Zusammenfassung Eine erste Übersicht über die Grundlagen und Anwendungen eines neuen Verfahrens zur Spurenanalyse von Metallen wird gegeben. Die Vorteile dieser von unserer Arbeitsgruppe entwickelten Methode zur Spurenanalyse von Metallen liegen in der geringen Analysenmenge, der hohen Empfindlichkeit und Selektivität, der einfachen Probenzubereitung für die Messung (kein Aufschluß notwendig) und der Zuverlässigkeit und Genauigkeit der Ergebnisse. Der Zeitbedarf für eine komplette quantitative Analyse liegt unter 30min. Die Konzentration monoisotopischer Metalle, wie zum Beispiel Aluminium, Cäsium, Mangan, usw., wird mit Hilfe einer Eichkurve bestimmt. Mit der Anwendung der Isotopenverdünnungsanalyse für die Quantifizierung von Metallen mit mindestens zwei stabilen Isotopen läßt sich die Genauigkeit der Ergebnisse weiter steigern. Bei diesem Verfahren wird einerseits die Umwelt nicht mit radioaktivem Material belastet, andererseits können selbst geringste Konzentrationsänderungen der Spurenmetalle in biologischem Material sicher nachgewiesen werden. Die Richtigkeit der erhaltenen quantitativen Daten wurde bei Testmessungen mit anderen analytischen Methoden, wie Atomabsorptionsspektroskopie und Thermionen-Massenspektrometrie abgesichert. Obwohl die größten analytischen Möglichkeiten der Felddesorptions-Massenspektrometrie auf dem Gebiet der hochmolekularen Naturstoffe liegen, ist es in den letzten Jahren möglich geworden, das Verfahren so zu modifizieren, daß mehr als 60 Metalle qualitativ und quantitativ untersucht werden konnten. In Tierversuchen konnte die Felddesorptions-Massenspektrometrie bei der Bestimmung der toxischen und teratogenen Wirkung von Thallium auf Ratten und Mäuse erfolgreich angewandt werden. Weiterhin konnten die Konzentrationsänderungen von Lithium, Rubidium, Magnesium, Calcium, Strontium und Barium in mütterlichen und fetalen Körperflüssigkeiten und Geweben von Kaninchen während der reproduktiven Phase bestimmt werden und wurden mit den verschiedenen Gestationsstadien korreliert. Einen interessanten Gesichtspunkt, der auf eine vorübergehende Essentialität von Spurenmetallen deutet, liefern die Resultate, daß Lithiumionen eine entscheidende Rolle bei der Skeletausbildung spielen und Rubidium charakteristisch mit der Milchproduktion zusammenhängt. Beim Menschen wurde dieses Verfahren erstmals für die Quantifizierung von Lithium in Körperflüssigkeiten unter physiologisch normalen Bedingungen und nach therapeutischer Gabe von Lithiumsalzen sowie für Calciumabsorptionsstudien angewandt. Wichtige Anwendungen für medizinische Fragestellungen waren die Untersuchung der zeitabhängigen Ausscheidung von Elektrolyten und Spurenmetallen im Urin von Multiple-Sklerose-Patienten sowie die Feststellung von Elektrolyt- und Spurenmetallkonzentrationen im Liquor cerebrospinalis von gesunden Probanden- und Multiple-Sklerose-Patienten. Bei diesen Untersuchungen gelangen die ersten Bestimmungen der Normalkonzentrationen und biologischen Varianzen von Lithium, Strontium und Barium in der menschlichen Hirnflüssigkeit.

Summary A first survey on the basic aspects and applications of a novel method for trace analyses of metals is given. The advantages of this methodology for analyses of trace metals which was developed by our group are: small sample amount, high sensitivity and selectivity, simple sample preparation for the measurement (no ashing) and reliability and precision of the results. The time consumption for one complete quantitative analysis lies below 30 min. The concentration of monoisotopic metals, as for example aluminum, cesium, manganese etc. is determined using a calibration curve. Using stable isotope dilution analysis quantification of metals with at least two stable isotopes further improved the precision of the results. If this technique is utilized, on one hand contamination of the environment by radioactive substances is avoided, on the other even the smallest changes in concentrations of trace metals are detected unambigeously. The accuracy of the resulting quantitative data has been confirmed by test measurements with other analytical methods such as atomic absorption spectroscopy and thermal ionization mass spectrometry. Although there is no doubt that the greatest analytical capacity of field desorption mass spectrometry is in the field of high-molecular weight natural products, it has been possible in the last years to modify the method for qualitative and quantitative investigations of more than 60 metals. In animal experiments field desorption mass spectrometry has been utilized successfully for the determination of toxic and teratogenic actions of thallium on rats and mice. Further the concentration changes of lithium, rubidium, magnesium, calcium, strontium and barium in maternal and fetal body fluids and tissues of rabbits during the reproductive phase were determined and correlated with different stages of gestation. Interesting aspects which point to a temporary essentiality of trace metals, are the facts that lithium ions play an important role during bone formation and rubidium ions are characteristically connected with the production of milk. In humans the method was applied for the first time for the quantification of lithium in body fluids under physiologically normal conditions and after therapy with lithium salts as well as for calcium absorption studies. Important applications for medical problems included investigations of the timedependent excretion of electrolytes and trace metals in urine of multiple sclerosis patients and the assay of these elements in cerebrospinal fluid of healthy humans and multiple sclerosis patients. These studies lead to the first determinations of the normal concentrations and biological variances of lithium, strontium and barium in human cerebrospinal fluid.
