    Notizen: Summary Using high performance liquid chromatography, the presence of N6-methyladenine has been found at a level of 0.1 mol percent in DNA extracted from Penicillium chysogenum. No 5-methylcytosine was detected. A mutant strain HP547, which is sensitive to the lethal effects of N-methyl-N1-nitro-N-nitrosoguanidine, methylmethane sulphonate and the base analogue 2-aminopurine shows an increased spontaneous mutation rate and no detectable DNA methylation. Comparison of restriction enzyme digests of wild type and undermethylated strains indicated that methylation was occurring at a different sequence to that of the Dam methylase system of E. coli.

    Notizen: Abstract An analytical model for thermal damage of retinal tissue due to absorption of laser energy by finite-sized melanin granules is developed. Since melanin is the primary absorber of visible and near-IR light in the skin and in the retina, bulk heating of tissue can be determined by superposition of individual melanin granule effects. Granules are modeled as absorbing spheres surrounded by an infinite medium of water. Analytical solutions to the heat equation result in computations that are quick and accurate. Moreover, the model does not rely on symmetric beam profiles, and so arbitrary images can be studied. The important contribution of this model is to provide a more accurate biological description of submillisecond pulse exposures than previous retinal models, while achieving agreement for longer pulses. This model can also be naturally extended into the sub-microsecond domain by including vaporization as a damage mechanism. It therefore represents the beginning of a model which can be applied across the entire pulse duration domain.

    Notizen: Abstract The effect of irrigation with water at salinity concentrations of 2.6 and 5.2 dS m−1 on the growth of pure swards of six cultivars of white clover (Trifolium repens L.) was examined over three irrigation seasons at Tatura, Victoria, Australia. After two irrigation seasons, soil EC e levels increased to 6 dS m−1 at 0–60 cm depth in the higher salinity treatment resulting in highly significant (p 〈 0.001) reductions in shoot dry matter production, flowering densities and petiole and stolon densities. These saline conditions also increased (p 〈0.001) concentrations of Cl and Na in the shoots and reduced (p 〈 0.001) leaf water potentials and canopy photosynthetic efficiency rates especially at high temperatures. In contrast, root growth increased at shallow depths (0–15 cm) under both saline irrigation treatments (p 〈0.001). Cultivars differed significantly in salt tolerance (p 〈 0.001), with cultivars Haifa and Irrigation exhibiting superior tolerance in terms of lower reductions in herbage yield (p 〈0.05) and petiole densities (p 〈0.001) during one irrigation season and lower concentrations of Na and Cl in the shoots (p 〈0.05) compared with the other four cultivars (Aran, Kopu, Pitau and Tamar). In addition, canopy photosynthetic efficiency rates (A *) in plots irrigated with water at 5.2 dS m−1 were higher in cultivar Haifa compared with cultivar Tamar (p 〈0.05). The salt tolerance ranking obtained for the six cultivars was in broad agreement with earlier greenhouse studies. Consequently, it appears that, while white clover is an extremely salt-sensitive species, it is possible to grow cultivars which display greater salt tolerance than other cultivars and which provide some scope to increase, or at least to maintain, pasture yields in areas where the soil salinity is low to moderate or where pumped saline groundwater is re-used for Irrigation.

    Notizen: Abstract Efforts to develop new crop varieties with improved salt tolerance have been intensified over the past 15–20 years. Despite the existence of genetic variation for salt tolerance within species, and many methods available for expanding the source of genetic variation, there is only a limited number of varieties that have been developed with improved tolerance. These new varieties have all been based upon selection for agronomic characters such as yield or survival in saline conditions. That is, based upon characters that integrate the various physiological mechanisms responsible for tolerance. Yet over the same time period, knowledge of physiological salt responses has increased substantially. Selection and breeding to increase salt tolerance might be more successful if selection is based directly on the physiological mechanisms or characters conferring tolerance. Basic questions associated with using physiological selection criteria are discussed in the paper. These are centred around the need for genetic variation, the importance of the targeted mechanism, the ease of detection of the physiological mechanism (including the analytical requirements) and the breeding strategy. Many mechanisms, including ion exclusion, ion accumulation, compatible solute production and osmotic adjustment have been associated with genetic variation in salt tolerance. Yet their successful use in improving salt tolerance, via physiological selection criteria, is largely non-existent. Consideration is given to the role of physiological criteria in the short and long term in improving salt tolerance. In several glycophytic species, particularly legumes, physiological selection based on ion exclusion from the shoots shows promise. Recent results for white clover indicate the potential for using a broad physiological selection criterion of restricted Cl accumulation in the shoots, with scope for future refinement based upon the specific physiological characters that combined result in ion exclusion.

    Notizen: Abstract Two divergent populations of T. repens cv. Haifa developed from two generations of recurrent selection for shoot chloride concentration, were grown in the greenhouse at 0 and 40 mol m−3 NaCl. Over two harvest cycles at 40 mol m−3 NaCl, the population selected for a low concentration of chloride in the shoot maintained a significantly lower chloride and sodium concentration compared with those plants selected for a high shoot chloride concentration. The distribution of chloride in the shoots was further examined in a subsample of plants from both populations. In all plants, concentrations of chloride were lower in the expanding and fully expanded leaves than in the older leaf tissue or petioles. While there were no significant differences in the photosynthetic rates between lines, shoot yields and relative leaf expansion rates were higher in the low chloride population. Plant death was greater in plants selected for high shoot chloride. These results suggest that selections based on measurements of low shoot chloride concentrations may be successful in developing a cultivar of T. repens with improved salt tolerance.
