    Notes: Summary In cats the effect of Metergoline (Liserdol, Farmitalia) was investigated on the performance of a delayed response task and was related to the associated changes in the concentrations of dopamine, noradrenaline and serotonin in the brain. Metergoline (14 μg/kg/day, i.m.) administered during the first 10 days of the training significantly decreased response latencies at delay periods of 0, 3, 9 and 27 sec without significantly affecting the number of errors. Biochemical assays showed that the improved performance correlated with the increased dopamine content of limbic structures and the decreased serotonin content in all brain structures. The concentrations of dopamine in the neostriatum remain unchanged. Response latencies and the number of errors in cats treated with Metergoline were significantly different from those in animals treated with L-DOPA (30 mg/kg/day) probably reflecting the specific biochemical disturbance induced by the two drugs. These results suggest an inhibitory influence of serotoninergic pathways on areas like the meso-limbic system retaining high concentrations of dopamine.

    Notes: Une diminution significative de la sérotonine dans toutes les structures du cerveau est démontrée chez le chat suìte à un traitement chronique à la p-chlorophénylalanine (250 mg/kg i.p., 2 doses, à intervalles de 72 h). Les concentrations de noradrenaline ne sont pas affectées par un tel traitement. Par ailleurs, les concentrations de dopamine sont significativement augmentées dans le lobe piriforme, le septum, l'hypothalamus et le mésencéphale et normales dans. le néostriatum, le cortex frontal, le tuberculum olfactorium et le cervelet. Des chats porteurs d'une lésion sélective du noyau raphae dorsalis, sans atteinte des autres noyaux, démontrent une baisse significative de la sérotonine du néostriatum et du lobe piriforme. Comparativement aux chats traités à la p-chlorophénylalanine, les chats lésés présentent une augmentation significative des concentrations de la dopamine du lobe piriforme alors que cells du néostriatum demeurent normales. Ces résultats permettent de dissocier les structures appartenant au système limbique des structures appartenent au système moteur extrapyramidal à la fois sur une base biochimique et morphologique. De plus, ces données suggèrent que. le rôle physiologique de la dopamine du système moteur extrapyramidal est fonctionnellement différent du rôle de la dopamine impliquée dans les structures du système limbique.〈section xml:id="abs1-2"〉〈title type="main"〉AbstractCats treated chronically with p-chlorophenylalanine (250 mg/kg, i.p., 2 doses, 72 h apart) demonstrate a significant decrease in serotonin concentrations in all structures within the brain. The noradrenaline level was not affected. Dopamine concentrations were however significantly increased in the piriform lobe, septum, hypothalamus and mesencephalon and unchanged in the neostriatum, frontal cortex, tuberculum olfactorium and cerebellum. In cats bearing a selective lesion of nucleus raphae dorsalis without involving other raphae nuclei, a significant decrease in serotonin was observed in the neostriatum and piriform lobe. Compared to p-chlorophenylalanine treated cats, lesioned cats show a significant increase in dopamine content in the piriform lobe but not in the neostriatum. A single dose of p-chlorophenylalanine (250 mg,kg, i.p.) as well as a selective lesion of nucleus raphae centralis superior caused a significant decrease in serotonin level without affecting dopamine concentration. Such findings allow differentiation of a group of structures belonging to the limbic system from those of the extrapyramidal motor system on a biochemical as well as on a morphological basis. Moreover these results suggest that dopamine in the nigrostriatal pathway has a different function from dopamine involved in the limbic system.

    Notes: Abstract— Pyridoxine (50mg/kg, per os) given for 7 consecutive days did not modify the content of dopamine, noradrenaline, and serotonin in the neostriatum of the brain 3, 6 and 18 h after the last dose, but significantly increased DOPA/5HTP decarboxylase activity in both the neostriatum and liver. The administration of l-DOPA and pyridoxine (100 and 50mg/kg, per os, respectively) together for 7 days increased DOPA/5HTP decarboxylase activity in the brain to the same extent as did l-DOPA and pyridoxine given individually. Liver DOPA/5HTP decarboxylase activity remained normal when both drugs were administered together. However it decreased significantly after l-DOPA administration for 7 days but not after pyridoxine treatment. In cats under treatment with l-DOPA for 7 days, actinomycin D given for the final 3 days prevented the increased DOPA/5HTP decarboxylase activity induced by l-DOPA in the neostriatum and mesencephalon but had no effect on the enzymatic activity in the liver. These findings indicate that differences exist between brain and liver DOPA/SHTP decarboxylase activity in uivo. In addition, denatured supernatant from livers of animals treated with l-DOPA contained a dialysable compound which inhibits DOPA/SHTP decarboxylase activity in the supernatant from livers of untreated cats. In animals who received pyridoxine along with l-DOPA, no such inhibitor was found. These results may explain the mechanism by which l-DOPA exerts its beneficial effects and why pyridoxine administered with l-DOPA reduces the therapeutic effectiveness of l-DOPA in Parkinson's disease. These findings are consistent with the possibility that a tetrahydro-isoquinoline derivative formed in vivo in the liver after l-DOPA therapy for 7 days might affect DOPA/5HTP decarboxylase activity in the liver but not in brain. A tetrahydroisoquinoline derivative did not appear to be formed when l-DOPA and pyridoxine were administrated together suggesting that pyridoxine protected the enzyme and favored a more rapid degradation of l-DOPA peripherally with less l-DOPA available for the CNS.

    Notes: Abstract We present an exact solution of the anomalous chiral Schwinger model using Fermionic variables. We implement infrared regularization by considering the model on a spatial circleS 1. Quantum effects modify the gauge constraints through the appearance of Schwinger terms in the gauge algebra. We perform a careful analysis of the resulting second class gauge constraints by implementing Dirac's method at the quantum level and obtain the spectrum of the theory. We get a consistent unitary Lorentz invariant theory for particular values of the counterterms. We find that when we regulate the fermionic sector of the model without reference to the gauge fields Lorentz invariance requires that we add both Lorentz variant and gauge variant counterterms.
