    Notes: Abstract Recombinant fusion proteins offer important new therapeutic approaches for the future. This report describes the use of three different genetic strategies (i.e. “mono-”, “bi-” and “tri-cistronic” vectors) to achieve stable secretion from BHK cells of a glycosylated antibody-cytokine fusion protein designed for use in anti-tumour therapy. It describes selection of a robust and effective production cell line based on stability of secretion of the product, quality of mRNA and protein products and performance in in vitro bioassays for potency. The data obtained at this stage were utilised in the selection of a suitable candidate production cell line. The relative productivity and general performance of the cells in stirred tank and fixed bed culture systems indicated that a variety of cell culture technologies provided robust tools for production of a highly selected cell clone. Consistency of the product glycosylation was determined by analysis of released oligosaccharides using matrix-assisted laser desorption ionisation – time of flight mass spectrometry and high-performance anion exchange chromatography. These investigations showed consistent expression of three glycoforms of the fusion protein which varied in their relative proportions in different culture systems and at different time points in a fixed bed reactor with continuous perfusion. In conclusion, this study dealt with a range of important scientific and technical issues which are essential for regulatory approval and commercial success of a recombinant protein and elucidates some useful markers for process development for similar recombinant biologicals.

    Description / Table of Contents: Résumé Nous présentons le cas d'une jeune fille de 14 ans traitée pour un thorax en carène (pectus carinatum). Elle a été traitée initialement par un corset plâtré pendant six semaines, suivi d'un corset orthopédique pendant sept mois. Le même corset a ensuite été porté seulement la nuit pendant huit mois. Sept ans plus tard la patiente a un résultat cosmétique excellent, sans récidive. Chez un sujet bien motivé, en cours de croissance, le port d'un corset orthopédique peut être efficace dans la correction d'un thorax en carène, pour raison esthétique.
    Notes: Summary We report the case of a 14 year old girl who desired treatment for a cosmetically objectionable pectus carinatum deformity. She was initially managed with a corrective underarm body cast for six weeks followed by fulltime bracing for seven months. Subsequently, she was braced only at night for another eight months. Seven years after the onset of treatment she had an excellent result without recurrence of deformity. In a well-motivated, skeletally immature individual bracing can be an effective treatment for cosmetically displeasing pectus carinatum.

    Notes: Abstract Since the discovery of heavy-fermion superconductivity in uranium compounds in the early 1980s, other uranium compounds have been discovered that are fully as interesting to study. However, as we look forward in the year 2000, we now have higher-purity, single crystals of the element itself. Preliminary resistivity and ac susceptibility measurements show the improved quality of the samples and thus hold the promise of understanding many aspects of its superconductivity, which have remained untouched for almost 25 years.
