    Notes: Summary Unitary K+ currents in single cells isolated from ventricular muscle of newborn rat hearts were measured in response to different potentials and [K] o . TheI/V curves were linear for potentials more negative than the zero-current voltage: especially in high [K] o (150nm KCl), no clear outward currents could be detected indicating a drastic rectification in the inward direction. The channel is mainly selective to K+ but Na+ ions are also carried (P Na/P K=0.056). The channel conductance is proportional to the square root of [K] o but Na+ ions seem to have a facilitatory effect on γK, the single-channel conductance. The channel activity, measured asP o, i.e. the probability to find the channel in open state, decreased as the membrane was hyperpolarized. This behavior was tentatively explained by an inactivation process as the membrane becomes more negative. The rate constants of the transitions between the different states were calculated according to a C−O−C model. A control of the gating process by permeant ion K+ was postulated, based on the increase of one of the rate constants from the closed to the open state with [K] o . Finally, the macroscopicI/V curves calculated fromP o and δi, the unit current, were found to be characteristic of a ion-blocked inward rectifier.

    Notes: Abstract Sarcoplasmic reticulum (SR) vesicles were prepared from either canine or sheep heart and fused into lipid bilayers to study their ionic channels. A 92±5 pS anion-selective channel was recorded in asymmetric 50 mm trans/250 mm cis CsCl buffer system. Reversal potentials and theoretical equilibrium potentials for Cl− ions obtained under various experimental conditions allowed us to confirm the Cl− selectivity of this SR channel. The majority (69%) of channel recordings (n = 45) displayed steady-state kinetics and a slight voltage dependency of the open probability. However, 31% of the channels inactivated after their incorporation. We now report that the channel might be reactivated by depolarizing voltage steps. Furthermore, the use of either PKA or PKG in association with adequate phosphorylating buffers lengthens the deactivation process at the end of the voltage pulses, but does not prevent the inactivation. It was assumed that the change in gating mode was due to a voltage-sensitive association/dissociation mechanism with a phosphorylated protein of the SR membrane such as phospholamban (PL). We demonstrated that a specific monoclonal antibody raised against canine PL inhibited the activity of the channel and prevented its reactivation by depolarizing steps. 400 to 800 ng/ml of Anti-PL Ab consistently and sequentially turned off the channel activities. In contrast, heat inactivated Anti-PL Ab had no effect. We propose that phospholamban may be a primer of the SR Cl− channel whereby Cl− anions would play the role of counter-charge carrier during rapid Ca2+ release and Ca2+ uptake by the SR.

    Notes: Abstract The behavior of single chloride channels in sarcoplasmic reticulum of rabbit and trout skeletal muscle was examined by fusing isolated vesicle fractions into planar lipid bilayers. The channel exhibited a full open state with a unit conductance of 65 pS (in 100 mM Cl-) and several subconductance states with reversal potentials which were dependent on the chloride gradient across the bilayer. Open probability was 0.6–0.95 for membrane potentials ranging from-60 to+60 mV. The kinetic behaviour could be described by assuming one time constant for the fully conducting channel, and at least two time constants for the non-conducting channel. In the presence of methane sulfonate, sulfate and phosphate anions, a decrease in the unit current amplitude but not open time argued in favor of a competition between these anions and Cl- at the transport site of the channel. Chloride channel activity was not affected by variations of Ca2+ concentration in both chambers or by the presence of Mg2+. Similarly, neither millimolar ATP nor the presence of the drugs taurine (up to 10 mM), lidocaine (2–40 μM) or the calmodulin antagonist W7 (5–150 μM), modified channel behavior. Finally, pH variations between 6.8 to 8 were without effect.

Characteristics of non-optimized pH-stat continuous stirred tank reactors withL. bulgaricus andS. thermophilus entrapped in the same or separate particles were examined. In these reactors the highest production rate were 9.4 g.l−1.h−1 for lactic acid and 3.4×1011 C.F.U. 1−1.h−1 for inoculation when microorganisms were entrapped separately. A stable balance of the yoghurt bacteria liberated in continuously prefermented milk was observed.

    Notes: Dipeptidyl peptidase II (DPP II), E.C. 3.4.14.2, a serine class endopeptidase, is widely used as a lysosomal marker in cytochemical studies. To date most ultrastructural studies of ameloblasts use the presence of acid phosphatase activity to identify cellular organelles to be lysosomal. Using decalcified rat mandibles, with kidney tissue as a positive control, DPP II activity, was assessed with specific substrate Lysyl-alanine-4-methoxy-2-naphthylamide in ameloblasts at an ultrastructural level. Reaction product (RP) indicative of DPP II activity was observed only within lysosome-like organelles. These RP-labelled organelles were only localized in the supra- or para-nuclear regions of the ameloblasts, which corresponds with previous studies using acid phosphatase cytochemistry. However, in contrast with these studies, RP was not detected in the distal region of the ameloblasts, viz., in the Tomes' processes of the secretory ameloblasts or near the ruffled border in the maturation ameloblasts. The transitional ameloblasts were notable for the intensity of staining of their RP-labelled organelles. We propose that DPP II may have a role in programmed cell death which is thought to occur in this transition zone.Biochemical analysis of rat incisor enamel organ homogenates, indicated tissue fixation resulted in an 82% reduction in DPP II activity, although the specific activity of DPP II was not affected. © 1992 Wiley-Liss, Inc.
