    Notizen: Summary Treatment of Kluyveromyces cells with several polycations partially neutralized their negative charge, even after washing; chitosan at 0.5 mg/ml rendered the zeta potential positive. Adhesion of the cells to various supports (glass, polycarbonate, polystyrene) was promoted by treating the cells or the support with chitosan; this confirms the importance of electrostatic interactions in the adhesion process. When the support was treated, a dense and regular monolayer of adhering cells was obtained; when the cells were treated the adhering layer was more heterogeneous due to floc formation. Cell ghosts of K. lactis were prepared by CHCl3/ethanol permeabilization and glutaraldehyde treatment. Treating the ghosts with chitosan provided immobilization of 1.9 to 7.3×106 ghosts per cm2 according to the support; treating the support allowed to obtain 5.5 to 5.7×106 ghosts per cm2. The lactase activity of the immobilized ghosts was found to be comparable to that of ghosts in suspension, i.e. in the range of 0.9 to 2.2 U/mg dry cell weight, at 30°C, corresponding to 0.02 to 0.14 U/cm2 of support. A test in microreactor with a lactase solution showed that 50% of the initial activity was still found, after about 25 days of continuous operation at 30°C.

    Notizen: Abstract. Saccharomyces cerevisiae LBG H620 and DSM 2155 strains were continuously cultivated under carbon (C)-limited, phosphorus (P)-limited and nitrogen (N)-limited growth conditions. Cell and protein concentrations in feed, foam, and residue as well as the degree of cell recovery and the rate of foaming were measured, and the concentration and enrichment factors were evaluated at different dilution rates (D). The LBG H620 cells were reduced, while the DSM 2155 cells were enriched in the foam. The highest concentration factors in DSM 2155 cells were attained if they were cultivated under strong P-limitation at a low D. Fairly high concentration factors were also found under C-limitation. Under N-limitation, low concentration factors were found with low Ds. At the beginning of the continuous cultivations, all of the cells were recovered, but with advancing time the degree of recovery and cell concentration and the enrichment factor ratio diminished. The cellular properties of the yeast were characterized by flow cytometry, and the surface properties by measurements of their hydrophobicity, electrophoretic mobility, and chemical composition (using X-ray photoelectron spectroscopy, XPS). These investigations indicated that the large difference in flotation between the two strains is due to different surface properties. Strain DSM 2155 has higher surface hydrophobicity and lower electrokinetic potential. Cell wall properties and the cell flotation depend on medium composition and age of the culture.

    Notizen: Summary Immobilization of Saccharomyces cerevisiae as a monocellular layer adhering to a glass support has been achieved by adsorbing Al or Fe(III) ions on the cells or by coating the support with Al(OH)3 or Fe2O3 particles. The initial rate of glucose to ethanol conversion was measured in pure glucose solutions for free cells in suspension, either untreated or submitted to the chemical treatments used for immobilization, and for immobilized cells, using a small reactor (50–70 ml) in closed circuit. The conversion rate of immobilized cells is in the range of 3.5 to 5×10-14 mol/cell·h while that of untreated suspended cells is in the range of 5 to 7×10-14 mol/cell·h.

    Notizen: Abstract The ability of Klebsiella oxytoca NRRL-B199 to use either lactose or the mixture of glucose and galactose as substrate for the production of 2,3-butanediol was studied in batch fermentations with different conditions of aeration and pH. 2,3-butanediol was undetected, or present in minute concentration in the fermentation broths with lactose, while it was the main product from glucose+galactose with final concentrations of up to 18.8 g/l in media at pH 6.0. Under conditions optimal for 2,3-butanediol synthesis, when aeration limited growth, the rate of biomass growth was more tightly related to the aeration rate in lactose medium than in glucose+galactose medium. These relations suggest that the growth rate is very low on lactose but still considerable on glucose+galactose when aeration rate tends toward zero. Correspondingly, the metabolism is more oxidative in the former medium, yielding mainly acetate as product.

    Notizen: Klebsiella oxytoca (NRRL-B199), although able to produce 2, 3-butanediol from glucose, converted lactose mainly into acetic acid. By addition of a preparation of lactase (β-galactosidase, EC 3.2.1.23), the fermentation of lactose in a stirred vessel was three-times faster and resulted in a high concentration of 2, 3-butanediol. The lactase confined in dead cells of Kluyveromyces lactis (CBS 683) was prepared by permeabilization with solvents and fixation with glutaraldehyde. The cells were coimmobilized by adhesion to glass wool after treatment of the latter with chitosan, which ensured cell-support electrostatic attraction. The cell loading (dry weight) was ca. 9 gL-1 for the yeast and ca. 2 gL-1 for the bacteria. In the presence of culture medium, the adhesion of both cells was stable and the bacteria tended to form biofilms. The stability of the coimmobilized cells was demonstrated by the continous conversion of lactose into 2, 3-butanediol at 30oC during 25 days. The coimmobilization system gave output concentrations (14 gL-1) and rate of production (1 gL-1 h-1) of 2, 3-butanediol from lactose, similar to those obtained in the literature with immobilized cells and glucose. Compared to the literature data on direct conversion of lactose using pure cultures, the present results showed higher butanediol concentrations and 10 to 100 times higher rates of production.

    Notizen: The adsorption of aluminum ions by Saccharomyces cerevisiae has been investigated by determining adsorption isotherms and electrophoretic mobility. The adsorption of aluminum ensures a neutralization of the cell surface charge and allows adhesion of the cells to glass and polycarbonate. Glass slides have been taken as a negatively charged model support, allowing the authors to study in detail the process of adhesion. The cells are simply pretreated by an aluminum solution near pH 4. Bringing the Al-pretreated cells in contact with the support by sedimentation and washing the support and sediment makes it possible to obtain a single, dense, regular layer of cells adhering strongly to the support. Adhesion can also be obtained from a suspension flowing parallel to a vertical support, provided the flow velocity is sufficiently small; the amount of cells immobilized per unit support area is about one-half that obtained by sedimentation. The immobilized cells show a specific activity for ethanol production from glucose which is similar to cells in suspension.
