    Notes: Summary In this immunocytochemical study, the brains of nine squirrel monkeys (Saimiri sciureus), ranging from 8 to 27 years of age, were examined for the presence and distribution of β/A4 amyloid, a 4-kilodalton peptide. In aged squirrel monkeys, amyloid is associated primarily with intracerebral and meningeal capillaries and arterioles and occurs to a lesser degree as small and/or diffuse deposits in the neural parenchyma and in the dense cores of senile plaques. Cerebrovascular amyloid is found primarily in neocortex, amygdala, and septum verum and is rare or nonexistent in other subcortical gray structures, white matter, cerebellum, and spinal cord; this pattern of localization is comparable to that in humans with cerebral amyloid angiopathy. There is a significant correlation between cerebrovascular and parenchymal deposits of amyloid. However, cerebrovascular amyloid is always the most abundant form in squirrel monkeys, even in cases of severe cerebral amyloidosis. In contrast to squirrel monkeys, aged rhesus monkeys (Macaca mulatta) develop, mostly parenchymal deposits of amyloid and have relatively less vascular amyloid. This species difference in the histological distribution of amyloid suggests that separate mechanisms may influence the accumulation of amyloid in cerebral blood vessels and in the neural parenchyma. These data also indicate that the squirrel monkey can serve as a model for investigations of cerebrovascular amyloidosis.

    Notes: Summary Patients with Alzheimer's disease (AD) and some aged controls may have diminished functions of the pineal gland. In this immunocytochemical study, we stained pineal glands from cases of AD and young and aged controls for cytoskeletal elements and amyloid. We found no evidence of neurofibrillary tangles (NFT) or the accumulation of neurofilaments, tau, A68, or β/A4 amyloid deposition in pinealocytes or associated structures in cases of AD or controls. In both AD and controls, we observed dense immunoreactivity for phosphorylated neurofilaments in marginal plexuses associated with processes of pinealocytes, boutons, and knob-like endings. The accumulation of phosphorylated neurofilaments in the processes of pinealocytes appears to be a normal morphological characteristic of the pineal gland and may not represent a pathological change.

    Notes: Neutron diffraction measurements have been performed on liquid equiatomic potassium–lead alloy at T=870, 900, and 930 K to determine the influence of temperature on the structure. A well-defined first sharp diffraction peak (FSDP) at Q=0.96 A(ring)−1, similar to that found in glassy materials, was obtained in the total structure factors S(Q) at the three temperatures. While the position of the peak remained unaffected by temperature changes, its magnitude decreased with increasing temperature. The other features of S(Q) were almost unaltered by temperature, as expected. The FSDP can be related to the presence of chemical bonds or clusters whose stability decreases with increasing temperature. A molecular model based on the random packing of K4Pb4 structural units is shown to give a remarkable correspondence with the measured structure factors.

    Notes: The x-ray structure factor of water measured under ambient conditions with synchrotron radiation is compared with those predicted on the basis of partial structure factors describing the nuclear positions obtained by neutron diffraction and of different assumptions for the electron distribution. The comparison indicates that a charge of approximately 0.5 e is transferred from each hydrogen atom to the oxygen on the same molecule, implying an effective dipole moment of 2.9 D, in good agreement with theoretical estimates. © 2000 American Institute of Physics.

    Notes: Amyloid precursor proteins (APP) are a member of a larger family of proteins that include the amyloid precursor-like proteins (APLP) APLP1 and APLP2. We have examined the expression and metabolism of APLP2 and document that APLP2 is expressed at high levels in the nervous system and in peripheral tissues. Furthermore, several APLP2 isoforms encoded by alternatively spliced transcripts are posttranslationally modified by a chondroitin sulfate glycosaminoglycan (CSGAG) chain. Furthermore, CSGAG modification is regulated by the insertion of sequences encoded by an alternatively spliced exon. Notably, expression of the CSGAG form of APLP2 appears restricted to embryonic neurons and mature neuronal populations that undergo regeneration, such as olfactory sensory neurons. Thus, differences in posttranslational modifications between the APLP2 isoforms and APP are likely to underlie differences in the regulation and function of these homologues. Our present efforts are directed towards using gene targeting strategies to disrupt the expression of the mouse APP/APLP2 genes to define the normative roles of the encoded molecules in development, plasticity, regeneration, and repair.

    Notes: Wide-angle neutron diffraction measurements have been performed on single-phase and phase-separated high-silica glasses in the system Li2O–SiO2. The structure factors of the single-phase glasses vary modestly with increasing Li2O concentration compared to the changes observed when Na2O or K2O is added to SiO2, and vestiges of the intermediate-range order characteristic of v-SiO2 are retained to 33 mol % Li2O. The structure factors of single-phase and phase-separated glasses are identical to within 1%–2%, indicating that phase separation results in very minor disruptions to local atomic arrangements. When combined with evidence obtained from previous studies, these data indicate that single-phase high-silica glasses in this system contain compositional heterogeneities analogous to those produced by phase separation. © 1995 American Institute of Physics.

    Notes: The structure of polyethylene oxide (PEO) was investigated by neutron scattering in both semicrystalline and liquid states. Deuterated samples were studied in addition to the protonated ones in order to avoid the large incoherent scattering of hydrogen and identify features in the pair correlation functions attributable to C–H pairs. Analysis of the deuterated sample gave additional information on the C–O and C–C pairs. The results are compared with molecular-dynamics simulations of liquid PEO. © 1998 American Institute of Physics.
