    Notes: Summary The distribution and severity of the neuropathological changes in 57 cases of Alzheimer's disease, and 11 patients with Down's syndrome were investigated with reference to the cerebellum: A modified silver stain and a monoclonal antibody raised against amyloid β-protein were used to identify amyloid plaques. The highest incidence of amyloid plaques in the cerebellum (93%) was found in the group of patients who developed dementia before 65 years of age. This figure dropped to 56% in those patients with dementia beginning after 75 years. In 37 of these cases the distribution of the pathological changes of the disease were also examined in the brain stem. The severity of the pathological changes in the cerebellum corresponded to the involvement of the brain stem nuclei with connections to the cerebellar cortex. The possibility that the disease process spreads to the cerebellum by involving the fibres from the brain stem is discussed with reference to previous anatomical and neurochemical studies.

    Notes: Summary Pancreatic tissue from a case of neonatal hypoglycaemia with nesidioblastosis has been studied by routine light and electron microscope techniques and by highly sensitive light and electron microscope immunolocalization methods. A hyperplastic nodule within the pancreas from this case contained enlarged distorted haemorrhagic islets, with a variable rim of exocrine tissue. Islet cells in these areas were shown to contain more than one hormone in separate granules. An immunoperoxidase system using hapten-labelled primary antibodies and photochemical amplification applied to serial semithin sections suggested a consistent overlap between insulin and glucagon immunoreactive cells. Serial ultrathin sections of tissue embedded in LR White showed that some heteromorphous cells with predominantlyβ-granules also contained a minority population of granules which had either glucagon or glicentin immunoreactivity. In adjacent studies, the same techniques confirmed that the majority population of granules did indeed contain insulin, and immunocolloidal gold methods were used to show that glucagon and glicentin containing granules were present in the same cells. The significance of these findings is discussed, including the possibility that cells containing more than one granule type might represent a subpopulation of facultative cells in transit from producing one hormone to producing a second. The importance of sensitive immuno-electron microscopy in the investigation of endocrine lesions is stressed.

    Notes: Summary Light (LM) and electron (EM) microscope comparisons of the cytochemistry and immunocytochemistry of neuritic plaques and neurofibrillary tangles, the main histopathological changes in the brains of Alzheimer's disease sufferers, have been almost impossible because of the disparity between the two technologies. By embedding unosmicated brain tissue in the acrylic resin LR White, direct comparisons can be made between techniques applied at the LM level with those at the EM level. After partial dehydration in 70% ethanol, the tissue is embedded by rapid infiltration and polymerization at 0°C, which has been shown to maximally preserve tissue immunoreactivity. Semithin sections are then receptive to routine LM stains, silver stains e.g. Gomori's methenamine silver, and immunocytochemistry with immunoperoxidase or immunocolloidal gold. Serial thin from the use of a mouse monoclonal antibody against β-amyloid and a rabbit polyclonal antibody against ubiquitin are presented. LR White resin includes no elements other than carbon, oxygen and nitrogen, of which it is composed, so that sections of it are valuable for sensitive X-ray energy dispersive microanalysis.

    Notes: Abstract This study used biochemical and immunohistochemical methods to characterize the chondroitin sulphate-rich proteoglycans from human alveolar bone obtained from an oral source. Proteoglycans were extracted from bone by a sequential 4 m guanidine HCl extraction process, and purified by DEAE-ion exchange chromatography. SDS-PAGE and Western blot analysis, using CS-56 monoclonal antibody, demonstrated one major proteoglycan species with a core protein of 58 kDa, glycosaminoglycan chains of 45--66 kDa and a mean molecular weight of 205 kDa. This work confirmed the biochemistry of chondroitin sulphate-rich proteoglycans from a novel source of adult human alveolar bone, and pointed towards a proteoglycan with a high glutamate, glycine, aspartate, alanine, serine and leucine content. Sections of alveolar bone were embedded in LR White resin, labelled with CS-56 antibody and examined with the light and electron microscopes. At the light microscope level, labelling was restricted to the osteocyte lacunae and canaliculi. Ultrastructural observations showed that the labelling was localized to fine filamentous material in the walls of the osteocytes and canaliculi. Sparse labelling was associated with the collagen fibres immediately subjacent to the lamina limitans, but no labelling of the mineralized matrix was observed. These findings also indicated subtle differences in the distribution of chondroitin sulphate compared with previously reported work, which may indicate species or age differences in the samples used in this study.     Notes: Summary A simple and versatile technique for the preparation of ultra-thin sections, which can be stained immunohistochemically directly on electron microscope grids, is presented. An anti-hapten immunoperoxidase procedure has been adapted for use on tissue fixed in a purified monomeric glutaraldehyde-picric acid mixture, and embedded in ‘L R White’, a recently formulated plastic resin. This plastic tolerates the use of partial dehydration of tissue, resulting in higher antigenic yields. In addition, no etching of ultra-thin sections is necessary, and the whole immunostaining procedure can be completed in less than 2 h. A comparison of commonly used fixatives is discussed. High-resolution micrographs showing general staining (uranyl acetate-lead citrate) of rat pancreas, and immunostaining of insulin and TSH in storage granules in perfusion-fixed rat tissue and of lambda-chain immunoreactive cells in immersion-fixed human tonsil are included as examples.
