    Notizen: Summary During molting, the epithelium of the posterior caeca (PC) of the midgut in the terrestrial crustacean, Orchestia cavimana, is active in calcium turnover. In the preexuvial period, epithelial cells that progressively differentiate into cell-type III secrete ionic calcium (originating from the old cuticle) from the base to the apex of the cell within a typical extracellular network of channels; the calcium is then stored in the PC lumen as calcareous concretions. Immediately after exuviation, the epithelial cells rapidly differentiate into cell-type IV, reabsorbing calcium from the concretions through successive generations of spherites which quickly appear, grow, and then disappear from the apex to the base of the same extracellular network. The PC epithelium is thus alternatively calcium-loaded and unloaded. When the calcium-reabsorbing process is complete (average 48 h after exuviation), the epithelial cells again differentiate into two different regional cellular types (cell-type I in the distal segment and cell-type II in the proximal segment) characteristic of the intermolt period. The dynamic changes in the PC epithelium during the postexuvial period are discussed, including the characteristic features of cell-type IV and of the reabsorption spherites.

    Notizen: Abstract Decaying alder leaves in water from Alpine Aedes breeding sites, particularly their toxicity to larval Culicidae, were investigated and characterized with comparative toxicological and chemical methods. Bioassays that used third-instar Aedes aegypti as a reference species indicated that the larvicidal effect of crude leaf litter varied with decaying age of the litter, while no toxicity was detected from leaching water of the mosquito breeding sites. Ten-month-old leaf litter was the most toxic. Comparison of the different soluble and insoluble fractions obtained after sequential extraction of decomposed litter allowed us to localize the toxicity factor to an insoluble cell-wall fraction. The toxicity seems to be linked to phenolic activity. It is higher than that found for tannic acid solutions used as a reference to mimic the larvicidal effects of the molecules naturally occurring in decaying litter. The pattern of establishing the larvicidal effect of alder leaf litter in water of Alpine Aedes breeding sites is discussed.

    Notizen: Abstract The sensitivity of larval Culicidae to vegetable tannins was investigated in different taxa representative of the fauna from alpine hydrosystems (Aedes rusticus, Culex pipiens) and foreign noxious fauna (Aedes aegypti, A. albopictus). Bioassays reveal that tannic acid at concentrations of 0.1–6 mM is significantly more toxic for C. pipiens than for Aedes taxa, and A. aegypti is more sensitive than A. albopictus and A. rusticus. Comparison of the rank order of sensitivity among taxa with the associated levels of cytochrome P-450, esterase, and glutathione-S-transferase activities suggests that cytochrome P-450 and esterases may be involved in the detoxification of tannins. A possible involvement of these detoxifying enzymes is also revealed in vivo by the synergistic effects of S,S,S-tributyl phosphorotrithioate (esterase inhibitor) and piperonyl butoxide (P-450 inhibitor). The differential sensitivity to tannins among taxa is discussed in terms of ecological implications within mosquito communities from alpine hydrosystems, where the acquisition of tannins–detoxifying enzymatic systems may be considered as a key innovation.

    Notizen: Both structural and functional changes are observed within the posterior caeca (PC) of Orchestia during the molt cycle. During the intermolt period, there are two segments which are structurally different: a distal segment lined by type I epithelial cells and a proximal segment lined by type II cells. During molting, the PC cells are active in calcium turnover. Calcium is secreted and stored as calcareous concretions in the caecal lumen during the preexuvial period; then during the postexuvial period it is reabsorbed to mineralize the new cuticle. During the preexuvial period, cellular type III differentiates along the whole length of the PC in poster-anterior sequence and functions in ionic calcium secretion, from the basal part to the cellular apex. During the postexuvial period, this cellular type turns into cellular type IV engaged in calcium reabsorption from successive generations of spherites, from the cellular apex to the basal part.The role played by the caecal epithelium during both formation and reabsorption of the concretions was investigated by experiments in which caeca were transplanted to host pericardial cavities or were blocked by causing an abdominal hernia. The main structural characteristic features of cellular type III are as follows: an extracellular network of channels extends from basal to apical ends; microvilli are long and often apically dilated; multivacuolar complexes are localized in extracellular channels and within dilated tips of microvilli before secretion into caecum lumen; bundles of microtubules are oriented in parallel around the luminal orifices of the extracellular network; ribosomes are abundant in cytoplasm. Cellular type III develops progressively from the distal end of the caecum to the proximal one as the preexuvial period advances and concretions form in the caecum lumen.
