Notes: Thymic immunosuppression in adult Xenopus laevis laevis, the South African clawed toad, is antigen-dependent and antigen-specific, but it is not genetically restricted. In this report, we show that combination in vitro with a thymus from an immunized Xenopus laevis laevis can suppress antibody production to a hapten from spleen fragments, if the spleen fragments are derived from a subspecies of Xenopus laevis or a species of Xenopus which shares the same diploid chromosome number (2N = 36). Some aspects of the evolution of thymic immunosuppression are considered in the light of these results.

Notes: Summary Temperature treatment of sera of erythrocyte immunized adults of 3 amphibian species has shown that cooling enhances antigen-antibody complexing and does not alter complement function in vitro. These findings bear on understanding in vivo temperature immunosuppression in poikilotherms.

Notes: Implants were made into forelimbs of Triturus viridescens using fresh, frozen and boiled kidney and liver of T. viridescens and R. pipiens. Limbs were recovered at intervals up to 70 days post-implantation.Kidney implants from Wisconsin R. pipiens gave twice as many extensive accessory structures as did Vermont frog kidney. Total induction percentages, however, were similar.Quantitative and qualitative parameters for implant-induction of accessory structures were investigated. The decrease in antigenicity and increased rate of cytolysis of frozen implants resulted in increased similarity between frog and newt kidney in rate and pattern of breakdown and in rates of induction. Modification of rate and duration of the release of the stimulating factor from the implant did not result in induction by liver implants. No evidence was found for any increase in innervation prior to or coincident with blastema formation. Implantation and implant cytolysis may cause hypersensitivity of limb tissues to the normal innervation pattern or trophic stimuli from the implant may act with those from the injured limb tissues to produce growth. The general pattern of host reaction to the implanted material was studied and described.

Notes: Abstract Spontaneous and induced cancers are rare in non-isogeneic or inbred amphibians. Neoplastic cells become immortalized through loss of a normal capacity to die by apoptosis. Mature lymphocytes of mammals require activation and entry into the cell cycle in order to become susceptible to apoptosis. Whether Xenopus lymphocytes differ from mammalian lymphocytes in this regard is examined. In vitro exposure of PMA, or its analogue, MPMA, to adult splenocytes of Xenopus laevis was used to affect apoptosis. Flow cytometric analysis of FITC-Annexin V/propidium iodide (PI) fluorescence (apoptosis) and BrdU uptake (DNA synthesis) were assayed concurrently in the same lymphocyte population over time. Significant increases in apoptotic levels were induced throughout a 72 hour period in PMA-treated cells only. Lymphocytes were also separated by size for analysis. Several sub-populations of lymphocytes were identified, the most interesting of which was small and apoptotic within 4 hours, after PMA exposure. PMA-induced DNA synthesis did not become elevated until after 24 hours. “Direct” apoptosis, i.e. without cell cycle entry, was found only in these small, mature lymphocytes. Since small lymphocytes make up the vast majority of those being analyzed, “direct” apoptosis may be a determining mechanism in the resistance to neoplasia observed in Amphibia. Cells that die more readily are less likely to transform into neoplastic cells.
