    Notizen: Abstract An in vitro biocompatibility assessment test has been successfully applied to vascular prostheses. It consists of an organotypic culture of chick embryo vascular explant which preserves wall vessel cell interactions and enables cell growth and migration on biomaterial. This study is an attempt to identify the cell phenotype expressed by scanning electron microscopy, by immunostaining with specific monoclonal antibodies against smooth muscle cell (SMC) or von Willebrand factor (vWF), and by uptake of acetylated low-density lipoprotein. Early migrating cells had an endothelial-like phenotype independent of embryonic or adult vessel, veinous or arterial explants. SMC appeared labelled beyond the inner endothelial area. At the periphery, SMC displayed actin bundles specific to stationary phase. Adult cell cultures differed from embryonic cultures in that endothelial-like cells (EC) were more stable and increased their vWF labelling, whereas in embryonic cultures, some EC rounded up with subsequent detachment. Explant culture in liquid instead of agar medium activated this degenerative process suggesting the effect of diffusible chemotactic factors. Organotypic culture in agar medium provides an in vitro co-culture system of EC and SMC and enables further investigation of spatial and temporal evolution of vascular tissues in contact with various substrate.

    Notizen: Abstract Studies were carried out to compare the behaviour of human umbilical vein endothelial cells (HUVEC) and L-929 fibroblastic cells towards proteins crosslinked by glutaraldehyde (GTA) or carbodiimide (CDI) proposed for coating of vascular prostheses. CDI crosslinking of bovine serum albumin used alone, or mixed with gelatin, allowed higher rates of cell growth and DNA synthesis than GTA crosslinking independent of cells. Assessment of the plating efficiency revealed a similar behaviour of both cells towards membranes and reference plastic surface in terms of percentages of bound cells. HUVEC proliferation onto CDI crosslinked gelatin and/or albumin membranes did not differ significantly whereas the growth of L-929 was enhanced onto gelatin albumin membranes in comparison with both gelatin membranes and the reference surface. The analysis of DNA synthesis corroborated the results of the growth curves and elicited a delay of the growth phases in HUVEC cultured onto CDI crosslinked membranes, unlike the L-929 fibroblast.

    Notizen: Abstract We examined the regulation of procoagulant activity and the production of fibrinolytic components by human vascular endothelium grown on coating membranes of gelatin, pure or mixed with albumin, crosslinked by carbodiimide ((G)C, (AG)C) in comparison with plastic culture dishes. Confluent monolayers were stimulated by human recombinant interleukin (IL-1β) and responses in terms of tissue factors like procoagulant activity, tissue plasminogen activator (tPA) and plasminogen activator inhibitor (PAI-1) were followed for up to 72 h. Procoagulant activity of cell extracts displayed similar patterns whatever the substratum tested. Quantitative immunological assays revealed a 2-fold increase in tPA antigen released from monolayers grown on (G)C and on (AG)C compared to cells grown on plastic. Exposure of monolayers to IL-1β reduced the secretion of tPA antigen which still reached higher values on coated than on uncoated substratrum. We found that the quasi-totality of tPA formed stable complexes with PAI-1, thereby suppressing measurable fibrinolytic activity. IL-1β stimulated the release of PAI-1 antigen quantified by immunoassay and the kinetics of secretion were comparable on both coated and uncoated substratum.

    Notizen: An alternative to the usual technique of preclotting porous textile vascular prostheses prior to surgical implantation is to render them impermeable to blood by impregnation with a cross-linked albumin filler matrix. This material subsequently becomes the foundation for cellular development. The compatibility of such impregnated fabrics with newly formed endothelial cells has been evaluated by an in vitro organotypic culture method. This technique enables the characterization and numeration of cells that develop on blood contact surfaces and enables determination of their rate of development. Woven, knitted, and velour fabrics were evaluated folllowing coating with albumin and either storage in Tyrode solution or 40% ethanol or desiccation by critical point drying. Preclotted cardiovascular repair fabrics prepared according to conventional surgical protocol served as controls. The identification of the newly formed cells was confirmed histologically. The most extensive and rapid cellular development was observed on the woven fabric and is believed may have resulted from the smoother surface topography of this substrate. Good cellular development was noted particularly on fabrics which had been stored in Tyrode solution. Ethanol had a deleterious effect on the apparent compatibility.

    Notizen: In an attempt to find an alternative procedure to the preclotting of porous textile vascular prostheses, the feasibility of an albumin coating and ethanol preservation technique has been evaluated by implanting albuminated polyester prostheses as substitutes in the thoracic aorta of dogs. Nine different grafts representing woven, knitted, and velour constructions were implanted for periods ranging from 4 h to 6 months. At the sacrifice, the grafts were excized for measurement of the thrombogenicity of the flow surface, for pathological examination by light microscopy and SEM, and for quantifying the changes in the textile structure during implantation. Also the kidneys were removed and examined for infarcts caused by trapped circulating emboli. The healing characteristics of the nine different grafts proved similar and followed the same sequence of events as preclotted control grafts. The albumin coating and ethanol preservation resulted in a somewhat slower rate of healing. Yet, given sufficient time, a more completely healed graft was achieved as evidenced by the presence of endothelial-like cells throughout the length of the graft. In addition, the albumin is believed to reinforce the textile structure by reducing the tendency to stretch and dilate in vivo.

    Notizen: Cell adhesion and spreading of chick embryo vascular and corneal explants grown on rough and smooth poly (methyl methacrylate) (PMMA) were analyzed to test the cell response specificity to substratum surface properties. Different degrees of roughness were obtained by sandblasting PMMA with alumina grains. Hydrophilic and hydrophobic components of the surface free energy (SFE) were calculated according to Good-van Oss's model. Contact angles were determined using a computerized angle meter. The apolar component of the SFE, γsLW, increased with a slight roughness whereas the basic component, γs-, decreased. The acido-basic properties disappeared as roughness increased. Incubation of PMMA in culture medium, performed to test the influence of the biological environment, allowed surface adsorption of medium proteins which annihilated roughness effect and restored hydrophilic properties. An organotypic culture assay was carried out in an attempt to relate the biocompatibility to substratum surface state. Cell migration was calculated from the area of cell layer. Cellular adhesion was determined by measuring the kinetic of release of enzymatically dissociated cells. A slight roughness raised the migration area to an upper extent no matter which cell type. Enhancement of the cell adhesion potential was related to the degree of roughness and the hydrophobicity. © 1997 John Wiley & Sons, Inc. J Biomed Mater Res, 36, 99-108, 1997.

    Notizen: Using canine models, a representative selection of polyester or Dacron vascular prostheses, including woven, knitted, and velour types, were evaluated for their relative healing characteristics and for their structural changes during implantation. Following residence periods ranging from 4 h to 6 months at the site of the thoracic aorta, the dogs were sacrified, and the grafts were excized for measurement of the thrombogenicity of the flow surface and for pathological examination by light microscopy and SEM. The kidneys were also removed and examined for infarcts caused by any trapped circulating emboli. The extent of healing, the presence of embolizing nuclei, and the thrombogenicity and morphology of the lumen surface were also assessed. The healing characteristics of each type of device proved similar. Velour fabrics exhibited more extensive encapsulation, but frequently their internal capsules failed to incorporate all the fibers. In all cases, cellular development on the lumen was limited to areas contiguous to the anastomoses. The initial porosity of the devices as measured by water permeability did not appear to influence the healing sequence to a significant extent. The grafts did exhibit differences in structural stability depending on whether they were of a knitted or woven construction. We suggest that users consider these different mechanical and structural properties when making their choice of a graft. Despite these differences, we believe that the healing process is far more host dependent than graft dependent.
