    Notes: Summary Partial sequences of 18s rRNA were obtained for 2 gymnosperms and 12 angiosperms from a wide range of families and these were analyzed with 5 other published sequences to form a phylogenetic tree. Using 16 published sequences of the large subunit of rubisco (rbcL), also from a wide range of angiosperm families, another phylogenetic tree was derived and the two approaches were compared. Both phylogenetic trees gave good grouping within families but in neither case was there resolution of the branching order of major taxa. Superficially the long rbcL sequences (whose base composition was homogeneous among all species) seemed very promising, but analysis showed that a large proportion of the variation did not affect the amino acid sequence. Although silent substitution contained some phylogenetic information, at the level required to order major taxa, much of it was random and obfuscating. It was concluded that neither macromolecule alone was likely to yield a solution to the problem of angiosperm phylogeny and therefore that studies of both, at least, will be required. For this reason, a method wa described for obtaining both DNA and RNA of good quality from the same preparation and which had been used successfully with a wide range of species including many with pungent leaves.

    Notes: Abstract Among specimens of the spectacled hare-wallaby Lagorchestes conspicillatus Gould (Marsupialia, family Macropodidae) 4 males had 15 chromosomes and 2 females 16 chromosomes. The sex chromosomes are X1X1X2X2 in the female and X1X2Y in the male, the Y being metacentric and both X chromosomes are acrocentric. In about 96% of sperm mother cells at meiosis the sex chromosomes form a chain trivalent and in more than 99% of these this orients convergently so that the X1 and X2 move to the same pole. Evidence is presented that L. conspicillatus has evolved from a form with 22 chromosomes including a small X and a minute Y. Autoradiographic studies show that the proximal fifth of the X1 chromosome replicates late. This is probably the ancestral X chromosome which has been translocated to an autosome. The fate of the original Y is obscure but an hypothesis is proposed that it forms the centromeric region of the Y. A single male had 14 chromosomes and was heterozygous for a translocation involving the centric fusion of two acrocentric autosomes. In about 30% of sperm mother cells the autosomal trivalent did not disjoin regularly but, despite this, all secondary spermatocytes observed at metaphase 2 had balanced complements of chromosomes. It is assumed that unbalanced secondary spermatocytes died before reaching metaphase.

    Notes: Abstract Relative DNA values and percent lengths of chromosome arms have been studied for six species of the super-family Dasyuroidea, five species of Phalangeroidea and four species of Perameloidea. By multiplying by relative DNA values all percent lengths have been expressed in the same units. Two species are said to “share” a chromosome if neither of the arms differs at the 5% level of probability. It is argued that if species “share” at least two two-armed chromosomes, this can be taken as evidence of relationship. A “standard dasyurid” karyotype has been defined and individual species of Dasyuridae show only small deviations from it. The nature and significance of the bi-modal distribution of chromosome numbers in marsupials is discussed.

    Notes: Abstract Echymipera kalabu (Peramelidae: Marsupialia) does not have the full chromosome complement in all its adult somatic tissues. The chromosomes missing are the Y-chromosome in the male and an X-chromosome in the female. The full complement is present in the corneal epithelium and the reproductive tissue. A parallel mosaicism to this exists with respect to small supernumerary chromosomes which are found in certain animals of this species. These supernumeraries must be subject to the same control system as that which is responsible for the elimination of the sex chromosomes.

    Notes: Summary The effect of nisoldipine on renal function after 6 weeks treatment was investigated in hypertensive patients with and without renal impairment. Nisoldipine was well tolerated and an effective antihypertensive agent when administered over a period of 6 weeks. There were no significant changes in glomerular filtration, cardiac output, plasma renin activity or serum biochemistry during nisoldipine administration. Effective renal plasma flow was unaffected by treatment in the patients with normal renal function, but in the patients with renal insufficiency, the value decreased by a mean of 12%. Nisoldipine had no major untoward effects on renal function after 6 weeks administration, but minor changes in renal haemodynamics in the patients with renal insufficiency would suggest that careful monitoring of renal function is indicated in such patients.

    Notes: The interaction potential energy surface (PES) of H4 is of great importance for quantum chemistry as a test case for molecule–molecule interactions. It is also required for a detailed understanding of certain astrophysical processes, namely collisional excitation and dissociation of H2 in molecular clouds, at densities too low to be accessible experimentally. The 6101 ab initio H4 energies reported in 1991 by Boothroyd et al. demonstrated large inaccuracies in analytic H4 surfaces available at that time. Some undesirable features remained in the more accurate H4 surfaces fitted to these energies by Keogh and by Aguado et al., due in part to the relatively sparse coverage of the six-dimensional H4 conformation space afforded by the 6101 ab initio energies. To improve the coverage, 42 079 new ab initio H4 energies were calculated, using Buenker's multiple reference (single and) double excitation configuration interaction program. Here the lowest excited states were computed as well as the ground state, and energies for the original 6101 conformations were recomputed. The ab initio energies have an estimated rms "random" error of ∼0.5 millihartree and a systematic error of ∼1 millihartree (0.6 kcal/mol). A new analytical H4 PES was fitted to these 48 180 ab initio energies (and to an additional 13 367 points generated at large separations), yielding a significant improvement over previous H4 surfaces. This new PES has an rms error of 1.43 millihartree relative to these 48 180 ab initio energies (the fitting procedure used a reduced weight for high energies, yielding a weighted rms error of 1.15 millihartree for these 48 180 ab initio energies). For the 39 064 ab initio energies that lie below twice the H2 dissociation energy, the new PES has an rms error of 0.95 millihartree. These rms errors are comparable to the estimated error in the ab initio energies themselves. The new PES also fits the van der Waals well to an accuracy of about 5%. For relatively compact conformations (energies higher than the H2 dissociation energy), the conical intersection between the ground state and the first excited state is the largest source of error in the analytic surface. The position of this conical intersection forms a somewhat complicated three-dimensional hypersurface in the six-dimensional conformation space of H4. A large portion of the position of the conical intersection has been mapped out, but trying to include the conical intersection explicitly in an analytic surface is beyond the scope of the present paper. © 2002 American Institute of Physics.
