    Notes: Summary Insulin resistance is a critical component underlying the altered glucose homeostasis in a variety of metabolic and non-metabolic disorders. Aging, body fat distribution, obesity, diabetes mellitus or hypertension are well recognized conditions associated with an impaired tissue sensitivity to insulin action. Apart from such constant factors, insulin sensitivity can be acutely modified by independent variables such as physical exercise, dietary factors, alcohol intake or harmless drugs. To evaluate the day-to-day intra-individual variation in insulin sensitivity, glucose homeostasis and lipid profiles, we investigated the insulin sensitivity index (SI) (determined by the minimal model method of Bergman), basal and post-glucose-load insulin and glucose levels, serum total triglyceride and lipoprotein cholesterol fractions in 15 healthy young men (24± 1 year, mean± SEM), on two different occasions at an interval of 3 weeks (days 1 and 21), after 3 days of a standard dietary regimen and after an overnight fast. Blood pressure, heart rate, body weight and 24 h urinary sodium excretion were almost identical in the two phases. SI(day 1) varied from 4.2 to 15.8·10−4·min−1 pro μU/ml (mean: 10.2±0.9) and correlated with SI(day 21) (11.2±1.2· 10−4·min−1 pro μU/ml r = 0.78,p〈0.0007). The slope of the relationship did not differ from 1 (1.01, p〉0.90), the intercept was close to the origin (0.8,p〉0.73) and the coefficient of variation was 14.4%. Other variables of carbohydrate and lipid metabolism that were strongly correlated to each other on the two different days were: total serum lipids (p〈0.0009 to 0.0002) and the fasting and peak plasma insulin (p〈0.001 to 0.0001) and glucose levels (p〈0.005 to 0.001). These findings indicate that the assessment of in vivo insulin sensitivity using the minimal model method is practical and can be reproduced with accetable intra-individual variation in young healthy subjects.

    Notes: Summary Pregnant rats received a single administration of 0.5, 1.0, or 2.0 mg/kg of cadmium on day 18 or 20 or gestation Maternal animals were killed on day 21 and samples of the caudate nucleus from fetal brains were examined using the transmission electron microscope. A 2.0 mg/kg dose of cadmium administered to maternal rats on day 20 of gestation caused the formation of vacuoles in the endothelial cells of capilaries in the fetal brain. Significant endothelial cell vacuolization was not observed in the brain of fetuses from other treatment groups. The vacuoles occurred singly, were spherical in shape, were located adjacent to the intercellular junctions and caused focal distortion of the endothelial cell. Vacuoles were the only ultrastructural alteration observed in the caudate nucleus of fetal brains.

    Notes: Abstract  Oxidation of a flotation-derived, low-sulfide tailings containing approximately 0.4 wt.% S was compared with simultaneously oxidized tailings containing 1.0 wt.% S and 2.5 wt.% S to assess their acid generating characteristics. Each tailings type was exposed to oxidation for three years in laboratory columns and in lysimeter pits in the field. In these tailings the sulfide mineral of principal concern with respect to acid generation is pyrrhotite (Fe 1-x S). In past studies the alteration of pyrrhotite has been characterized by initial replacement with marcasite (FeS2) and ferric iron sulfates, which are followed by development of ferric oxyhydroxides such as goethite and lepidocrocite. Macroscopic characterization of the tailings shows varying and progressive degrees of oxidation correlative with the three different sulfur contents. As expected, the tailings with the lowest sulfur content are the least oxidized, and those with the highest sulfur content have reacted the most. The column tests, which represent accelerated reaction conditions relative to those for the lysimeter pits, show much higher degrees of oxidation, and a markedly more distinct boundary between the oxidized and unoxidized zones; as well, differences among the three tailings types are more pronounced.

    Notes: Abstract The HLA-linked “SB” antigens comprise a new segregant series of B-cell alloantigens mapping between HLA-DR and glyoxylase. They can be detected by secondary proliferative responses of lymphocytes primed against HLA-A, B, C, DR, MB- and MT-compatible stimulators. To assess genetic complexity of the SB-gene region, alloreactive cloned T-cell lines were derived from four reagents detecting specificities designated SB2 and SB3. In two families, products detected by seven different clones segregated with the HLA haplotypes bearing the SB2 or SB3 specificities as recognized by the uncloned reagents. There were no indications that the cloned cells differed from the oligoclonal reagents in their fine specificity. In contrast to previous results with an S134-associated specificity, in population studies of 25 S132-positive and 23 SB3-positive donors, no evidence could be found for subtypes of either specificity. Thus, even at the level of recognition by cloned T-cells, both S132 and SB3 appear to be remarkably homogeneous in the population.

    Notes: Summary To delineate a possible interaction of atrial natriuretic peptide ANF-(99–126) with autonomic nervous system function in humans, a spectrum of indices were assessed in 10 healthy young men during a 90 min iv administration of a) synthetic ANF-(99–126) 50 μg bolus followed by 0.025 μg·kg−1·min−1, b) the potent vasodilator sodium nitroprusside (SNP) 0.35 μg·kg−1·min−1, or c) vehicle 0.9% NaCl 40 ml and 20% albumin 5 ml, in random sequence. Plasma immunoreactive ANF (irANF) rose from 32 to 1700 pg·ml−1 during the ANF-(99–126) infusion and was stable during SNP or vehicle. Infusion of ANF-(99–126) and SNP, but not vehicle, decreased diastolic blood pressure (BP) on average by −9 and −7.5%, respectively; systolic BP was largely unchanged. Heart rate (HR, +15 and 12%) or plasma norepinephrine (NE) rose similarly during ANF-(99–126) and SNP infusions, and the systolic BP response to orthostasis was similar (−18 mm Hg). The following autonomic indices did not differ significantly after the 3 infusions: responses of HR and NE to orthrostasis; reflex bradycardic response to phenylephrine (PE)-induced rise in systolic BP (+20 mm Hg); responses of BP to hyperventilation, PE, or 3 min of sustained handgrip; and beat-to-beat variation (R-R interval) during deep breathing. The immediate orthostatic HR response (30/15 R-R interval ratio) fell similarly during infusion of ANF-(99–126) or nitroprusside. The findings indicate that in healthy men the function of the autonomic nervous system is not notably impaired by high circulating ANF levels. ANF-(99–126) infused in moderate dosage seems to lower BP largely by non-autonomic mechanisms.

    Notes: Summary The effect of the new ACE-inhibitor, fosinopril, on insulin sensitivity (SI), glucose homoeostasis and lipid profile has been examined in 24 young, healthy, normotensive men. SI, fasting plasma glucose and insulin, serum total triglycerides (Tg) and lipoprotein cholesterol (C) fractions, and ACE activity were assessed after subjects had taken placebo for 1 week and after 3 further weeks either on placebo (12 subjects) or fosinopril 20 mg daily (12 subjects), administered in a doubleblind, randomized order. Measurements were made after 3 days on a standard diet (2500 kcal/d, 45% carbohydrates, 40% fat and 15% proteins) and after an over-night fast. Compared with control values at the end of the runin placebo phase, fosinopril reduced plasma ACE activity (from 106 to 24 nmol·ml−1·min−1), Significantly increased plasma potassium and lowered upright systolic blood pressure. It also improved the k-value of the glucose disappearance rate after glucose load (from −1.70 to −1.88%·min−1) and tended to increase SI slightly although not significantly (from 10.2 to 12.0·10−4·min−1·μU−1·ml−1). Fasting plasma glucose, insulin, serum total, high-, low-, and very-low density lipoprotein cholesterol fractions and total triglycerides were unchanged following fosinopril and placebo. The findings indicate that in healthy lean humans, ACE inhibition with fosinopril is neutral with regard to lipoprotein and carbohydrate metabolism, and that it may slightly enhance cellular glucose disposal. This calls for further evaluation in individuals at high risk of developing insulin resistance and in patients with impaired insulin sensitivity related to hypertension, obesity, decreased glucose tolerance and diabetes mellitus.
