    Notes: In mushroom, presence of the mannitol cycle has not been reported so far although the polyol is supposed to be generated by the reduction of fructose by mannitol dehydrogenase. This study submits evidence for the presence of the mannitol cycle in Pleurotus ostreatus. The key enzyme of the cycle, mannitol-1-phosphate dehydrogenase (M1PDH), was present appreciably in all the developmental stages of the mushroom. However, the enzyme level dropped significantly at the onset of sporulation. The presence of M1DPH was confirmed by isozyme analysis and RT-PCR mediated amplification of a ∼400 bp DNA fragment.

    Notes: Mycelial derived protoplasts of the mushroom, Termitomyces clypeatus, did not regenerate in synthetic growth medium but did regenerate if casein hydrolysate was added to the same medium. Aspartic acid, glutamic acid, histidine, of serine and alanine when added individually had a similar effect on regeneration to that of casein hydrolysate. Krebs cycle acids also supported protoplast regeneration in the presence of glucose. Glucose alone, in the regenerating medium supported osmotically stable germ tube formation from protoplasts while Krebs cycle acid alone could not even change the osmotic fragility of the protoplasts. Electron microscopy of germ tube preparations indicated the presence of an incomplete wall structure when regeneration took place in the presence of glucose alone.

    Notes: Summary An amyloglucosidase from a mycelial culture of the mushroom Termitomyces clypeatus hydrolysed larch wood xylan independently and synergistically with an endo-β(1→4) xylanase of the same fungus. The glucoamylase saccharified xylan predigested with xylanase at a faster rate compared to that of xylanase acting on amylase-digested xylan. However, overall saccharification of xylan in both cases was the same. Only glucose was liberated from xylan by amylase digestion whereas xylose, xylobiose and other oligosaccharides were liberated during xylanase digestion. The synergistic response of enzyme combinations was reflected in the liberation of glucose from xylan, rather than xylose. Glucoamylase and xylanase activities on soluble and insoluble fractions of larch wood xylan with different xylose and glucose contents suggested that synergism in xylanolysis by the presence of glucoamylase was dependent on the activity of the participating xylanase on the xylan preparation. It is suggested that possibly α-glucosidic linkages are present in xylan and that amyloglucosidase might be involved in xylanolysis.

    Notes: Abstract During sporulation, laccase activity of Pleurotus florida decreased to a minimum level in spite of increase in the number of isozymes. An endogenous laccase substrate was detected especially in the gill structure of the sporophore, which competitively inhibited oxidation of guaiacol by the enzyme during in vitro assay. Appearance of the laccase substrate in the gill structure may be linked with the sporulation phenomenon.
