    Notizen: The normal mode analysis in dihedral angle space is carried out on a famous double-stranded DNA dodecamer d(CGCGAATTCGCG) to reveal the dynamical characteristics and internal motion of this molecule. We have observed the coupled motion of sugar and base. We have also noted the dependence of internal motion in the dodecamer on location of the nucleotide relative to the helical ends, and the dependence of atomic fluctuations on distance from the helix axis. The similarity of atomic fluctuations between two strands suggests the presence of a twofold pseudosymmetry axis with its origin located at the center of the molecule. Our results show that the atomic fluctuations of duplex DNA are mainly determined by a small number of low-frequency normal modes, the normal modes with frequencies below 30 cm−1 make major contributions to the site-dependence of atomic fluctuations. © 1997 American Institute of Physics.

    Notizen: We have carried out normal mode analysis for four B-DNA molecules with different lengths and sequences. By focusing our attention on motions of the helical axes of the molecules and comparing them with those of the ideal rod obeying vibrational theory, we have shown that DNA molecules behave like isotropic and homogeneous elastic rods. Then, we have calculated dynamic constants representing their rigidities for bending and twisting. In the analysis of their twisting motions, we have shown that sliding and shifting motions between adjacent bases are important for the motions. © 1999 American Institute of Physics.

    Notizen: Abstract PURPOSE: We report a patient with a recurrent pelvic tumor after abdominoperineal resection of a rectal carcinoma who was treated sufficiently by repeated intra-arterial infusions of 5-fluorouracil. METHODS: A continuous, 24-hour 5-fluorouracil administration was made through the bilateral internal iliac artery at a dosage of 250 mg/m2/day by the subcutaneous reservoir located at both upper legs using a Baxter infusor. RESULTS: In this patient pain in the hip and pelvis was relieved. A complete regression in the infused field of pelvic tumor was observed not only with computed tomography and magnetic resonance imaging but also confirmed by operative findings at the seventh month after the intra-arterial infusion. The abnormal serum level of carcinoembryonic antigen and carbohydrate antigen 19-9 was decreased to within the normal range at the 19th and 3rd week respectively. When the repeated recurrence was supected in follow-up, normalization of the re-elevated carcinoembryonic antigen and carbohydrate antigen 19-9 levels was also obtained by repeating the same treatment. The side effects and complications were tolerable, consisting of local skin erosion on the hips and lower extremity neuropathy caused by the 5-fluorouracil. CONCLUSIONS: Clinical local regression of a pelvic recurrence was observed in a patient with rectal recurrent tumor who received continuous intra-arterial chemotherapy. Local recurrence of rectal cancer may be controlled effectively and safely by repeating long-term, continuous, intra-arterial 5-fluorouracil infusion.

    Notizen: Abstract The purpose of this study was to clarify the functional outcomes of colonic J-pouch anastomosis (J-LAR) for lower rectal cancer in comparison with those of traditional straight anastomosis (S-LAR). A questionnaire regarding anorectal function was conducted 1 year after operation on patients who underwent J-LAR (n=15) and S-LAR (n=30). The clinical functions were assessed by an incontinence scoring system. The physiologic function was assessed by anorectal manometry and the balloon expulsion test. No patients demonstrated a diverting stoma. The bowel frequency (range) 1 year after operation was 4.8 (3–6) in the S-LAR group and significantly decreased to 1.8 (1–3) in the J-LAR group (P〈0.05). Complete evacuation was 50.2% (40%–60%) in the S-LAR group and significantly increased to 80.6% (60%–90%) in the J-LAR group (P〈0.05). Neorectal compliance was 2.2 (1.4–2.9) ml/mmHg in the S-LAR group and significantly increased to 3.1 (1.3–3.5) ml/mmHg in the J-LAR group (P〈0.01). No significant difference was observed between the two groups regarding the maximum resting or maximum voluntary squeezing pressure. In conclusion, our findings suggested colonic pouch anastomosis performed after a low anterior resection to support the compliance of the (neo)rectum to be an important factor for retaining a satisfactory bowel frequency.

    Notizen: Abstract The aim of this study was to examine the functional outcome of transsacral rectopexy performed with Dexon mesh for recurrent complete rectal prolapse. Anorectal function was assessed by anorectal manometry and defecography, before and from 1 year after surgery in five patients who were followed up for 1–3 years. The fecal incontinence score recovered from a preoperative mean score of 3.8 to a postoperative mean score of 1.2, and constipation was improved in four patients (80%). The straining anorectal angle (S-ARA), measured by defecography, improved from a preoperative value of 120.6°±6.9° to a postoperative value of 98.5°±3.5° (P〈0.05), and the perineal descent (PD) improved from a preoperative value of 16.2±2.5 cm to a postoperative value of 8.1±1.3 cm (P〈0.05). The maximal resting pressure (MRP) increased from a preoperative value of 20.5±3.7 cmH2O to a postoperative value of 40.5±4.8 cmH2O (P〈0.05).     Notizen: Abstract The internal motion of yeast phenylalanine transfer RNA is studied by normal mode analysis in extended dihedral angle space, in which the flexibility of five-membered ribose rings is treated faithfully by introducing a variable for its pseudo-rotational motion. Analysis of global molecular motions reveals that the molecule is very soft. We show that this softness comes not from the property of the “material” comprising the molecule but from its slender shape. Analysis of thermal distance fluctuations reveals that this molecule can be regarded as consisting dynamically of three blocks. Thermal fluctuations of the mainchain dihedral angles show rigidity of the anticodon region. They also show flexibility of regions around nonstacking bases. Base-stacking interactions cause suppression of the correlated functions of mainchain dihedral angles beyond a ribose ring. We analyze the thermal fluctuation of parameters describing the positions of two adjacent bases. Fluctuations of relative translational parameters in the anticodon and acceptor stem regions are found to be larger than those in other stem regions. The relative translational motions cause the two stem regions to undergo global twisting and bending motions. We show that the role of pseudo-rotational motion of sugars is important in regions around bases which are involved in nonregular interactions.

    Notizen: Abstract We have isolated and characterized a thrombopoietin (TPO)-dependent BF-TE22 cell line endogenously expressing murine Mpl, which is a subclone of murine pro-B Ba/F3 cells. TPO stimulated the proliferation of BF-TE22 cells in a dose-dependent manner, and also induced the expression of megakaryocyte lineage-specific AP-51 and CD61 cell surface antigens. The results indicate that the murine Mpl on BF-TE22 cells can transmit both proliferation and megakaryocyte lineage-specific differentiation signals to cells. Furthermore, it was shown that IL-3 inhibits the TPO-induced differentiation signals of BF-TE22 cells. These results suggest that the signals mediated by IL-3 predominate over those of TPO in BF-TE22 cells. Thus, BF-TE22 cells will be useful for the biological and biochemical studies of the TPO-Mpl signal transduction mechanism.

    Notizen: Summary In vitro phosphorylation reactions using extracts of Streptomyces griseus cells and γ-[32P]ATP revealed the presence of multiple phosphorylated proteins. Most of the phosphorylations were distinctly inhibited by staurosporine and K-252a which are known to be eukaryotic protein kinase inhibitors. The in vitro experiments also showed that phosphorylation was greatly enhanced by manganese and inhibition of phosphorylation by staurosporine and K-252a was partially circumvented by 10 mM manganese. A calcium-activated protein kinase(s) was little affected by these inhibitors. Herbimycin and radicicol, known to be tyrosine kinase inhibitors, completely inhibited the phosphorylation of one protein. Consistent with their in vitro effects the protein kinase inhibitors inhibited aerial mycelium formation and pigment production by S. griseus. All these data suggest that S. griseus possesses several protein kinases of eukaryotic type which are essential for morphogenesis and secondary metabolism. In vitro phosphorylation of some proteins in a staurosporine-producing Streptomyces sp. was also inhibited by staurosporine, K-252a and herbimycin, which suggests the presence of a mechanism for self-protection in this microorganism.
