    Notes: Abstract The evidence for an impact of platelet-derived 5-hydroxytryptamine (5-HT) on local tissue perfusion is reviewed. By interacting with 5-HT2 serotonergic receptors, 5-HT, directly or through amplification, activates platelets, endothelial and vascular smooth muscle cells producing platelet aggregation, vascular permeability increase and large vessel constriction. Pharmacodissection in experimental animals with selective serotonergic 5-HT2 receptor antagonists, e.g. ketanserin, shows that 5-HT largely contributes to the platelet-mediated increase in vascular permeability, to platelet-vessel wall interaction during hemostasis, to cardiopulmonary dysfunction provoked by thromboembolism and to the platelet-mediated inhibition of peripheral collateral circulation. Clinical results obtained with ketanserin further substantiate an involvement of platelet-derived 5-HT in the pathogenesis of impaired tissue perfusion in some cardiovascular conditions.

    Notes: Summary Isolated kidneys of Wistar rats and spontaneously hypertensive rats (SHR) were perfused with Tyrode's solution. In 2- and 6-month old SHR, the maximal increase in perfusion pressure caused by norepinephrine was higher than in 2- and 6-month old Wistar rats, but the sensitivity, as judged from the dose of the agonist required to reach 50% of the maximal response was the same. Both the maximal response and the sensitivity to serotonin were significantly augmented in 6-month old SHR and Wistar rats when compared to the young animals. This hypersensitivity was more pronounced in SHR than in Wistar rats. Infusion of serotonin potentiated the vasoconstriction induced by a bolus of norepinephrine. This amplification, due to activation of S2-serotonergic receptors, was more pronounced in the old animals. No amplification occurred when norepinephrine was infused instead of serotonin. Tachyphylaxis to the amplifying effect of serotonin was observed and was less pronounced in kidneys from old than from young animals. The amplifying effect of serotonin was inhibited by ketanserin at concentrations which did not, or only moderately, inhibit the response to norepinephrine.

    Notes: Summary Oxatomide is a new potent inhibitor of anaphylactic and allergic reactions. After oral administration, the compound both inhibits the release of endogenous histamine and prevents the effects of exogenous histamine, at comparable doses. The combination of these effects appears to be the basis of the effectiveness of oxatomide in allergic reactions and may lead to clinical applications different from classical antihistaminics and from cromoglycate.

    Notes: Abstract A fully automated system to quantify different parameters of gastrointestinal motility and gastroduodenal co-ordination in conscious dogs was designed and built around a personal technical computer (HP 9816). Online it performs sampling of contractions on four different sites of the digestive tract for two dogs simultaneously, data reduction, drift correction and storage of data on hard disk. Offine it performs baseline determination, peak detection, calculation of motility parameters such as amplitude and interval, plus co-ordination between gastric and duodenal motility and presentation of the results in both tabular and graphic form. To illustrate the possibilities and advantages of the computer analysis the early postprandial motor patterns for antrum, duodenum and jejunum were quantified during the first hour after administration of a small test meal.

    Description / Table of Contents: Sommaire Ordinairement, les désordres du rythme cardiaque sont traités avec des médicaments qui augmentent la période réfractaire du muscle cardiaque. Les changements dans la période réfractaire du muscle cardiaque peuvent être étudiés avec la stimulation du pouls simulé. On décrit un nouvel instrument qui est capable de stimuler un muscle papillaire isolé, soit couplé soit seul, avec une séquence programmée des stimuli et avec recyclage automatique. La période réfractaire apparaît comme une indication digitale, en millisecondes, et le résultat digital peut être alimenté dans d'autres ordinateurs digitaux. Le stimulateur-ordinateur est facilement programmé et possède des nombreuses caractéristiques qui sont applicables aussi dans d'autres domaines de la médicine et de la biologie.
    Abstract: Zusammenfassung Unregelmässigkeiten im Herzrhythmus werden mit Medikamenten behandelt, welche die Refraktärperiode des Herzmuskels vergrössern. Änderungen in der Refraktärperiode des Herzmuskels können durch gepaarte Impulsstimulierung untersucht werden. Es wird ein neues Instrument beschrieben, das fähig ist, einen isolierten, papillären Muskel in dem gepaarten und einzelnen Modus mit einer programmierten Reihe von Stimuli und mit automatischer, erneuter Kreislaufbildung zu stimulieren. Die Refraktärperioden erscheinen als eine Zahlenanzeige in Millisekunden, und die digitale Auslese kann andere digitale Computer speisen. Der “Anreger” Computer wird leicht programmiert und hat viele eingebauten Einrichtungen, die auch auf anderen Gebieten der Medizin und Biologie verwendbar sind.
    Notes: Abstract Disorders of cardiac rhythm are commonly treated with drugs which increase the refractory period of heart muscle. Changes in the refractory period of heart muscle can be studied with paired pulse stimulation. A new instrument is described which is capable of stimulating an isolated papillary muscle in the paired and single mode with a programmed sequence of stimuli and with automatic recycling. The refractory period appears as a digital indication in milliseconds and the digital read-out can be fed into other digital computers. The stimulatorcomputer is easily programmed and has many built-in features which are also applicable in other fields of medicine and biology.
