    Notes: Abstract Objective: A single cross-over, comparative pharmacokinetic study of oral and rectal formulations of 200 mg artesunic acid in 12 healthy Malaysian volunteers is reported. Methods: Plasma concentrations of artesunic acid and dihydroartemisinin were determined simultaneously by HPLC with electrochemical detection. The test drug was well tolerated and no undesirable adverse effects were observed. Results: Comparison of pharmacokinetic parameters of artesunic acid after oral and rectal administration showed statistically significant differences in t max and AUC, with no changes for C max and t 1/2. As for dihydroartemisinin, differences were observed for t max and C max but not for AUC. Conclusion: There appear to be pharmacokinetic differences between oral and rectal modes of administration. The significance of these findings should be explored in malaria patients before appropriate therapeutic regimens are devised.

    Notes: Abstract The 24 h variation in the passive avoidance response of rats was measured. Different groups of 18 animals were tested at 4 hourly intervals throughout 24 h. In all cases, retention was measured 48 h after the first trial. All animals were housed under controlled conditions for 14 days before testing. A significant 24-h rhythm in passive avoidance response was found, the maximum and minimum effects being in the middle of the light and dark phases respectively.

    Notes: Summary Ultrastructural analysis of specific heart granules in the left atrial myocardium of the rat shows that the so-called A and B types are not different populations but represent varying planes of section through a uniform set of organelles. Histograms of the apparent diameters of granules in individual animals establish that they are unimodal and tilt series of micrographs through selected granules show that the appearances of A and B profiles are interchangeable during the tilting manoeuvre. Moreover, serial sections reveal that equatorial sections through a granule yield the typical appearance of A profiles while peripheral sections through the same granule yield a B type appearance. Immunocytochemical studies were conducted on acrylic resin-embedded specimens using antiserum raised to ANP-28 and visualized by labelling with protein-A gold. A type profiles are more heavily decorated with gold particles than B profiles, a feature which could be partially explained if there is a higher concentration of peptide at the centre of the granule core. Some peripherally sectioned caps of granules which project B type profiles do not exhibit immunoreactivity because their orientation within the thickness of the section prevents access of the antiserum but, after resectioning at right angles to the original plane, their ANP content is revealed by a second phase of immunogold labelling.

    Notes: Summary There was no statistically significant difference in basal concentrations of immunoreactive atrial natriuretic peptide (ANP), as assessed by radioimmunoassay, between right and left atrial muscle of control rats; similarly, stereological analysis showed no statistically significant difference in the fractional volume of myocytes occupied by specific heart granules, or in numerical density of granules, between right and left atria. Nevertheless, correlated radioimmunoassay and ultrastructural investigations showed that the major source of elevated plasma levels of ANP after expansion of blood volume was the right atrium. Substantial expansion of blood volume caused an increase in the proportion of peripherally located granules in myocytes of both atria, but reduction in the number of granules and in the concentration and total content of ANP occurred in the right atrium only. Bilateral cervical vagotomy also caused a statistically significant elevation of plasma ANP concentration, accompanied by a statistically significant reciprocal reduction in right atrial ANP content; no statistically significant change occurred in left atrial ANP. When blood volume was expanded after bilateral vagotomy, there was a further statistically significant increase in plasma ANP concentration; this was accompanied by further reduction in right atrial ANP and, moreover, the combined manoeuvre also elicited a statistically significant reduction of ANP in the left atrium. Ultrastructural studies confirmed that, under these conditions, myocytes in both atria showed a marked depletion of specific heart granules.

    Notes: Each of the major cardiac valves (two arterial and two atrioventricular) is made up of a fibrous annulus with a characteristic configuration, and cusps or leaflets comprising a layer of endocardium folded over a fibrous lamina. Each of the arterial valves (aortic and pulmonary) has an annulus shaped like a three-pronged coronet to which are attached three equal-sized semilunar cusps. The arterial wall beyond each cusp forms a pocket or sinus which is crucial in the efficient closure of these valves. The coronary arterial orifices in the aorta lie high in two of the sinuses or above them and are unaffected by valve action. Narrowing of the annulus is a significant component of closure of each cardiac valve, more so for the atrioventricular valves than the arterial. Despite their traditional terminology, the left and right atrioventricular valves (mitral and tricuspid) both possess more than three leaflets each. Closure of these valves is not dependent on the number of leaflets and it is easiest to regard leaflet tissue as a continuous veil or skirt tapering towards the ventricles, where it is tethered to papillary muscles by means of chordae tendineae. Closure of these valves is biphasic, an incomplete phase in late diastole and complete closure during ventricular systole. Movement of atrioventricular leaflet tissue is slight as it is held down by tension of the chordae tendineae. © 1993 Wiley-Liss, Inc.
