    Description / Table of Contents: Résumé La concentration plasmatique d'insuline immuno-réactive, d'acides gras non estérifiés et de glucose a été déterminée chez des sujets normaux et dans un groupe d'obèses non diabétiques avant et après une période de restriction alimentaire. La concentration d'insuline immuno-réactive à jeun et après administration orale de glucose était anormalement élevée dans le groupe des sujets obèses et montra une diminution significative après une période de restriction alimentaire ayant entraîné une perte de poids.
    Abstract: Zusammenfassung Es wurden Messungen der Plasma spiegel von immunreaktivem Insulin, unveresterten Fett-säuren und Glucose bei normalen Personen und bei einer Gruppe von fettleibigen Nichtdiabetikern vor sowie nach einer Periode der Diätbeschränkung vorgenommen. Die Konzentration des immunreaktiven Insulins lag bei den fettleibigen Patienten sowohl im Nüchternzustand als auch nach einer oralen Glucosebelastung abnorm hoch und ging nach Diätbeschränkung und Gewichtsverlust wesentlich zurück.
    Notes: Summary Measurements of plasma levels of immunoreactive insulin, non-esterified fatty acids and glucose have been made in normal subjects and a group of obese non diabetic patients, before and after a period of dietary restriction. Levels of immuno-reactive insulin, both in the fasted state and after an oral glucose load, were abnormally high in the group of obese patients and were significantly lower after dietary restriction and loss of body weight.

    Description / Table of Contents: Résumé Après l'injection intraveineuse de glucose (750 mg/kg) en même temps qu'une dose traceuse de U-14C-glucose, à des rats blancs adultes à jeun, les pourcentages suivants de la dose administrée ont été retrouvés dans tous les organes et tissus: 1. après cinq minutes: muscle squelettique 30.3, peau 28.1, sang 13.1, tissu adipeux 10.7, foie 8.9. 2. Quarante minutes après l'injection, les valeurs correspondantes étaient les suivantes: 35.0, 11.1, 5.0, 4.6 et 9.4%. Le 14CO2 dégagé était négligeable après 5 min: après 40 min il représentait 8% de la dose totale administrée.— Après l'administration intragastrique de 1500 mg/kg de glucose, donné en même temps qu'une dose traceuse de U-14C-glucose dans les mêmes conditions expérimentales, le tube digestif contenait au bout de 15, 90 et 180 min, 60.5, 14.8 et 8.4% respectivement de la dose totale de 14C administrée. A ces moments là le foie contenait respectivement 2.9, 10.7 et 15.0%; la peau contenait 7.5, 7.1 et 5.4%; le tissu adipeux 2.0, 3.8 et 3.5%, et le 14CO2 dégagé était de 0.4, 11.8 et 31.3%. On donne des détails sur la captation du 14C-glucose par d'autres organes et tissus, et on essaye de dresser un bilan du produit injecté.
    Abstract: Zusammenfassung Nach i.v. Injektion von 750 mg Glucose/kg mit einer Sprüdosis U-14C-Glucose fanden sich bei fastenden, erwachsenen weißen Ratten folgende Prozentsätze der verabreichten Menge in Gesamt-Organen und -Geweben: 1. Nach 5 min: Skeletmuskel 30.3, Haut 28.1, Blut 13.1, Fettgewebe 10.7, Leber 8.9. 2. 40 min nach der Injektion lauteten die entsprechenden Werte: 35.0, 11.1, 5.0, 4.6, 9.4%. Nach 5 min ließen sich in der Ausatmungsluft nur Spuren von 14CO2 nachweisen, nach 40 min lagen 8% der verabreichten Dosis in dieser Form vor. — Nach intragastrischer Zufuhr von 1500 mg Glucose/kg mit einer Spürdosis von U-14C-Glucose unter den gleichen Versuchsbedingungen enthielt der Verdauungstrakt nach 15, 90 und 180 min jeweils 60.5, 14.8 bzw. 8.4% der zugeführten Radioaktivität. In der Leber fanden sich zu diesen Zeiten 2.9, 10.7 und 15.0%, in der Haut 7.5, 7.1 und 5.4, im Fettgewebe 2.0, 3.8 und 3.5%. Die Ausatmungsluft enthielt 0.4, 11.8 und 31.3% als 14CO2. Es folgen weitere Einzelheiten zur Aufnahme von Radioglucose durch andere Organe und Gewebe sowie der Versuch einer Bilanz für das injizierte Material.
    Notes: Summary After intravenous injection of glucose, 750 mg/kg, together with a tracer dose of [U-14C] glucose, into fasted, adult white rats, the following percentages of the administered dose were found in whole organs and tissues: 1. after five minutes: skeletal muscle 30.3, skin 28.1, blood 13.1, adipose tissue 10.7, liver 8.9. 2. Forty minutes after injection the corresponding values were: 35.0, 11.1, 5.0, 4.6 and 9.4%. Expired 14CO2 was negligible after five minutes: after 40 min it comprised 8% of the total dose administered. — After intragastric administration of 1500 mg/kg of glucose given with a tracer dose of [U-14C] glucose under the same experimental conditions, the alimentary tract contained, after 15, 90 and 180 min, 60.5, 14.8 and 8.4% respectively of the total 14C dose given. At these times the liver contained 2.9, 10.7 and 15.0%; skin contained 7.5, 7.1 and 5.4%; adipose tissue 2.0, 3.8 and 3.5%, and expired 14CO2 0.4, 11.8 and 31.3% respectively. Details of the uptake of 14C glucose by other organs and tissues are given, and a balance sheet for the injected material is attempted.

    Notes: The acarofauna and Der p I allergen concentrations in dust samples from mattresses and lounge room carpets obtained from 20 homes from two coastal cities, Perth and Bunbury, were determined. All samples were shown to contain mites and the geometric mean numbers of total mites/g of mattress and carpet dust for Perth and Bunbury were 480 and 263, and 585 and 992, respectively. Carpets from both centres had a significantly (P 〈 0.02) greater mean number of mite species (Perth 9.1, Bunbury 9.0) than mattresses (Perth 5.2, Bunbury 5.7). The predominant mite species were D. pteronyssinus, E. maynei and Tarsonemus spp. D.farinae was found to be absent from all dust samples examined. E. maynei was present in the 10 Bunbury homes and in 50% of the Perth homes, ranging from 0 to 81% of mites identified. The arithmetic mean Der p I concentrations in the mattresses and carpets in Perth and Bunbury were 4.2 and 4.1, and 3.8 and 9.2 μg/per gram of fine dust, respectively, and Der p I concentration correlated with mite counts (r = 0.75; P 〈 0.001). The concentration of Der p I equivalent per 100 mites was 1.5 μg. The data are consistent with the view that asthmatic patients in Western Australia have significant exposure to a variety of house dust mites and that E. maynei may be clinically significant.

    Notes: The presence of a high molecular weight glycoprotein component bearing the hapten phosphorylcholine (Pc) has been demonstrated in some but not all extracts of house dust mite using a phosphorylcholine specific IgA myeloma protein (S107). The Pc bearing component was isolated from house dust mite extracts by precipitation at equivalence using whole myeloma serum and the isolated fraction was found to be allergenic as judged by a direct RAST assay using a highly house dust mite allergic human serum pool. RAST inhibition studies using free hapten and direct RAST studies using pneumococcal polysaccharide C-substance indicated that the Pe moiety did not contribute significantly to the immunodominant portion of the isolated allergen. Further studies indicated that the S107 isolated material was immunochemically different from the tridacnin reactive material and the con A reactive material previously isolated from extracts of house dust mite.

    Notes: Five allergen extracts of Dermatophagoides pteronyssinus, Lolium perenne, Alternaria tenuis, Aspergillus fumigatus and Cladosporium herbarum, obtained from four different manufacturers, were examined by inhibition of RAST, content of protein and carbohydrate, contents of phosphorylcholine (Pc) and tridacnin reactive components, and by skin test. Inhibition of RAST was used as a primary method for establishing allergenic potency and demonstrated wide variations for each preparation supplied by the different manufacturers. The extracts also varied widely in protein and carbohydrate content and in the ratio of these parameters, indicating internal heterogeneity. Pc content was significantly related to RAST potency for extracts of A. fumigatus and A. tenuis, suggesting that Pc content may be used as a primary standarization procedure for these extracts. Skin test reactions undertaken at a single concentration did not show any significant variation in weal size between preparations of a given allergen extract. However, of particular importance to practising clinicians is the finding that varying numbers of patients showed negative skin reactions to one preparation of a particular allergen yet were positive to the corresponding preparations supplied by the other companies.

    Notes: There have been only a few studies of how allergens cross the airway epithelium to cause allergic sensitization. House dust mite fecal pellets (HDMFP) contain several proteolytic enzymes. Group 1 allergens are cysteine peptidases, whilst those of groups 3, 6 and 9 have catalytic sites indicative of enzymes that mechanistically behave as serine peptidases. We have previously shown that the group 1 allergen Der p 1 leads to cleavage of tight junctions (TJs), allowing allergen delivery to antigen presenting cells.In this study we determined whether HDMFP serine peptidases similarly compromise the airway epithelium by attacking TJs, desmosomes and adherens junctions.Experiments were performed in monolayers of MDCK, Calu-3 or 16HBE14o-epithelial cells. Cell junction morphology was examined by 2-photon molecular excitation microscopy and digital image analysis. Barrier function was measured as mannitol permeability. Cleavage of cell adhesion proteins was studied by immunoblotting and mass spectrometry.HDMFP serine peptidases led to a progressive cleavage of TJs and increased epithelial permeability. Desmosomal puncta became more concentrated. Cleavage of TJs involved proteolysis of the TJ proteins, occludin and ZO-1. This was associated with activation of intracellular proteolysis of ZO-1. In contrast to occludin, E-cadherin of adherens junctions was cleaved less extensively. Although Calu-3 and 16HBE14o-cells expressed tethered ligand receptors for serine peptidases, these were not responsible for transducing the changes in TJs.HDMFP serine peptidases cause cleavage of TJs. This study identifies a second general class of HDM peptidase capable of increasing epithelial permeability and thereby creating conditions that would favour transepithelial delivery of allergens.
