    Notes: Abstract Polarised targets as used in high energy physics experiments may be of considerable interest in biological structure research using polarized neutrons. So far, this promising method has been facing difficulties in getting reasonable polarization of the target nuclei. We report on a polarized “frozen spin” target which has been prepared from an enzyme dissolved in a mixture of heavy water and deuterated propanediol doped with a completely deuterated paramagnetic radical. Clusters of 700 protons defined by the structure of lysozyme embedded in a fully deuterated matrix were polarized to 75% within an hour by 4 mm microwave irradiation in a magnetic field of 2.5 tesla at a temperature of 0.3 K. The polarisation behaviour of biological targets can be compared to the best frozen spin target materials in high energy physics research.

    Notes: A cold atomic hydrogen beam source has been built and tested at BNL. The flow channel of the source, consisting of a discharge tube, a constricting nozzle, a thermal transition section, and an accommodator, was designed to operate in the laminar flow range. A novel feature of this source is a 0.3-mm gap between the transition Teflon tube which guides the flow and an accommodator which cools the flowing gas; across this gap the wall temperature may step from 100 K in the Teflon to 3 K in the accommodator. A record pulsed H° beam flux of 9.4×1018 H° sr−1 s−1 was obtained at an accommodator temperature of 5.8 K. The gas dynamic analysis of the system is in excellent agreement with the experimental results.

    Notes: Results from a novel variation on the Stern–Gerlach method for magnetic separation are presented. A superconducting solenoid lens for spin selection and focusing of a cold atomic hydrogen beam has been built and tested at Brookhaven National Laboratory. The solenoid consists of three coils connected in series, with the current in the outer two coils counter to the current in the middle coil, resulting in a large magnetic field (up to 5.2 T) with strong gradients. Beam density enhancement (focusing) of up to a factor of 10 was achieved. Excellent qualitative agreement exists with ray-tracing calculations. Quantitatively, the experimental results show a lens performance which is a factor of at least 3 better than what these calculations predict.

    Notes: Polarized neutron scattering from dynamically polarized targets has been used for the study of hydrogenous materials in several laboratories. A new variant, which is less dependent on specific deuteration, is proposed. This is based on the observation that a radio frequency (RF) field swept over a selected part of an NMR line depolarizes the spins depending on their spatial distribution in and around paramagnetic molecules. An RF field swept over a narrow frequency interval has little effect on polarized nuclear spins in the paramagnetic centers, whereas remote nuclear spins can be depolarized. When applying the sweep over a much larger frequency range, all spins are depolarized. This was observed both by NMR and by polarized neutron scattering after selective depolarization of the proton spins in a solid solution of protonated EHBA–CrV, Na(C12H20O7CrV).D2O, in a mixture of deuterated glycerol and water. The decay time of proton spin polarization in domains associated with the paramagnetic centers varies between 5 h in a deuterated solvent and less than 1 min in a protonated solvent. The size of such a domain is barely larger than 1 nm. The same observation is also made when the nuclear polarization is reversed by adiabatic fast passage. If the RF sweep is limited to frequencies close to the central peak of the proton NMR line, the proton spins far away from the paramagnetic centers are reversed, whereas those of EHBA–CrV and close to the centers maintain their original polarization. This method may become a useful tool in neutron scattering once the paramagnetic centers can be fixed as spin labels to well defined sites in a macromolecule, preferably in a crystal lattice.

    Notes: Bovine serum albumin (BSA) was dissolved in a mixture of deuterated glycerol and heavy water. The clusters formed by the 3300 proton spins in each BSA molecule were dynamically polarized up to P = 40%. Spin-contrast variation in small-angle neutron scattering was studied at several target polarizations. Zero contrast, and hence minimum polarized neutron small-angle scattering, is expected at PH = 60% from extrapolation of present data. The three basic scattering functions of spin-contrast variation look very similar because the shape of the BSA molecule and its proton distribution are congruent. Neutron small-angle scattering of BSA is similar to X-ray small-angle scattering at room temperature, indicating no deterioration of the molecular structure of BSA on solidification.

    Notes: A new polarized target for neutron scattering has been designed by CERN and tested successfully using the reactor FRG-1 at the GKSS Research Centre. The nuclear spins are aligned with respect to the external field – parallel or antiparallel – by dynamic nuclear polarization (DNP). To avoid absorption of neutrons by 3He, the frozen solutions of biomolecules are immersed in liquid 4He which in turn is thermally coupled to the cooling mixture of 3He/4He of the dilution refrigerator. Compared with earlier experiments where the sample had been cooled directly by 3He, the rate of detectable neutrons increased by a factor of 30. Another factor of 30 is due to the installation of the cold source and the beryllium reflector in FRG-1. Polarized neutron scattering from apoferritin in deuterated solvent shows that the proton spin polarization is homogeneous in apoferritin molecules. After saturation of proton nuclear magnetic resonance (NMR), polarized neutron scattering is dominated by deuteron spin contrast. With the deuterated large subunit of E. coli ribosomes, three different basic scattering functions are derived from spin-contrast variation, reflecting the known scattering-length-density distribution of the architecture of rRNA and ribosomal proteins. The planned in situ structure determination of a mRNA fragment is discussed in the light of the present results.

    Notes: Contrast variation is a commonly employed method of structure analysis by small-angle scattering. Thermal neutrons and X-ray synchrotron radiation have led to a renaissance of non-destructive labelling methods like anomalous scattering and spin-contrast variation for the study of the in situ structure of macromolecular components. Anomalous scattering from sulfur and phosphorus has been used to identify ribosomal proteins and rRNA of the large subunit of the E. coli ribosome. For polarized neutron scattering from dynamic polarized targets, rRNA and the ribosomal proteins were deuterated separately and studied in deuterated and in undeuterated solvents. The structural parameters of rRNA and the whole of the ribosomal proteins obtained by these methods are in good agreement with earlier results obtained from neutron scattering in H2O/D2O mixtures.

    Notes: Abstract Hydrogen, deuterium, and carbon-13 nuclear spin systems have been studied in partially deuterated 1,2-ethanediol (CD2OH)2, doped with paramagnetic Crv complexes, between 0.1 and 0.5 K, using the technique of dynamic polarization. Various steady-state and transient measurements demonstrate the existence of a thermal contact between the different spin species of this sample and the electron spin-spin interaction reservoir. The lowest spin temperature attained was about 1.2 mK in a magnetic field of 25 kG, which corresponds to a proton polarization of 97%, to a deuteron polarization of 40%, and to a carbon-13 polarization of 48%.

    Notes: Abstract We have performed transverse fieldμSR experiments on several different samples of copper in the temperature range below 2 K, including isotope separated Cu and impurity doped polycrystalline Cu. We do not observe any strong effect of the isotope composition of the sample. A63Cu and a natural Cu sample of identical purity both yield 0.16μs−1 for the low-temperature plateau, while an increased linewidth in the65Cu case may be related to the strong effects of Fe impurities. Careful transverse field measurements on large single crystals at 0.08 K reveal non-Gaussian lineshapes in accordance with the picture of diffusing muons at this temperature. This allows us to reject several of the existing models for muon behaviour in copper below 2 K.
