Summary Report of a new test, controlling the antilymphocytic in vitro-effect of the antilymphocytic serum (ALS) of Pichlmayr, Brendel and coworkers. The test is practised with fresh samples of blood, mixed with an anticoagulant (Vetren). The test consists of two phases: a) incubation of the blood with different concentrated ALS (seven solutions, 1:3600 till 1:12 ALS, control sample with NaCl solution); b) Examination of the vitality of lymphocytes by means of testing their resistance against osmotic damage in hypotonic environment. Valuation by establishing a „blood picture of haemolysis“, using blood smears on slides, coloured as usual. The result (percentage of intact immunocompetent lymphocytes) is demonstrated in a curve. The lymphocytic effect of ALS is pointed out by a concentration adequate 50 percent of intact immunocompetent lymphocytes: “toxic concentration 50” (CT 50). The advantages of the test are: use of whole blood; separated effect on lymphocytes and polynucleated white cells; perception of little differences; simple technic.

Zusammenfassung Bericht über einen neuen Test zur in vitro-Kontrolle der antilymphocytären Wirkung des antilymphocytären Immunserums (ALS) von Pichlmayr, Brendel u. Mitarb. Der Test wird an frisch entnommenem Vetren-Vollblut durchgeführt; er verläuft in zwei Phasen: a) Inkubation des Blutes mit ALS in unterschiedlicher Konzentration (7 Verdünnungsstufen, 1:3600 bis 1:12 des ALS bei der Inkubation; Kontrolle mit physiologischer NaCl-Lösung); b) Durchführung einer Resistenzbestimmung mit Prüfung der osmotischen Resistenz (hypotones Milieu). Auswertung durch Herstellung eines Leukocytolyse-Blutbildes an Objektträger-Ausstrichen. Das Ergebnis — Prozentzahl intakter immunokompetenter Lymphocyten — wird in einer Kurve in bezug auf die Konzentration des ALS bei der Inkubation dargestellt (Konzentrationswirkungskurve, logarithmischer Maßstab). Die lymphotoxische Wirkung eines ALS wird gekennzeichnet durch diejenige Konzentration des ALS, bei der noch 50% der immunokompetenten Lymphocyten erhalten sind: „Toxische Konzentration 50“, CT 50. Der Test hat folgende Vorteile: Verwendung von Vollblut; getrennte Anzeige der antilymphocytären und der antigranulocytären Wirkung; bessere Differenzierung als beim cytotoxischen Test nach Pappenheimer; einfache Technik.
