    Notizen: Abstract Two genetic loci regulate hepatic α-fetoprotein (AFP) mRNA levels in adult mice. The raflocus controls basal levels and the Riflocus determines the degree of induction during liver regeneration. We have investigated the function of each locus during L-ethionine-mediated AFP induction using adult female mice with different Rif/rafgenotypes. A single intraperitoneal injection of L-ethionine (0.5 mg/g body weight) resulted in significant triglyceride accumulation in hepatic parenchymal cells and increased AFP synthesis 48–96 h following injection. Hepatic AFP mRNA levels in Balb/cJ mice (high basal level/high induction level during regeneration) were 10- to 30- fold higher than Balb/cCRBL or C3H/He mice (low basal level/high induction level) following ethionine injection, indicating that raf-mediated differences persisted throughout the course of acute ethionine poisoning. The magnitude of this induction was similar to that seen during carbon tetrachloride-induced regeneration. In contrast, C57BL/6 mice (low basal level/low induction level during regeneration) contained hepatic AFP mRNA levels similar to Balb/cCRBL and C3H/He mice following ethionine injection. Thus, Rif-dependent differences seen during liver regeneration were not seen during acute ethionine poisoning. This leads us to conclude that (1) hepatic AFP mRNA induction by ethionine may not be mediated by the Riflocus if Rifis a transcriptional inducer, or (2) if Rifis a transcriptional repressor, it is inactivated equally in all strains during acute ethionine poisoning, unlike during liver regeneration. Hepatic albumin mRNA levels were not affected by ethionine treatment in vivo. L-Ethionine elevated AFP mRNA levels in primary mouse hepatocyte cultures; however, ethionine treatment also increased albumin mRNA levels in vitro.

    Notizen: Summary The excretory organs of Amphioxus occur as segmentally arranged structures throughout the pharyngeal region and may be divided into three components: the solenocytes, the renal tubule, and the renal glomerulus. The solenocytes possess foot processes that rest upon the coelomic surface of the ligamentum denticulatum. The tubular apparatus of the solenocytes consists of ten triangular rods surrounding a central flagellum. The distal end of the tubular apparatus enters branches of the renal tubule. The renal tubule eventually opens into the atrial cavity of Amphioxus. The renal glomerulus is a sinus within the connective tissue of the ligamentum dentieculatum where it connects elements of the branchial circulation with the dorsal aorta. The renal glomerulus, like other blood vessels of Amphioxus, lacks an endothelial lining. If Amphioxus is adapted to artificial sea water at different concentrations there is no change in kidney morphology suggesting that Amphioxus is either is osmotic with its environment or is osmoregulating with other organs.

    Notizen: Summary Exposure of fibroblasts to tunicamycin has been found to be cytotoxic for transformed cells, but not for nontransformed cells. With two mouse epidermal cell lines of common origin, we observe a contrary pattern: The malignant cells are more resistant to tunicamycin than their nonmalignant counterparts, as measured by growth and viability. With respect to the glycosylation of sugar precursors, the incorporation of mannose is more inhibited than that of glucosamine, while fucose is least impacted. Sugar incorporation is less reduced for the malignant cells, by a factor of two for fucose and more modestly for the other two sugars. There are no significant morphological changes; in particular, the desmosomal junctions are not affected. On polyacrylamide gels, we note intensity variations in several protein bands in response to tunicamycin, but little difference between malignant and nonmalignant cells when using either Coomassie stains or Concanavalin A overlays.

    Notizen: In order to clarify the morphology of the circulatory system of amphioxus the blood vessels were investigated using modern techniques of light and electron microscopy.The pattern of circulation in amphioxus is forward ventrally and backwards dorsally. In addition, circulating corpuscles, usually associated with the blood of higher chordates, are absent.The circulatory system of amphioxus consists of well defined contractile vessels and vascular spaces or sinuses within a connective tissue matrix. The contractile vessels have a discontinuous endothelial lining resting on a basal lamina and are enclosed by a simple layer of contractile myoepithelial cells. Discontinuous endothelial linings occur throughout the vascular tree, including major and minor afferent and efferent vessels and blood sinuses. This is in contrast to higher animals where the endothelium forms a more or less continuous lining along the inner surface of the boundary layer.It is suggested that the endothelial cells of amphioxus, like the endothelial cells in capillaries of higher chordates, most likely play a role in the physiology of the circulatory system by removing residues of filtration from the basal lamina, thereby facilitating an exchange of materials to and from the surrounding tissues.

    Notizen: Summary The endothelial cell of Amphioxus has an endocytotic capability since both ferritin and horseradish peroxidase, when injected into the endostylar artery, became localized within endothelial cell cytoplasmic vesicles. The endothelial cells also contain vesicles within which the lysosomal enzyme acid phosphatase has been localized. Because of the presence of an endocytotic capability and lysosomes, these cells possess both the means and the method for the acquisition and disposal of exogenous proteins.
