    Cytokine mRNA decay is accelerated by an inhibitor of p38-mitogen-activated protein kinase (1999)

    Notes: Abstract. Objective: To identify the site(s) in tumor necrosis factor (TNFα), interleukin-6 (IL-6), and macrophage inflammatory protein-1α (MIP-1α) biosynthesis that is blocked by SB202190, a selective inhibitor of p38-mitogen activated protein kinase (p38). ¶Materials: Human blood monocytes isolated by centrifugal elutriation. ¶Methods: Monocytes were stimulated with lipopolysaccharide in the presence of 0, 0.3, 1 and 3 μM SB202190. Induced TNFα, IL-6, and MIP-1α protein and mRNA were measured by ELISA and quantitative RT-PCR, respectively. The half-lives of cytokine mRNA levels were determined following treatment of cells with actinomycin D or SB202190. ¶Results: SB202190 suppressed 〉60% of lipopolysaccharide-induced TNFα, IL-6, and MIP-1α protein and mRNA expression. Suppressed mRNA levels could be attributed to a 〉2 to 7-fold reduction in cytokine mRNA half-lives. In contrast, SB202190 did not destabilize mRNAs encoding interferon-induced gene 15 protein and glyceraldehyde-3-phosphate dehydrogenase. ¶Conclusions: Specific mRNA destabilization represents an important and novel site of action for the cytokine suppressive effects of p38 inhibitors.

    Notes: Abstract Two-dimensional and three-dimensional Green-Naghdi (GN) models equipped with a numerical wave-absorbing beach have been developed to simulate nonlinear, regular, and irregular wave propagation. The numerical beach is introduced near the downstream boundary to absorb outgoing waves. An appropriate amount of numerical damping and an appropriate length of numerical beach are investigated using numerical experiments. The results show that the GN models with a numerical beach work very well in simulating wave propagation in water in a small computational domain.

    Isolation and partial characterization of a full-length cDNA clone for 3α-hydroxysteroid dehydrogenase: A potential target enzyme for nonsteroidal anti-inflammatory drugs (1991)

    Notes: Abstract Homogeneous 3α-hydroxysteroid dehydrogenase (3α-HSD; EC 1.1.1.50) of rat liver cytosol is a monomeric (MR 34000) NAD(P)+ dependent oxidoreductase which displays 9-, 11- & 15-hydroxyprostaglandin dehydrogenase activity. The enzyme is potently inhibited by nonsteroidal anti-inflammatory drugs (NSAIDs), suggesting that 3α-HSD may be a target enzyme for NSAIDs. A monospecific, polyclonal anti-sera raised against the purified enzyme was used to screen a λgt11 expression library and oligonucleotide probes complementary to the 5′ and 3′ ends of immunopositive clones were used to isolate a 2.1 kb full-length cDNA. Digestion of the full-length cDNA with Eco RI generated two fragments of 1.1 and 1.0 kb in length. Both fragments were subcloned into pGEM3 and partially sequenced. The 1.1. kb fragment contains the C-terminus of 3α-HSD which was confirmed by an in-frame stop codon and comparison of the predicted amino acid sequence to peptide sequence obtained from two endo lys-C peptides of 3α-HSD. The 1.0 kb fragment is 5′ to the 1.1 kb fragment and is sufficient in length to contain the remainder of the entire open reading frame for 3α-HSD. Dideoxysequencing reveals significant sequence homology with bovine lung prostaglandin PGF2α synthase. These findings support the role 3α-HSD in inflammation and suggest that hydroxysteroid dehydrogenases, hydroxyprostaglandin dehydrogenases and prostaglandin F2α synthase may be members of a common gene family.

    Notes: Zusammenfassung Es wurde das Fließverhalten einiger Perbunanlatices in einem Konzentrationsbereich von 0,5 bis 47,3% und in einem Temperaturintervall von 18 bis 35°C eingehend untersucht. Unterhalb von 25% verhalten sich die Latices wieNewtonsche Flüssigkeiten. Bei höheren Konzentrationen hängt ihr Fließverhalten von der Schubspannung ab: Bis etwa 100 dyn · cm−2 zeigen die Emulsionen ebenfalls dasNewtonsche, darüber hinaus jedoch strukturviskoses Verhalten. Die relative bzw. spezifische Viskosität erwiesen sich imNewtonschen Bereich bei allen Konzentrationen als temperatur-unabhängig. Die Meßergebnisse lassen sich durch eine modifizierteRobinson-Beziehung beschreiben. Mit Hilfe einer einfachen Arbeitshypothese, daß die Latexteilchen durch eine dünne immobilisierte Flüssigkeitsschicht (Hydrathülle) vergrößert werden, gelingt es, unsere sämtlichen Meßergebnisse aus demNewtonschen Bereich sowie einige aus der Literatur bekannte Daten über das Fließverhalten von Naturlatex durch eine gemeinsame Kurve darzustellen.

Ein spezieller Ausgleichsvorgang — die „Rückfederung“ der Substanz bei der Entlastung — wird eingehend behandelt, und die Anwendung dieses Vorganges für die Bestimmung der Relaxationszeit einer elasto-viskosen Substanz, sowie zur Prüfung der Frage, ob und inwiefern es sich dabei um das Maxwellsche Verhalten handelt, diskutiert. Entsprechende, von W. Meskat durchgeführte experimentelle Messungen werden anschließend besprochen.

    Notes: Zusammenfassung Die elastoviskosen Eigenschaften von Flüssigkeiten und Gelen werden bekanntlich unter anderem-mit Hilfe von Schwingungs-Elastoviskosimetern untersucht. Ein derartiger Test spiegelt nicht unmittelbar die interessierenden Eigenschaften der Prüfsubstanz, sondern das mechanische Verhalten des aus der Prüfsubstanz und der Meßapparatur bestehenden Simultansystems wider, welches durch die Massenträgheit der Apparatur und der Prüfsubstanz beeinflußt wird. Dieses mechanische Verhalten des Systems wird in der Arbeit theoretisch eingehend untersucht (das ebene Problem), und die exakten Auswertungsformeln zur Bestimmung der elastoviskosen Eigenschaften der getesteten Substanzen werden entwickelt. Ferner wird der Resonanztest diskutiert und, fußend auf einer Approximation durch eine äquivalenteMaxwell-Substanz, ein Fluchtlinien-Nomogramm zur Berechnung des Verlustwinkels im Resonanzpunkt angegeben. Die aufgestellten theoretischen Beziehungen wurden zur Auswertung einiger experimenteller Ergebnisse in einem Frequenzbereich von 10–103 Hz herangezogen.

Die Arbeit schließt mit einer kritischen Diskussion desOstwald de-Waeleschen Reibungsgesetzes.
