    Notes: Abstract Cloning and sequencing of an approximately 6.0-kb chromosomal DNA fragment from Helicobacter felis revealed five complete open reading frames. The deduced amino acid sequence of one ORF exhibited sequence similarity to the FtsH protein, an ATP-dependent metalloprotease, from various bacterial species. The encoded protein consists of 638 amino acid residues with a molecular mass of 70.2 kDa. The hydropathy profile of the FtsH protein predicted two N-terminal transmembrane regions that were confirmed experimentally. Insertion of ftsH into a new versatile expression vector resulted in overexpression of FtsH protein in Escherichia coli. In addition, the E. coli ftsH gene could be replaced by the H. felis homologue to allow reduced growth and tenfold increased lysogenization by temperate phage λ.

    Notes: Inactivation of Helicobacter pylori cadA, encoding a putative transition metal ATPase, was only possible in one of four natural competent H. pylori strains, designated 69A. All tested cadA mutants showed increased growth sensitivity to Cd(II) and Zn(II). In addition, some of them showed both reduced 63Ni accumulation during growth and no or impaired urease activity, which was not due to lack of urease enzyme subunits. Gene complementation experiments with plasmid (pY178)-derived H. pylori cadA failed to correct the deficiencies, whereas resistance to Cd(II) and Zn(II) was restored. Moreover, pY178 conferred increased Co(II) resistance to both the cadA mutants and the wild-type strain 69A. Heterologous expression of H. pylori cadA in an Escherichia coli zntA mutant resulted in an elevated resistance to Cd(II) and Zn(II). Expression of cadA in E. coli SE5000 harbouring H. pylori nixA, which encodes a divalent cation importer along with the H. pylori urease gene cluster, led to about a threefold increase in urease activity compared with E. coli control cells lacking the H. pylori cadA gene. These results suggest that H. pylori CadA is an essential resistance pump with ion specificity towards Cd(II), Zn(II) and Co(II). They also point to a possible role of H. pylori CadA in high-level activity of H. pylori urease, an enzyme sensitive to a variety of metal ions.

    Notes: Abstract Precursor mRNA is complexed with proteins in the cell nucleus to form heterogeneous nuclear ribonucleoprotein (hnRNP), and these hnRNPs are found associated in vivo with small nuclear RNPs (snRNPs) for the processing of pre-mRNA. In order to better characterize the ATP-independent initial association of U1 snRNP with hnRNP, an important early event in assembly of the spliceosome complex, we have determined some of the components essential to an in vitro reassociation of U1 snRNP with hnRNP. U1 snRNP reassociated in vitro with 40S hnRNP particles from HeLa cells and, similar to the in vivo hnRNP/U1 snRNP association, the in vitro interaction was sensitive to high salt concentrations. U1 snRNP also associated with in vitro reconstituted hnRNP in which bacteriophage MS2 RNA, which lacks introns, was used as the RNA component. Purified snRNA alone would not associate with the MS2 RNA-reconstituted hnRNP, however, intact U1 snRNP did interact with protein-free MS2 RNA. This indicates that the U1 snRNP proteins are required for the hnRNP/U1 snRNP association, but hnRNP proteins are not. Thus, the initial, ATP-independent association of U1 snRNP with hnRNP seems to be mediated by U1 snRNP protein(s) associating with hnRNA without requiring a splice-site sequence. This complex may then be further stabilized by intron-specific interactions and hnRNP proteins, as well as by other snRNPs.

    Notes: Abstract We report the results of a search for evidence of periodic unstableorbits in the electroreceptors of the catfish. The function of thesereceptor organs is to sense weak external electric fields. Inaddition, they respond to the ambient temperature and to the ioniccomposition of the water. These quantities are encoded by receptorsthat make use of an internal oscillator operating at the level of themembrane potential. If such oscillators have three or more degreesof freedom, and at least one of which also exhibits a nonlinearity,they are potentially capable of chaotic dynamics. By detecting theexistence of stable and unstable periodic orbits, we demonstratebifurcations between noisy stable and chaotic behavior using theambient temperature as a parameter. We suggest that the techniquedeveloped herein be regarded as an additional tool for the analysisof data in sensory biology and thus can be potentially useful instudies of functional responses to external stimuli. We speculatethat the appearance of unstable orbits may be indicative of a stateof heightened sensory awareness by the animal.

    Notes: Abstract We report the results of a search for evidence of unstable periodic orbits in the sensory afferents of the facial cold receptors of the rat. Cold receptors are unique in that they exhibit a diversity of action potential firing patterns as well as pronounced transients in firing rate following rapid temperature changes. These characteristics are the result of an internal oscillator operating at the level of the membrane potential. If such oscillators have three or more degree of freedom, and at least one of which also exhibits a nonlinearity, they are potentially capable of complex activity. By detecting the existence of unstable periodic orbits, we demonstrate low-dimensional dynamical behavior whose characteristics depend on the temperature range, impulse pattern, and temperature transients.

    Notes: Zusammenfassung Mit Hilfe eines einfachen Flüssigkeitsmodells, nämlich des Löchermodells, wurde die Selbstdiffusion in einatomigen Flüssigkeiten berechnet, wobei als Berechnungsgrundlage die Beziehung zwischen Diffusion und dem mittleren Quadrat der Verschiebung der Teilchen diente. Durch Heranziehung der Stokes'schen Formel für den Reibungswiderstand, die durch eine geringfügige Änderung den atomaren Verhältnissen besser angepaßt wurde (Faktor 2), konnten die Ergebnisse leicht auf die Viskosität einatomiger Flüssigkeiten übertragen werden. Es gelang so eine Absolutberechnung der Werte vonD undη, die an Hand der Erfahrung geprüft werden konnte und bei den einatomigen Metallen eine befriedigende Übereinstimmung lieferte. Eine direkte Übertragung der experimentell genauer nachprüfbaren Endformel für die Viskosität auf mehratomige Molekeln von kugelförmigem Bau liefert wegen des Versagens der einfachen Stokes'schen Formel im allgemeinen zu hohe Viskositätswerte.

    Notes: Abstract The response characteristics of facial thermoreceptors of the common vampire bat and of the mouse have been quantitatively studied. Cold receptors were identified in bat and mouse; warm receptors were only established in the bat. Cold and warm receptor populations of the two species share most of their properties with facial thermoreceptor populations of various mammalian species investigated so far. The temporal pattern of activity of cold receptors of the mouse corresponded to that observed in cats, dogs and monkeys: impulse groups at lower, and beating activity at higher temperatures. At maintained temperature, no impulse groups were initiated in cold receptors of the bat. However, cooling steps from various initial temperatures induced a transient grouped discharge in both cold receptor populations. A discharge in regular groups of impulses was occasionally generated in warm receptors of the bat at maintained temperatures and following warming steps. The data indicate that the temperature dependence of periodic activity in warm receptors is not as uniform as it is in cold receptors. It is concluded that cyclic processes are involved in sensory transduction of both warm and cold receptors, and that this cyclic behavior seems to be a general property of thermoreceptors of presumably all vertebrate species.

    Notes: Abstract An isolated preparation of the cat tongue was developed to study quantitatively the relation between single cold receptor activity and calcium concentration. The activity recorded from lingual cold fibres of the isolated preparation was identical to that observed by previous authors in lingual cold receptors of whole animals. A perfusing solution containing excess calcium (5 mmol/l) suppressed discharge at constant temperatures as well as the dynamic response to rapid cooling. Reduced calcium levels (0.5 mmol/l) enhanced cold fibre activity at adapting temperatures above 15°C. Cold receptor activity was linearly inversely related to the logarithm of calcium concentration at adapting temperatures above 20°C. The linear relation was valid for both the discharge at constant temperatures and the dynamic peak frequency during cooling steps. From this relation it was calculated that at 25° C an elevation of 0.1 mmol/l external calcium above the control value (1.53 mmol/l) reduces static firing rate by 0.35 impulses per second. It is concluded that neither the in vitro conditions nor the application of various external calcium concentrations impair cold receptor function. The data are consistent with the hypothesis of a calcium-stimulated outward current controlling the static and dynamic response characteristics of cold receptors.
