    Notes: Abstract The pulmonary macrophages collected with bronchial mucus from patients with cystic fibrosis and other chronic lung diseases were investigated histochemically for acid phosphatase, beta-glucuronidase and beta-glucosaminidase activity, and also for mucopolysaccharide content. In macrophages from patients with cystic fibrosis decreased beta-glucuronidase activity was observed, together with large cytoplasmic vacuoles suggesting storage of partially digested material. It thus seems very likely that apart from the increased synthesis of mucopolysaccharides described in cystic fibrosis by several authors, the degradation of mucosubstances produced can be impaired.

    Notes: Summary A method histochemical localization of prostaglandin synthetase using DAB, potassium cyanide and polyunsaturated fatty acid has been revised. The arachidonic acid-induced DAB oxidation observed in the secretory epithelium of sheep vesicular glands and in collecting tubules as well as interstitial cells of rabbit kidney medulla was found to be insensitive to antiinflammatory cyclooxygenase (formerly referred as prostaglandin synthetase) inhibitors, such as indomethacin, aspirin, mefenamic acid and paracetamol, whereas aminotriazole caused complete inhibition of the reaction. Furthermore, DAB was oxidized in the presence of polyunsaturated fatty acids inconvertible to prostaglandins (linoleic and linolenic acid) as well as in the presence of H2O2 — in the latter case reaction possessed identical features with that induced by fatty acids. Ultrastructurally, the reaction product was localized on the membranes of nuclear envelope and endoplasmic reticulum. On the ground of the results obtained a hypothesis is presented, that the polyunsaturated fatty acid-induced DAB oxidation is due to a peroxidatic activity of the investigated tissues. Possible relations between such peroxidatic activity and prostaglandin biosynthesis are discussed.

    Notes: Summary The effect of various stabilizers — 0.9% NaCl, 7.5% sucrose and 7.5% polyvinyl pyrrolidone (PVP) on reactions of mitochondria with fresh and photooxidized diaminobenzidine (DAB) was investigated. NaCl and PVP abolished the DAB staining of mitochondria incubated without exogenous cytochrome c; NaCl, however, was ineffective when exogenous cytochrome c or oxidized DAB were present in the incubation medium. PVP considerably decreased the intensity of all reactions performed in the presence of exogenous cytochrome c or oxidized DAB. The mechanism of the observed effect is not likely to involve osmotic protection of mitochondria; extraction of endogenous cytochrome c from fresh frozen tissue by NaCl and greatly increased viscosity of PVP-containing media seem to be most probable explanations.

    Notes: Abstract  A significant reduction of catalase activity, a peroxisomal marker enzyme, occurs in human hepatic neoplasias, but no information is available on other peroxisomal proteins. We have studied by means of immunohistochemistry four specific proteins of peroxisomes (catalase and three enzymes of lipid β-oxidation) in human hepatocellular tumors of various differentiation grades from adenoma to anaplastic carcinoma. In all tumors, except the adenomas, the tumor cells contained fewer peroxisomes than extrafocal hepatocytes and the reduction of antigenic sites in the tumor types generally correlated with the degree of tumor dedifferentiation as assessed by classical histopathological criteria. Two poorly differentiated tumors had no detectable peroxisomes at all. There were no major differences in the intensities of the immunocytochemical staining for all four studied peroxisomal antigens in different tumors, suggesting that the neoplastic transformation affects the biogenesis of the entire organelle and not merely the individual peroxisomal enzyme proteins. Some tumors exhibited a distinct peripheral distribution of peroxisomes. In cases with associated liver cirrhosis, the hepatocytes in the adjacent liver showed marked peroxisome proliferation, forming large perinuclear aggregates, occupying occasionally the entire cytoplasm. Taken together, our observations indicate that peroxisomes are significantly altered in both hepatocellular tumors and liver cirrhosis and, thus, could be responsible for some of the metabolic derangements observed in those disease processes.

    Notes: Summary Rat liver fixed by perfusion with low glutaraldehyde concentrations was incubated in diaminobenzidine-containing medium to stain for peroxidase. Endogenous peroxidatic activity was found not only in Kupffer cells but also in the endothelial cells lining the sinusoids and central veins. The reaction product was localized in the nuclear envelope and endoplasmic reticulum. The peroxidatic activity in endothelial cells showed a concentration-dependent sensitivity to glutaraldehyde: in liver samples fixed with 0.25% glutaraldehyde, approx. 23% of the sinusoidal endothelial cells and 65% of central vein endothelium were peroxidase-positive; with 0.5% glutaraldehyde, only approx. 8% of the sinusoidal endothelial cells contained detectable amounts of the reaction product; with 1.5% glutaraldehyde all endothelial cells were consistently peroxidase-negative. No peroxidatic activity could be found in liver endothelial cells following isolation by centrifugal elutriation. Endothelial cell peroxidase may possibly be involved in defense responses of liver and/or, as a part of prostaglandin synthase system, in prostanoid production.

    Notes: Summary The cell membrane of smooth muscle cells from four large arteries of rabbits was investigated by freeze-fracture. On the surface of the smooth muscle smooth zones could be distinguished arranged in an alternate manner with zones characterised by numerous openings of surface vesicles. Vesiculate zones formed bands of variable width, generally parallel to the long axis of the cell, or irregular patches of different size. The density of vesicles within bands and patches varied from a very loose packing to a dense, quasi-hexagonal array. Comparative morphometric studies of the cell membrane replicas showed differences in the ratio vesiculate area: smooth area and also different surface vesicle frequencies expressed either as mean values or as values for vesiculate zones, in the arteries investigated. Communicating junctions (nexuses, gap junctions) characterized by a random arrangement of particles on the P-face and a more ordered, rhomboidal array of pits on the E-face were the only type of intercellular junctions found between arterial smooth muscle cells.
