    Notes: Summary Nine patients were studied 1.5–3 years after jejuno-ileostomy for obesity by an intravenous glucose infusion technique designed to imitate blood glucose concentrations after glucose ingestion. Whereas serum insulin and gastrin concentrations were normal, blood glucose concentrations were significantly depressed compared to preoperative levels as well as to levels in matched normal subjects. Thus, in the fasting state mean concentrations (± S.E.M.) of blood glucose, serum insulin and gastrin in the patients were, respectively, 3.3±0.2 mmol/l, 95±22 pmol/l and 38±4 pmol/l. The corresponding concentrations in the matched normals were 4.3±0.2 mmol/l, 70±18 pmol/l and 39±6 pmol/l. The glucose concentrations in the patients were low in all situations, i. e. in the fasting state, after oral glucose ingestion and during the intravenous glucose infusion. The results indicate that jejuno-ileostomy in obesity greatly facilitates peripheral glucose disposal. The mechanism behind this phenomenon is not yet known.

    Notes: Conclusion In the nephrectomized rat gastrin (and pentagastrin) does not evoke acid secretion but retains its ability to activate gastric hirtidine decarboxylase. The results suggest that the kidney is important for metabolism and excretion not only of gastrin but of humoral antagonists of gastrin-induced acid secretion as well.

    Notes: Summary In adult rats islet cell neogenesis can be stimulated by partial duct ligation. Duct to islet cell differentiation is thought to be regulated by growth factors such as gastrin and transforming growth factor-α (TGFα). To test this hypothesis, we examined the expression of gastrin and TGFα at the mRNA and protein level in pancreatic tissue following partial duct ligation. Pancreatic specimens were investigated on days 3, 5, 7 and 14 after duct ligation by means of non-isotopic in situ hybridization, immunocytochemistry and Western blotting. Gastrin mRNA was strongly expressed in newly developed duct-like cell structures in the ligated tail portion of the pancreas before the period of pronounced islet cell neogenesis (days 5 and 7), and immunostaining for gastrin peptides was positive at days 5–7. In the non-ligated head portion and in control pancreases, gastrin was not expressed. Expression of TGFα was found to be increased in the ligated tail portion of the pancreas on day 3 and particularly on day 5, while there was no enhanced signal in the non-ligated part. Western blotting revealed two different TGFα isoforms (18 kDa and 42 kDa) in the ligated tail part and three isoforms (18 kDa, 24 kDa and 42 kDa) in the non-ligated head part and in untreated pancreases. The induction of gastrin and TGFα expression preceded the peak in the bromodeoxyuridine pulse labelling index of beta cells, known from a previous study to occur on day 7. We conclude that pancreas duct ligation induces the overexpression of gastrin and TGFα in the first days following ligation. Since ductal cells are known to give rise to endocrine cells after duct ligation, gastrin and TGFα may play a role as growth factors in islet neogenesis. [Diabetologia (1997) 40: 887–893]

    Notes: Summary The insulin response to oral and intravenous glucose was measured in ten patients after resection of antrum, duodenum, proximal jejunum, and the head of pancreas (Whipple's operation). Compared to matched normal subjects the operation reduced neither the total nor the gut hormone induced part of the insulin response to oral glucose. The results suggest, that hormones from the first part of the intestinal tract are not necessary as incretins.

    Notes: Summary CSF concentrations of vasoactive intestinal polypeptide (VIP), cholecystokinin (CCK), noradrenaline (NA) and dopamine (DA) were measured in the lateral ventricles and at the lumbar level in patients with normal pressure hydrocephalus (NPH). The concentrations of VIP (n=15), NA (n=10) and DA (n=10) were significantly higher at the lumbar level than at the ventricular level, whereas the concentrations of CCK (n=9) were similar at the two sites. A signifikant positive correlation between the concentrations measured at the two levels was found for VIP (rs=0.65; p⩽0.01) and DA (rs=0.94; p⩽0.001). The results indicate that the concentrations of transmitter substances measured in CSF at the lumbar level not necessarily are indicative for concentrations measured more centrally. The negative correlations between Evans ratio and L-CSF VIP (rs=-0.76; p ⩽ 0.001), and between resistance to outflow and V-CSF as well as L-CSF CCK (rs=-0.75); p ⩽0.05) might be explained by a reduction in number of cortical neurons or by disturbances in CSF dynamics in patients with NPH.

    Notes: Abstract: The subcellular localization of in vivo synthesized cholecystokinin (CCK) in different parts of the rat brain was studied after intracisternal pulse injections of [35S]methionine. The rats were decapitated 1 h after the injection, and the brain was divided into cortex, hippocampus and remainder. Subcellular fractions were obtained according to Whittaker's method. De novo synthesized CCK in the crude mitochondrial-synaptosomal fraction, P2, and in the purified synaptosomal fraction was demonstrated by affinity chromatography, using antibodies specific for the COOH-terminal sequence of CCK. By subsequent gel chromatography two molecular forms of labelled CCK occurred, with elution constants, Kav, of 1.1 (corresponding to the COOH-terminal octapeptide) and of 1.40 (a component which may correspond to the COOH-terminal tet-rapeptide amide, CCK-4). The findings support the idea that the small molecular forms are the transmitter forms of CCK.

    Notes: Abstract: The regional distribution of cholecystokinin-like immunoreactivity (CCK-LI) in postmortem human brain material has been investigated. CCK-LI was concentrated particularly in the forebrain (all cerebral cortical areas, amygdala, and hippocampus), with smaller amounts in the basal ganglia, hypothalamus, and periventricular grey. Lowest amounts of CCK-LI were found in the thalamus, cerebellum, and spinal cord.
