    Notes: The outstanding semidwarf spring barley cultivar ‘Diamant’ was derived from the cv.‘Valticky’ by X-ray mutagenesis. More than 120 European spring barley varieties trace back to ‘Diamant’. This variety and nine German spring barley cultivars having ‘Diamant’ in their pedigree, as well as four cultivars not related to ‘Diamant’, were analysed by amplified fragment length polymorphisms (AFLPs) and simple sequence repeats (SSRs) in order to identify genomic regions associated with traits derived from ‘Diamant’. The total number of SSRs tested was 122, of which 88 were polymorphic taking all varieties into account, with 51 for ‘Valticky’ and ‘Diamant’, i.e. 42.8%. A varying level of polymorphism was detected on individual chromosomes (11.1–78.6%). In addition, AFLP revealed a high level of polymorphism. Out of the total number of 1591 bands, 670 were polymorphic for all varieties studied with 182 for ‘Valticky’ and ‘Diamant’, i.e. 11.4%. Based on SSR and AFLP data UPGMA cluster analysis was carried out, but no grouping according to the relatedness with ‘Diamant’ or ‘Valticky’, respectively, was found. It is concluded from these results that the mutagenic effect leading to the development of ‘Diamant’ out of ‘Valticky’ affected many genetic loci and is thus larger than expected.

    Notes: Based on the RAPD marker OP-C04H910 which is closely linked to the barley mild mosaic virus (BaMMV) resistance gene rym9 derived from the variety ‘Bulgarian 347’ the marker STS-C04H910 cosegregating with OP-C04H910 and generating a single additional band on plants carrying the recessive resistance encoding allele has been developed. Furthermore, the simple sequence repeats (SSRs) WMS6 and HVM67 have been integrated into the genetic map of the rym9 region on chromosome 4HL. Because of their close linkage to rym9 and distinct banding pattern STS-C04H910 and HVM67 are well-suited for marker- assisted selection, enhanced backcrossing procedures and pyramiding of resistance genes.

    Notes: In order to obtain an overview of the genetic diversity present within the set of pea cultivars released in Germany, 21 cultivars were analysed at the DNA level by random amplified polymorphic DNAs (RAPDs) and amplified fragment length polymorphisms (AFLPs), as well as for agronomic traits. Yield of grain cultivars ranged from 2.95 to 3.87 t/ha. Based on the screening of 60 RAPD primers and 32 Eco RI + 3/Mse I+3 AFLP primer combinations, 20 RAPD primers and 11 Eco RI + 3/MseI+ 3 primer combinations generating polymorphic and distinct fragments were chosen for estimation of genetic diversity. Twenty RAPD primers amplified a total of 314 scorable bands ranging from about 262 bp to 1996 bp. Of these, 175 fragments (55.7%) were polymorphic. Based on these data, genetic similarity (GS) was estimated between 0.80 (‘Lisa’ vs.‘Grapis’) and 0.94 (‘Bohatyr’ vs. ‘Sponsor’; mean GS = 0.88). Eleven AFLP primer combinations led to the amplification of 949 scorable fragments ranging from 43 to 805 bp and of these, 462 (48.7%) were polymorphic. Genetic similarity based on AFLPs was calculated between 0.85 (‘Lisa’ vs.‘Laser’) and 0.94 (‘Bohatyr’ vs. ‘Sponsor’, mean GS = 0.90). Correlation of genetic similarity estimated on RAPDs and AFLPs was estimated at r = 0.79** using Spearman's rank correlation coefficient and at r = 0.84 by the Mantel test, respectively. UPGMA cluster analysis carried out on these data separately for RAPDs and AFLPs and on the combined data reflected, to some extent, pedigree relationships and cophenetic correlations (r = 0.89 for RAPDs, r = 0.88 for AFLPs, and r = 0.93 RAPDs + AFLPs) indicate a good fit of respective clusters to genetic similarity data. The correlation of cluster analyses to pedigree information and the impact on parental genotype selection is discussed.

    Notes: The inheritance of durable resistance of selected spring barley varieties to Rhynchosporium secalis was investigated. Data from the F2 generation of a 4 × 4 diallel, without reciprocals and the F4 generation of three crosses selected out of this diallel, suggest that resistance in this sample of varieties tested is complex in inheritance. Significant additive effects were detected indicating that the resistance level of barley cultivars may be improved by the hybridisation of suitable varieties. However, the genes conferring resistance seem to be concealed by the expression of one completely dominant resistance gene in our set of varieties. These results are partly in conflict with previous results on the inheritance of resistance to R. secalis in the breeder's line ‘11258/228613A’ indicating that the effectiveness of this resistance gene may be greatly influenced by the genetic background of the current population of R. secalis.

    Notes: Experiments were carried out in order to identify RAPD-markers for the BaMMV/BaYMV resistance gene ym4 to facilitate efficient marker-based selection in practical breeding programmes. Linkage analysis was carried out with 283 doubled haploid (DH) barley lines out of a cross between the BaMMV/BaYMV susceptible cv. ‘Igri’ and the resistant cv. ‘Franka’. Using bulked segregant analysis, 310 RAPD-primers were screened for polymorphism between the parents. Four of these primers gave rise to specific bands linked to the resistance gene ym4.

    Notes: Summary In order to obtain information about the mode of inheritance of BaMMV resistance in germplasms carrying genes different from the “German” gene ‘ym4’ f1-tests for resistance as well as F2-segregation analyses of crosses to susceptible German cultivars were carried out by mechanical inoculation in the greenhouse. In the f1 the majority of plants of each combination tested reacted susceptible to BaMMV while in the F2 a good fit to a segregation of 1r:3s or 7r:9s was observed. Therefore, the results of these tests revealed that the BaMMV resistance of all the varieties tested is inherited either by a single or by two recessive genes. By testing intercrosses of these resistant varieties segregation of BaMMV-susceptile plants was observed in the majority of combinations, revealing a high degree of genetic diversity in the barley gene pool.

    Notes: Abstract  The genetic structure of the rym5 locus was studied in a population comprising 391 doubled-haploid lines that were evaluated for resistance to two strains of Barley Yellow Mosaic Virus (BaYMV-1, 2) and to Barley Mild Mosaic Virus (BaMMV). The absence of recombinants that are able to differentiate between the reaction to these different bymoviruses provides evidence that rym5 is a complex locus, which is either composed of several closely linked genes or of an allelic series of a single gene. For marker-assisted introgression of this locus into adapted barley germplasm, a CAPS (cleaved amplified polymorphic sequence) and a microsatellite marker were developed that flank the gene at distances of 0.8 and 1.3% recombination, respectively.

    Notes: Abstract  In the present study three novel genes from barley accessions 10247 (ym8), Bulgarian 347 (ym9), and Russia 57 (ym11), which confer resistance to Barley Mild Mosaic Virus (BaMMV), were mapped using molecular markers. Bulked segregant analysis of four progenies segregating for resistance to BaMMV was followed by fine-scale mapping of the resistance genes using individual F2 or BC1F2 plants. The resistance genes are inherited recessively and are located on the long arm of barley chromosome 4HL. A series of closely linked molecular markers are available for marker-assisted breeding programs. A marker (MWG2134) linked with resistance gene ym11 from Russia 57 was identified, which is diagnostic for the resistance gene.

    Notes: Abstract  Thirty-six Hordeum vulgare varieties and 12 H. spontaneum germplasms originating from different parts of the world and showing different reactions to the barley yellow mosaic virus complex (BaMMV, BaYMV, BaYMV-2) were analyzed for genetic similarity using RAPDs. On the basis of an analysis of 20 selected RAPD-primers corresponding to 544 bands genetic similarity according to Nei and Li (1979) was estimated to be between 0.685 and 0.964. Associations between the 48 genotypes were calculated using UPGMA-clustering and principal coordinate analysis. By applying these methods we were able to separate H. spontaneum accessions from H. vulgare varieties, and within these groups all the genotypes were clustered correctly according to their origin. Consequently, RAPD analysis can be considered a very useful and efficient tool for the fast estimation of genetic relationships in barley. The correlation between genetic similarity with respect to German varieties and adaptation of exotic barley varieties to German growing conditions is discussed.

    Notes: Summary To assess the possibilities offered by isozymes to locate resistance genes against barley mild mosaic virus (BaMMV), the isozyme patterns of 19 barley (Hordeum vulgare L.) genotypes carrying genes different from ym4 were determined. Of the 15 isozyme systems tested, only three were polymorphic, namely aconitate hydratase, esterase, phosphogluconate dehydrogenase, providing markers on four of the seven barley chromosomes. Studies of F2 progenies derived from three crosses between resistant genotypes and susceptible varieties failed to reveal linkage between resistance genes and isozymes. Another goal of the experiment was to study the linkage relationships between ym4 and the esterase locus (Est1-Est2-Est4). Our estimates of the recombination rate between these two loci (3.41 and 8.32%) were in the range of those reported between these esterases and one of the resistance genes of the Chinese variety Mokusekko 3.
