    Notes: The ABSCISIC ACID-INSENSITIVE 3 (ABI3) gene of Arabidopsis thaliana (L.) Heynh is known to play an important role during seed maturation and dormancy. Here, we present evidence suggesting an additional role for ABI3 during vegetative quiescence processes. During growth in the dark, ABI3 is expressed in the apex of the seedlings after cell division is arrested. The 2S seed storage protein gene, a target gene of ABI3 in seeds, is also induced in the arrested apex under similar darkness conditions. In addition, β-glucuronidase expression under the control of the ABI3 promoter is abolished by treatments that provoke leaf development in the dark [sucrose and abscisic acid (ABA) biosynthesis inhibitors] and induced by treatments that prevent leaf development (darkness and ABA). Furthermore, ABI3 expression is absent in apices of dark-grown de-etiolated (det 1) and abi3 mutants, both known to develop leaves or leaf primordia in the dark. The fact that the expression of the ABI3 gene is only observed in a fraction of the analysed plants suggests that ABI3 is probably only one of the components of a molecular network underlying quiescence. In addition to the expression of ABI3 in apices of dark-grown seedlings, the ABI3 promoter confers expression in other vegetative organs as well, such as the stipules and the abscission zones of the siliques. In conclusion, apart from its role in seed development, ABI3 might have additional functions.

    Notes: Pending cell death was visualized by thermographic imaging in bacterio-opsin transgenic tobacco plants. Cell death in these plants was characterized by a complex lesion phenotype. Isolated cell death lesions were preceded by a colocalized thermal effect, as previously observed at sites infected by tobacco mosaic virus (TMV) (Chaerle et al. 1999Nature Biotechnology 17, 813–816). However, in most cases, a coherent front of higher temperature, trailed by cell death, initiated at the leaf base and expanded over the leaf lamina. In contrast to the homogenous thermal front, cell death was first visible close to the veins, and subsequently appeared as discrete spots on the interveinal tissue, as cell death spread along the veins. Regions with visible cell death had a lower temperature because of water evaporation from damaged cells. In analogy with previous observations on the localized tobacco–TMV interaction (Chaerle et al. 1999), the kinetics of thermographic and continuous gas exchange measurements indicated that stomatal closure preceded tissue collapse. Localized spontaneous cell death could also be presymptomatically visualized in the Arabidopsis lsd2 mutant.

    Notes: The heavy metal lead was administered to Arabidopsis thaliana plants by foliar spray. At a concentration of l4mol m−3, the lead nitrate suspension induced densely distributed necrotic lesions on A. thaliana leaves. A number of Arabidopsis ecotypes were tested and a differential response to heavy-metal toxicity was noted. The necrosis provoked as a result of the phytotoxic effect of lead had a similar appearance to the necrotic lesions observed in a hypersensitive response of A. thaliana to inoculation with Xanthomonas campestris pv. campestris (Lummerzheim et al. 1993, Molecular Plant-Microbe Interactions 6, 532–544). In addition to this phenotypic resemblance, accumulation of polyphenols and callose depositions observed by microscopic analysis, as well as increases in the activities of the stress-related proteins β1,3-glucanases, chitinases and peroxidases, revealed significant similarities in the plant response to the two treatments examined, lead toxicity and bacterial infection. The results allow the establishment of markers for both types of stress.

    Notes: To gain better insights into the generation of oxidative stress by NaCl stress, we compared the effects of toxic, sublethal concentrations of NaCl (150 mM) with those of CuSO4 (100 μM) on the antioxidant defence responses of Nicotiana plumbaginifolia L. plants grown in hydroponic culture. Short- and long-term adaptive responses were investigated as well as the effects of root cutting to allow rapid absorption of ions. Our results show that photosynthesis is already affected after 12 h of both treatments and that photoinhibition is induced. Furthermore, the two treatments showed distinct patterns of induction of the genes of the antioxidant systems. Principally, NaCl stimulated catalase activity through activation of the Cat2 and Cat3 genes, whereas with CuSO4, transcript levels, protein levels, and enzymatic activities of glutathione peroxidase, ascorbate peroxidase, and superoxide dismutase were differentially induced. The antioxidants ascorbate and glutathione were found to be largely oxidized after 12 h, suggesting an inability of the recycling process to cope with the stress. In addition, both NaCl and CuSO4 treatments stimulated the accumulation of the osmolyte proline. Therefore, we conclude that the uptake and accumulation of different types of ions lead to the generation of distinct antioxidant responses in N. plumbaginifolia.

    Notes: The nucleotide sequence of the chloramphenicol-resistance gene (cmr) of Rhodococcus fascians NCPPB 1675 (located on the conjugative plasmid pRF2) allowed the identification of two possible open reading frames (ORFs), of which ORF1 was consistent with the mutational analysis. Biochemical analysis of cmr revealed that it does not encode an antibiotic-modifying enzyme. The amino acid sequence of 0RF1 predicted a hydrophobic protein, with 12 putative membrane-spanning domains, homologous to proteins involved in the efflux of tetracycline across the plasma membrane. Expression of the cmr gene was induced by addition of chloramphenicol to the growth media. The promoter of this gene was restricted to 50 bp upstream from a 200 bp 5′-untrans-lated mRNA region, the latter containing two inverted repeats. At the amino acid level, the cmr gene is 52% identical to a previously identified chloramphenicol-resistance determinant in Streptomyces lividans, indicating a wider dispersion of this type of cmr gene among the actinomycetes.

    Notes: Electrotransformation of Rhodococcus fascians by non-replicating plasmids containing a suitable resistance marker resulted in stable transformants by integration of these constructs at various sites in the genome, thereby generating different mutations. Tagged genes could be isolated in Escherichia coli owing to the presence of a CoIE1 replicon and an ampicillin resistance gene in the inserted sequences.Southern analysis and nucleotide sequencing revealed that recombination can occur at defined locations in the plasmid, while no site preference for target sequences could be detected. Low homology between the recombining sequences indicates illegitimate recombination. The specificity of the plasmid sites could be explained by assuming a linear recombination intermediate, generated by cleavage of the transformed plasmid.

    Notes: We have developed an efficient transformation system based on the use of polyethylene glycol and CaCl2 for the biocontrol agents, Trichoderma spp. Trans formation was obtained with the plasmid pAN7-1, carrying a bacterial hygromycin-resistance gene as a selectable marker, under the control of Aspergillus nidulans heterologous expression signals. The system described here yielded 200-800 transformants per μg of DNA. Transformants contained several copies of the plasmid integrated into their genome, apparently at the same site in the different transformants analysed. Stability of the transformants was achieved by inserting a 2.4kb homologous DNA fragment into pAN7-1. Southern blot analysis indicated that integration in the stable transformants occurs through non-homologous recombination.

    Notes: A molecular karyotype for three different Trichoderma species (T. harzianum, T. viride, and T. reesei) was determined by using two different systems: countour-clamped electric-field and rotating-electrode electrophoresis. Six chromosomal DNA bands were observed in T. harzianum and T. reesei and five in T. viride. The sizes of these molecules were estimated by their mobility relative to the Schizosaccharomyces pombe chromosomes and ranged between 2.2 and 7.4 megabase pairs (mbp). The estimated genome sizes range from 31 to 39 mbp. A number of genes were located in the different chromosomes by means of Southern analysis. The implications of these findings are discussed.
