    Notizen: The glycine decarboxylase multienzyme complex (EC 2.1.2.10) is located in the mitochondrial matrix and catalyzes a key reaction of the photorespiratory C-2 cycle of plants. The light-dependent control of the synthesis of the P-protein of this complex in pea (Pisum sativum cv. Alaska) leaves was studied using a partial (0.7 kb) cDNA clone. The size of the P-protein mRNA detected by this probe was 3.5 kb. When the mRNA hybrid selected by this probe was translated in vitro, a protein that migrated in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) with an apparent molecular mass of 114 kDa was formed. This protein could be immunoprecipitated with the P-protein-specific antibody. The mass of the mature P-protein on SDS-PAGE is 100 kDa. This suggests the presence of a presequence used to direct the protein to the mitochondrial matrix. Analysis of the concentration of P-protein mRNA showed an approximately 5-fold increase in the abundance of this transcript in light-grown as compared to dark-grown pea seedlings. The time course for the production of P-protein mRNA following exposure of dark-grown peas to white light was compared to that for the H-protein of glycine decarboxylase, the small subunit of ribulose bisphosphate carboxylase/oxygenase, and the chlorophyll a/b binding protein. All 4 mRNAs accumulated with similar kinetics where they remained low during the first 3 h of illumination, increased about 5-fold during the next 8 h, and then maintained a steady state over the next 12 h.

    Notizen: Processing of polymers plays an important role in application of polymers in biomedicalengineering, for instance in manufacture of scaffolds for tissue engineering applications. Rapidprototyping technologies like fused deposition modeling (FDM) has been widely used in processingpolymers for biomedical applications. The present work is focused on modeling of flow behavior inthe extrusion liquefier in FDM. A finite element (FE) model of extrusion liquefier was constructedon ANSYS after verification of internal geometry using X-ray imaging. Polycaprolactone (PCL) isused as the base bio polymer for analysis. Experiments were carried out to characterize the physicalproperties like thermal conductivity, specific heat, viscosity and shear thinning property of PCL.These values were used for behavior modeling in the extrusion liquefier. The thermal and flowbehavior in the extrusion liquefier is studied by varying input conditions and analyzing the velocity,pressure drop profiles at various zones of extrusion liquefier. Experimental values of parametersand the simulated flow model showed good correlation. The current model can be extended topredict the flow behavior of PCL/ Hydroxyapatite composites in a FDM head which in turn willreflect on the quality of scaffold constructed using the Biocomposite

    Notizen: Abstract Phage M-1 produced clear plaques with a halo in the lawn of Rhizobium japonicum D211. A one step growth curve of phage M-1 showed a latent period of 3 h, burst size of 55 and rise period of 2 h. The inactivation of phage M-1 was found to be dependent upon the concentraion of d-glucosomanine. The neutralization kinetics of phage M-1 by antiphage serum gave a K value (velocity constant) of 83.1 min−1. Transduction of str and kan was studied in the presence of antiphage serum and d-glucosamine. Cotransduction of different antibiotic resistance markers suggested that the system can be further explored for high resolution mapping in R. japonicum.

    Notizen: Abstract The formation of R-prime plasmids was selected in crosses involving soybean microsymbionts with genomic Tn5 insertions and carrying plasmid pJB3JI (with one IS2) copy as donors and Escherichia coli HB101 as recipient. Whereas the parent plasmid was 60 kb, recombinant plasmids between 76 kb and 121 kb were obtained. Restriction and Southern analyses confirmed the mobilization of Tn5 on four R-primes from Bradyrhizobium japonicum I-110 and on an R-prime plasmid from Rhizobium fredii HH303. The largest R-prime plasmid was obtained from the rescue of two symbiotically defective R. fredii mutant strains that required adenosine.

    Notizen: Abstract In order to study the transcriptional control of 15-LO expression, we have cloned and sequenced the human 15-LO promoter region. The 15-LO promoter is associated with a CpG island at the 5′-end of the gene, and sequence analysis reveals putative Sp1 and Ap2 binding site/s and absence of TATA or CAAT motifs. Transcription is initiated at one major site. Using deletion constructs, we have defined an active promoter region of 1056 bp. Gel-shift assays revealed that transcriptional factor(s) induced only in response to IL-13 treatment of human peripheral blood monocytes bind to the 15-LO promoter DNA. Two regions, DP1 (−140 to −92 bp) and DP2 (−353 to −304 bp) of the promoter were essential for transcription in HeLa cells and human peripheral monocytes. Hela nuclear extracts contained a specific nuclear factor(s) binding to 15-LO promoter DNA which are distinct from those derived from IL-13-treated human peripheral monocyte nuclear extracts. In addition, fluorescent in situ hybridization (FISH) results refined the previous localization of 15-LO to human chromosome 17p13.3.

    Notizen: We investigated the miscibility of polybutadienes with butadiene-styrene copolymers of varying styrene content. The methods of optical microscopy, differential scanning calorimetry, and small angle light scattering (SALS) were used. All polymers/copolymers used in this study have equivalent butadiene microstructures. Copolymers with high styrene levels are immiscible with polybutadiene. The tendency to miscibility increases with decreasing styrene content. An upper critical solution temperature is observed with the 23 percent styrene copolymer.

    Notizen: The rigid-ice model of frost heave is one of the most comprehensive frost-heave models but is restricted to one-dimensional cases in its present form. In this paper, the model is extended to two-dimensional problems. The complete formulation of the partial differential equations governing heat, moisture and ice transport in freezing soils is provided. The equations are subsequently solved using the Galerkin finite element method in space and the finite difference method in time. A computer program is developed for the two-dimensional rigid-ice model. A case of freezing around chilled gas pipeline is solved and the numerical results are compared with experimental values, with good agreement between the two sets of results.

    Notizen: The miscibility of various amorphous polybutadienes with mixed microstructures of 1,4 addition units (cis, 1,4 and trans 1,4) and 1,2 addition units have been investigated. The studies here involved optical transparency, differential scanning calorimetry, and small angle light scattering. It was found that a 90 percent (cis) 1, 4 addition polybutadiene was immiscible with high (91 percent) 1,2 addition polybutadiene. Reduction of the 1,2 content to 71 percent induced an upper critical solution temperature (UCST) with the cis 1,4 polymer. Polybutadienes with 50 percent and 10 percent 1,2 contents were miscible above the crystalline melting temperature of the cis 1,4 polybutadiene. Immiscibility of the 91 percent 1,2 addition polymer was also found with a 10 percent 1,2 polybutadiene. The latter polymer also exhibits an UCST with the 71 percent 1,2 polymer. The results are used to interpret the characteristics of blends of polybutadienes of varying microstructure.
