    Notes: Abstract: A nonganglioside factor(s) present in Sigma types II and III mixed bovine brain ganglioside preparations synergises with suboptimal amounts of serum to induce proliferation specifically in nondividing B103 neuroblastoma cultures. The active substance is nondialys-able and soluble in water as well as in chloroform-methanol mixtures of 1:1–4:1 (vol/vol). It is completely insoluble in ether and acetone at room temperature. Biological activity survives heating to 70°C in the presence of 0.1 M HC1 for 1 h as well as boiling at neutral pH. Loss of activity occurs on heating to 70°C for 1 h with 1 M HCl or 1 M NaOH. The activity is insensitive to digestion with neuraminidase, trypsin, pronase, and phospholipases A2 and C. The factor cochromatographs with gangliosides on Dowex AG 50W and Sephadex G100 and is partially recovered with GMI on DEAE-Sepharose, but may be isolated in a ganglioside-free fraction by sequential chromatography on Sephadex LH20 and silicic acid columns. The substance(s) has the properties of a water-soluble proteolipid protein, the amino acid composition being reported. It is not immunologically cross-reactive with antibodies to GMI ganglioside or the major proteolipid protein of myelin.

    Notes: Neurotransmitter choice is a crucial step in neural development. In the cerebral cortex, pyramidal neurons use the excitatory neurotransmitter glutamate, whereas non-pyramidal cells use the inhibitory neurotransmitter GABA. We are interested in how these two neuronal types are generated. We labelled precursor cells from embryonic rat cerebral cortex with a retroviral vector in dissociated cell cultures, and examined the neurotransmitter phenotype of their progeny immunohistochemically after 2 weeks in vitro. We discovered, first, that precursor cells in culture generate glutamatergic and GABAergic neurons in proportions similar to those in vivo. Second, we found that neuronal precursor cells gave rise to both GABAergic and glutamatergic neurons. These results suggest that neuronal precursor cells in the cerebral cortex have the potential to generate both neuronal subtypes. Moreover, these data are consistent with a stochastic model of neurotransmitter specification.

    Notes: Abstract  Organ cultures from neonatal rabbit retinae grew well over periods of up to 2 weeks in vitro. Proliferation in vitro declined in parallel with the decline seen in vivo, although the rate of proliferation in the explants was slightly reduced. The proliferation of progenitor cells in vitro produced the same cell types produced postnatally in vivo. Postnatally generated cell clones, labeled by means of a retroviral vector, consisted mainly of rods and Müller cells. The layers of the retinae developed as in vivo; an outer plexiform layer occurreed after the first 2 days in vitro. Ultrastructurally, ribbon synapses (outer and inner plexiform layer) and conventional synapses (inner plexiform layer) were observed. The photoreceptor cells grew well-developed inner segments and cilia but no mature outer segments. The cultured retinae contained a well-developed, regular lattice of Müller cells expressing vimentin as in vivo. The neuron-to-Müller cell-ratios were essentially the same as in vivo, viz. about 15 to 16 neurons, among them about 10 to 11 (rod) photoreceptor cells per Müller cell. When the glia cell-specific toxin α-aminoadipic acid (αAAA) was applied, the pattern of vimentin-positive Müller cells became irregular, or even locally missing. In such cases, the tissue became disorganized as indicated by a local disappearance of the regular layering, and development of many rosettes. It is concluded that an intact lattice of Müller cells is necessary for the migration of young neurons, and for correct formation of retinal layers.

    Notes: Abstract The Cuyahoga River is a heavily polluted tributary of Lake Erie located in N. E. Ohio (USA). One hundred seventy taxa of diatoms were identified from seven locations along the Cuyahoga River. The most frequently collected diatoms from each station were Cyclotella meneghiniana, Gomphonema parvulum, Navicula cryptocephala, N. cryptocephala var. veneta, N. lanceolata, N. menisculus, N. minima, N. minuscula, N. pelliculosa, Nitzschia amphibia, N. ovalis, N. palea, and Rhoicosphenia curvata. In the most heavily damaged region of the river these widely-distributed taxa were the only frequently collected diatoms. The relatively undamaged section of the river contained 94% more taxa than the most heavily damaged section. Chief among these additional taxa were Achnanthes lanceolata, A. lanceolata var. dubia, Cocconeisplacentula, Fragilaria vaucheriae, Melosira varians, Meridion circulare, Navicula mutica var. tropica, N. symetrica, Nitzschia dissipata, Stephanodiscus astraea, and Synedra rumpens. The degree of similarity between diatom assemblages along the Cuyahoga River as measured by a modification of Sørensen's index of similarity in species-frequency of occurrence composition was related to the degree of chemical-physical water quality similarities and to the extent of similarity between areas of geologic and biologic substrates. The most dissimilar diatom assemblages usually occurred between locations that were the most dissimilar with respect to chemical-physical water quality conditions rather than to dissimilarities with respect to sediment-substrate composition.
