    Notes: Small interfering RNA (siRNA) holds potential as a therapeutic approach to silence targetedgene of disease, but siRNA has limited stability in vivo. Therefore, delivery system of siRNA is the key tosiRNA therapeutic application. We attempted to develop a delivery system, which enables siRNA todemonstrate high stability and long blood circulation. We synthesized a series of bottlebrush-typecopolymers (BBCs) possessing polycationic backbone (less than 30 wt%) and abundant water-soluble sidechains (more than 70 wt%) as siRNA carrier. A siRNA complexed with the BBC was resistant tonuclease and stable in plasma. Especially, the BBC (10 wt% PLL and 90 wt% PEG) having highergrafting ratio (≈ 90 wt%) of water-soluble side chains showed 100-times enhanced stability of siRNA inmouse bloodstream in vivo. Surprisingly, even when the BBC and siRNA separately injected into mouseat 20 min interval, the BBC increased blood half life of the siRNA. These results suggest that the BBChas higher selectivity in its ionic interaction to siRNA than other anionic substance in blood components.To our knowledge, this is the first report of siRNA delivery carrier which prolonged blood circulation ofsiRNA without resource-consuming preparation process

    Notes: Brain-derived neurotrophic factor (BDNF) acutely modulates synaptic transmission to excitatory neurons in hippocampus and neocortex. The question of whether BDNF acts similarly on excitatory synaptic transmission to GABAergic neurons was eluded in previous studies using cortical slices. To address this question, we used transgenic mice in which expression of green fluorescence protein (GFP) is regulated by glutamic acid decarboxylase 67 (GAD67) promoter. In cortical slices prepared from these GAD67-GFP knock-in mice, we could detect GABAergic neurons under a fluorescent microscope. An application of BDNF rapidly depressed excitatory postsynaptic currents (EPSCs) evoked by layer IV stimulation in most GFP-positive neurons in layer II/III of the cortex. This effect was seen at synapses activated during the BDNF application and blocked by anti-TrkB IgG, indicating that the acute inhibitory action of BDNF is activity-dependent and mediated through TrkB. Paired-pulse ratios of the amplitude of EPSCs to paired stimulation at intervals of 10–100 ms were not significantly changed after BDNF application, suggesting that the site of depression may be postsynaptic. Responses to directly applied glutamate were also depressed by BDNF in most of neurons, being consistent with the interpretation of postsynaptic action of BDNF. The depressive action of BDNF was blocked by an intracellular injection of a Ca2+ chelator, suggesting that a rise in Ca2+ is involved in the acute depression of EPSCs. This action of BDNF was seen in 67% of parvalbumin (PV)-positive neurons, but in only 19% of PV-negative neurons, indicating that the depressive action is biased to PV-positive GABAergic neurons.

    Notes: Abstract Purpose. To characterize the physical and biochemical properties of the DNA terplex delivery system, which has previously been shown to deliver and express pSV-β-gal plasmid efficiently in cultured smooth muscle cells (SMC) (1). Methods. Atomic force microscopy (AFM), zeta-potential measurement (ZP), gel electrophoresis (GE), circular dichroism (CD), fluorescence quenching and 1H-NMR spectrometry were used. Results. AFM showed that the plasmid DNA of about 600 nm long in its extended state was condensed to the size of about 100 nm by terplex formation. The DNA condensing effect of the terplex system was as good as unmodified PLL, as shown by an ethidium bromide displacement assay. Zeta-potential measurement showed that the terplex system exerts a slightly positive surface charge (+2 mV) at a 1:1:1 weight ratio of DNA:LDL:stearyl-PLL, which showed the best transfection efficiency on SMC. GE indicated that electrophoretic mobility of the terplex system decreased with increasing amounts of stearyl-PLL, indicating that the surface charge of the terplex system became more positive as more stearyl-PLL was added. Results from CD showed that there was no significant changes in tertiary structure of plasmid DNA from the terplex formation. Presence of strong hydrophobic interaction between stearyl-PLL and LDL was confirmed by 1H-NMR, where about a 30% decrease in ε-methylene peak of PLL backbone was observed when stearyl-PLL was mixed with LDL, but this phenomenon was not observed when unmodified PLL was used. Conclusions. Our results indicate that the plasmid DNA, when formulated with the stearyl-PLL and LDL, forms a stable and hydrophobicity / charge balanced terplex system of optimal size for efficient cellular uptake and the DNA is still intact after the terplex formation. This information is expected to be utilized for the development of much improved transfection vectors for in vivogene therapy.

    Notes: Abstract. To elucidate the after-effect of exercise on left ventricular (LV) function, end-diastolic volume (EDV), end-systolic volume (ESV) and ejection fraction (LVEF) were evaluated at 1 h after exercise and at rest by technetium-99m tetrofosmin gated myocardial single-photon emission tomography (SPET) using an automated program in 53 subjects. The subjects were grouped as follows: normal scan (n = 16), ischaemia (n = 19) and infarction (n = 18), based on the interpretation of perfusion images. Postexercise LVEF did not differ from resting LVEF in the groups with normal scan and infarction. In patients with ischaemia, postexercise EDV (90±17 ml, mean ±SD) and ESV (44±15 ml) were significantly higher than EDV (84±15 ml, P = 0.001) and ESV (36±14 ml, P〈0.0005) at rest. LVEF was significantly depressed 1 h after exercise (53%±9% vs 58%±9%, P〈0.0001). In ischaemic patients with depressed postexercise LVEF, LVEF difference between rest and postexercise showed a significant correlation with the sum of defect scores, which were reversible from exercise to rest perfusion images (r = 0.92, P〈0.0001). These results indicate that exercise-induced LV dysfunction (myocardial stunning) continues for at least 1 h in ischaemic patients and that the extent of LVEF depression is determined by the severity of ischaemia.

    Notes: Radical copolymerizations were carried out with 2-hydroxyethyl methacrylate (HEMA) and a polyamine macromonomer (1). 1H NMR, IR, liquid chromatography and turbidity measurements showed the polymer obtained to be poly[(HEMA)-graft-polyamine]. The monomer reactivity ratio for the polyamine macromonomer was found to be similar to that of a low-molecular-weight analogous monomer. Microphase separated structure was observed on thin films of the copolymer by means of transmission electron microscopy. Using the column method, the biomedical behavior of the copolymer was estimated in terms of its interaction with rat lymphocytes. Strikingly, the copolymer surface was found to differentiate the lymphocyte subpopulations without any added adjuvant protein.
