    Notes: Summary The connecting cilium of rat retinal rods was studied by freeze-fracture and thin-sectioning techniques. Transverse strands of intramembranous particles could be observed on fracture face B of the ciliary plasma membrane. The strands were essentially similar to those found at the transitional zone of motile cilia (“ciliary necklace”). The large number of intramembranous particles obscured the pattern on fracture face A of the membrane. On longitudinal sections of the cilia, beads showing a periodicity similar to the necklace strands were observed. Each bead consisted of two structures apposed to both sides of the plasma membrane. Transverse sections of the cilia revealed radial Y-shaped structures that connected each ciliary doublet with the plasma membrane. Axial tubules, central sheath, radial spokes and dynein arms were missing in the connecting cilium. Comparing the fine structure of the retinal cilia with that of motile cilia it becomes evident that the connecting cilium is analogous in structure with the transitional zone of motile cilia. The present observations suggest that periodic membrane beads along the plasma membrane on thin sections correspond to strands of necklace particles as observed on freeze-fractured membranes. The arrangement of the particles in transverse strands is probably ensured by the radial connecting structures.

    Notes: Summary Nerve terminals in the palisade zone of the rat median eminence were investigated with freeze-fracture electron microscopy. Fracture face P of the specific terminals showed two populations of intramembranous particles (IMP): a large and a small variety. The large IMP-s often formed small irregular clusters. In nerve terminals the total number of both populations of IMP-s was considerably less than that found on P membrane faces of ependymal feet. On fracture face E of the nerve terminals, the number of IMP-s was about a quarter of that seen on fracture face P. On both fracture faces of most terminals a few small round impressions (or elevations respectively) were found which may be interpreted as broken necks of either exo- or endocytotic vesicles. Neither gap nor tight junctions occurred at lateral membranes of the specific axon terminals. Similarly, no membrane specializations were observed with freeze-fracturing on membrane areas adjacent to the basement membrane. The findings are discussed in relation to a possible exocytosis mechanism of the hypothalamic releasing and inhibiting hormones.

    Notes: Summary The epithelial surface of the taste disc on the fungiform papillae of the frog was studied by means of freeze-fracture and conventional electron microscopy. The plasma membrane of the apical process of taste receptor cells was found to exhibit a uniform population of 10-nm intramembrane particles on its fracture face P in a density of 1,000–1,800/μm2. In contrast to intramembrane particles in mucous cells or in other regions of the plasma membrane, these particles were observed to protrude approximately twice as high from the fracture face. Not more than 1–2% of the particles were found on fracture face E of the apical process of the taste cell. The apical regions of satellite and mucous secretory cells are also described. The occurrence of relatively large intramembrane particles in a membrane region presumed to function as the chemoreceptor area of taste cells suggests that the particles may be involved in chemoreception.

    Notes: Summary The microvillous border of normal, developing and degenerating planarian retinal clubs was examined using osmium and glutaraldehyde fixation. Well-developed normal photoreceptors contained well preserved microvilli both after osmium and glutaraldehyde fixation. Osmium-fixed regenerating and degenerating retinal clubs showed serried vesicles and anastomosing tubules at the site of the microvillous border. After glutaraldehyde fixation, chains of vesicles were absent and the photoreceptor consisted of a regular array of microvilli. This difference indicates the selective sensitivity of the photoreceptor membranes in regenerating and degenerating retinal clubs to the action of osmium tetroxyde.
