    Notes: Abstract  In many applications up to millions of sensors or actuators have to be interconnected to each other and/or to the outside world, making the monolithic integration of circuitry mandatory. This monolithic integration is also pursued for mass-produced transducers because of economical reasons. CMOS-integrated transducers are thus found in imaging transducers arrays and mass-produced physical sensors. In addition, integrated biochemical sensor arrays can be CMOS-integrated. In this article first a general overview of the field is given and then selected work in the imaging and biochemical field is highlighted. Concerning imaging transducer arrays an overview is given of visible and IR imagers, displays and of inkjet printheads. Concerning the new field of biochemical sensor arrays, two examples are described: a blood-gas sensor and an array of interdigitated electrodes. Finally an overview of the possible technological approaches regarding integrated processing of transducers is presented.

    Notes: The generation of reactive oxygen species (ROS) in UV-exposed skin is believed to contribute to the photoaging process. The stratum corneum (SC) contains a variety of enzymatic and non-enzymatic antioxidants to protect against various environmental sources of free radicals. We have previously shown a seasonal variation in SC catalase activity with strong deactivation in sun-exposed skin in the summer, whereas SC superoxide dismutase (SOD) activity remained intact in those conditions. This potentially leads to the local overproduction of H2O2. The oxidized lipid squalene hydroperoxide accumulates at the surface of sun-exposed skin in the summer and upon exposure to ultravoilet A (UVA) doses as low as 0.1 J cm−2 and adequate protection against excessive lipid peroxidation at times of UV exposure should be aimed for. We have been using the induction of lipid hydroperoxides at the skin surface by a single dose of UVA (1 J cm−2) as a model system to evaluate the protective effect of antioxidants in vivo. Topical treatment with the synthetic SOD/catalase mimetic molecule (EUK-134) 1 h before UVA exposure reduced the level of lipid peroxides at the surface of UVA-exposed skin but also baseline peroxide levels on non-irradiated skin were reduced in a dose-dependent fashion. In contrast to alpha-tocopherol, EUK-134 even reduced the level of lipid peroxides at the surface of UVA-exposed skin when it was applied after irradiation. We confirmed that this salen–manganese complex was able to reduce squalene hydroperoxide levels in vitro, suggesting peroxidase-like activity towards organic peroxides. These data support the concept that the synthetic SOD/catalase mimetic EUK-134 might be able to compensate for seasonal deficiencies in antioxidant defense capacity at the skin surface, thereby contributing to an optimal protection of the skin against the accumulation of oxidative damage.

    Notes: The bio-mechanical properties of the stratum corneum are of great importance for the subjective perception of skin conditions (dry vs. normal), as well as for its multiple functions. The recently developed Gas Bearing Electrodynamometer allows us to measure objectively the viscoelastic properties of the stratum corneum in vivo and to evaluate, in terms of skin softness, the changes of this parameter induced by the application of emollients.The principle of the technique, as designed by Christensen et al., is briefly described in context with our results.During the probe movement, there was a modification of the skin surface relief: compression of the skin furrows ahead of the moving probe, and extension-flattening of the relief behind the probe.The variations of the skin softness were studied in context with the reproducibility of the measurements: the coefficient of variation was found to be about 3%. Considerably higher variations were found as related to: the anisotropy of the skin surface relief, various areas of the body, and individual differences.The effects of emollients and water on the skin softness were studied. Water had a clear effect but of very short duration (around 10 minutes). Water-containing emollients of the type o/w emulsions displayed a distinct long-lasting effect (followed up to 6 hours). Its magnitude was generally higher than that obtained with w/o emulsions. It was found that the emollient's non-volatile components (oil phase) play an important role in the product's softening effect.In conclusion, results to date have established this technique as a promising tool for studying skin softness in general, and effects of topical applications, new formulae and specific ingredients in particular.     Notes: The ambivalent consequences of mitochondrial stimulation on cellular activity have been well established. Mitochondria supply the cell with energy through a process of oxidative phosphorylation but thereby generate free radicals, resulting in the accumulation of hydrogen peroxide in the cytoplasm. We have investigated the impact of cellular senescence as well as UV irradiation, on the balance between these two activities.The adenosine triphosphate (ATP) level, DNA and protein synthesis in fibroblasts obtained from donors between 30 and 90 years of age appeared to be significantly influenced by the aging process. Both DNA and protein synthesis could be stimulated by increasing intracellular ATP levels. In-vitro senescent fibroblasts showed a reduction in the level of ATP as well as a shift in mitochondrial membrane potential. At the same time, there was an increase in intracellular hydrogen peroxide with increasing population doubling, indicating a clear dysfunction of the metabolic machinery in the mitochondria of senescent cells. To counteract this degradation of the energy pool, we treated cells with creatine, which is known to restore the pool of phosphocreatine in the mitochondria. Creatine treatment significantly increased cell survival after UV exposure, stimulated the repair of UVB-induced DNA damage in keratinocytes and caused a significant reduction in the number of sunburn cells in a UVB-exposed reconstituted skin model. These results clearly indicate that restoration of the energy pool in mitochondria increased cellular self-defense mechanism. These data show the important role played by the mitochondrial energy metabolism on the aging process, and indicate a possible therapy that can be used to counteract this negative effect. Treatment with creatine seems to provide the necessary boost to the cellular metabolism, which leads to an induction of a significant amount of protection and repair to human skin cells.

    Notes: A multi-technique evaluation of the hair regreasing process is described. Preliminary experimentation in vitro was followed by a long-term in vivo study conducted on a single subject having greasy hair. First, a shampoo for very greasy hair and one for dry hair were applied to the left and right sides of the head respectively (split-head test) for 7 weeks; then a shampoo for greasy hair was applied to the entire head (whole-head test) for 5 weeks. Three parameters were studied: (1) hair spacing, determined by means of a special device; (2) regreasing of individual hairs, measured using the contact-reprint technique; (3) the amount of sebum extracted from individual hairs, performed by g.l.c. determination of squalene. Results obtained during the split-head period showed that the shampoo for very greasy hair showed higher average hair spacing, slower progression of regreasing, and better sebum-removal efficiency when compared to the shampoo for dry hair. During the whole-head test period with one shampoo, results showed that there was consistent asymmetry as regards hair spacing and progression of regreasing.     Notes: The X-ray structure of bovine ribonuclease A cocrystallized with the dinucleotide deoxycytidylyl-3′,5′-guanosine has been determined at 1.9 Å resolution and refined by restrained least squares to R = 0.218 for 7807 reflections. The structure established that the recently observed retrobound mode of attachment of substrate analogues cytidylyl-2′,5′-guanosine and deoxycytidylyl-3′,5′-guanosine found in soaked RNase A crystals is also present in the cocrystallized complex. Retrobinding is thus unlikely to be the result of restrictions imposed by the crystalline environment as the ligands soak into the lattice but rather a phenomenon specific to small nucleotides containing guanine.

    Notes: The X-ray structure of the inhibitor complex of bovine ribonuclease A with cytidylic acid (2′-CMP) has been determined at 1.6 Å resolution and refined by restrained least squares to R = 0.17 for 11 945 reflections. Binding of the inhibitor molecule to the protein is confirmed to be in the productive mode associated with enzyme activity. A study of conserved solvent sites amongst high-resolution structures in the same crystal form reveals a stabilizing water cluster between the N and C termini.
