from pressed sorghum juice and the solution resulting from enzymatic hydrolysis of sweet sorghum ensilage.

    Notes: Summary Cellulolytic and hemicellulolytic enzymes were produced on extracted sweet sorghum silage by mixed culture solid substrate fermentation with Trichoderma reesei LM-1 (a Peruvian mutant) and Aspergillus niger ATCC 10864. Optimal cellulose and xylanase levels of 4 IU/g dry weight (DW) and 180 IU/g DW, respectively, were achieved in 120 h-fermentation when T. reesei, inoculated at 0 h, was followed by the inoculation of A. niger at 48 h.

    Notes: Batch fermentations were run at varying agitation rates and were either pressurized to 1 bar (15.2 psig) or nonpressurized. Agitation and pressure both affect the level of dissolved hydrogen gas in the media, which in turn influences solvent production. In nonpressurized fermentations volumetric productivity of butanol increased as the agitation rate decreased. While agitation had no significant effect on butanol productivity under pressurized conditions, overall butanol productivity was increased over that obtained in the nonpressurized runs. Maximum butyric acid productivity, however, was found to occur earlier and increased as agitation increased. Peak hydrogen productivity occurred simultaneously with peak butyric acid productivity. The proporation of reducing equivalents used in forming the above products was determined using a redox balance based on the fermentation stoichiometry. An inverse relationship between the final concentrations of acetone and acetoin was found in all fermentations studied. The results show that agitation and pressure are important parameters for solvent productivity in acetone-butanol fermentation.
