    Notizen: Conjugates based on N-(2-hydroxypropyl)methacrylamide (HPMA) represent a new generation of antibody-targeted polymeric anticancer drugs with both cytotoxic and immunoprotecting/immunomobilizing activity. 20–90% of mice that are cured of EL4 mouse T-cell lymphoma, BCL1 mouse B-cell leukaemia and 38C13 mouse B-cell lymphoma by injection of doxorubicin-HPMA conjugate develop a long-lasting memory and systemic antitumour resistance. It is suggested that the main activity of the polymeric drug, directly after application is – due to the high level of the drug – of cytotoxic and cytostatic nature. Thereafter, long-term conjugates persist at low concentration in the circulation, which are capable of mobilizing the defence mechanisms of the host. Until now, seven patients with generalized carcinoma were treated with doxorubicin-HPMA–human-Ig conjugate. Disease stabilization, lasting from 6 to more than 18 months, was recorded.

    Notizen: Relationships were established for five sensory methods of oral and nonoral viscosity evaluation between viscosity scores and instrumentally measured dynamic viscosity for model and real Newtonian fluid foods. These relationships were then used to predict the effective shear rates under which the sensory tests were performed. The highest shear rates were predicted for viscosity perception by compression of samples between tongue and palate, and the lowest for pouring the fluid foods from a teaspoon. Mixing with a teaspoon, slurping and swallowing exhibited nearly the same dependencies of apparent shear rates on equivalent instrumental viscosity. All relations were of the hyperbolic type. The resulting relationships between the apprent shear rates and equivalent instrumental viscosity are in good agreement with a similar relationship predicted by Shama and Sherman (1973a) (see Cutler et al. 1983) for oral perception.

    Notizen: A copolymer of N-(2-hydroxypropyl) methacrylamide (HPMA) and N-methacryloyltyrosinamide was prepared and fractionated using Sepharose 4B/6B (1:1) chromatography to produce eight HPMA copolymer fractions of narrow polydispersity and mean molecular weight (Mw) ranging from 12 to 778 kD. These fractions were radioiodinated and injected intravenously, subcutaneously, and intraperitoneally into rats. Their bloodstream-concentration profiles were monitored and rates of excretion assessed. Following intravenous administration the circulating blood volume available to the copolymers was not molecular-weight-dependent. A molecular-weight threshold limiting glomerular filtration was identified at approximately 45 kD, and preparations greater than this threshold were lost from the bloodstream only slowly by extravasation. Molecular weight did not influence the movement of copolymers from the peritoneal compartment to the bloodstream after intraperitoneal injection. The transfer rates observed could be accounted for by bulk phase lymphatic drainage alone, no transcapillary routes being implicated. Following subcutaneous administration the largest HPMA copolymer fraction (Mw = 778 kD, diameter approximately 30 nm) showed increased retention at the site of injection, approximately 20% of the dose remaining there after 21 days. This could result from physical restriction of movement or from internalization into local phagocytic cells. The smaller copolymer fractions moved readily into the bloodstream whence they were either lost in the urine or they gradually penetrated into other tissues and organs. Long-term (21 days) body distribution of copolymers following both intraperitoneal and subcutaneous administration showed size-dependent accumulation in organs of the reticuloendothelial system.

Die Zugabe einer kleinen Menge von Losungsmittel (Methanol) zur Polymerisationsmischung verursachte hohere Polymerisationsgeschwindigkeit und hoheres Molekulargewicht, aber größere Mengen von Losungsmittel erniedrigten beide Grokn. Das Auftreten eines Maximums bei R, und R, in Abhangigkeit von der Monomerkonzentration wurde nach der klassischen Bamford-Jenkins-Theorie diskutiert.
    Notizen: Kinetics of radical heterogeneous polymerization of N-(2-hydroxypropyl)methacrylamide was studied in acetone at 55°C. The values of reaction order with respect to the monomer (1.6) and to the initiator (0.5) indicate a bimolecular termination and degradative transfer during the chain growth. The over-all rate constant of polymerization was found to be 5.6.10-4. Addition of a small amount of a solvent (methanol) to the polymerization mixture led to an increased polymerization rate and increased molecular weight of the resulting polymer, but a higher amount of the solvent brought about a decrease of both characteristics. The Occurence of maxima on the dependences of Rp, as well as M̄w on the monomer concentration was interpreted in terms of the classical Bamford and Jenkins theory.

Die Copolymerisation von N-Äthylacrylamid mit N-Butylmethacrylamid wurde auch in Dimethylformamid und in Methanol durchgeführt. Die Resultate zeigen, daß die am Amidstickstoff gebundene Alkylgruppe die Reaktivität der untersuchten Verbindungen praktisch nicht beeinflußt. Das Lösungsmittel übt einen etwas stärkeren Einfluß auf die Copolymerisationsparameter aus. Wegen der ähnlichen Struktur der beiden Comonomeren werden die Parameter nur als Folge der verschiedenen Abhängigkeit der Reaktivität des N-Äthylacrylamids und N-Butylmethacrylamids vom Lösungsmittel beeinflußt.
    Notizen: The radical copolymerization of 14C-labelled N-ethylacrylamide with N-ethylmethacrylamide, N-(2-hydroxypropyl)methacrylamide and N-butylmethacrylamide in dimethylsulphoxide at 60°C was investigated. The copolymerization of N-ethylacrylamide with N-butylmethacrylamide was also studied in dimethylformamide and methanol. The results show that the structure of the alkyl group bonded to the amide nitrogen does not affect the reactivity of the compounds under investigation to any considerable extent. The solvent exhibits a somewhat stronger influence upon the copolymerization parameters. Because of the similar structure of both comonomers the copolymerization parameters will be affected by the solvent only as a result of the different dependence of the reactivity of N-ethylacrylamide and N-butylmethacrylamide on the character of the medium.
