    Notes: Abstract: Uptake of 59Fe from blood into brains of anaesthetized rats and mice has been studied by intravenous infusion of [59Fe]ferrous ascorbate or of 59Fe-transferrin, the results not being significantly different. Uptakes in the rat were linear with time, but increased at longer times in the mouse. Transfer constants, Kin (in ml/g/h × 103), for cerebral hemispheres were 5.2 in the adult rat and 5.6 in the mouse. These Kin values corresponded to 59Fe influxes of 145 and 322 pmol/g/h, respectively. 59Fe uptake into the mouse brain occurred in the following order: cerebellum 〉 brainstem 〉 frontal cerebral cortex 〉 parietal cortex 〉 occipital cortex 〉 hippocampus 〉 caudate nucleus. In genetically hypotransferrinaemic mice, 59Fe uptake into brain was 80–95 times greater than in To strain mice. Pretreatment of young rats and mice with monoclonal antibodies to transferrin receptors, i.e., the anti-rat immunoglobulin G OX 26 and the anti-mouse immunoglobulin M RI7 208, inhibited 59Fe uptake into spleen by 94% and 98%, respectively, indicating saturation of receptors. The antibodies reduced 59Fe uptake into rat brain by 35–60% and that into mouse brain by 65–85%. Although a major portion of iron transport across the blood-brain barrier is normally transferrin-mediated, non-transferrin-bound iron readily crosses it at low serum transferrin levels.

    Notes: Lee Valley Water plc has, in a joint study with the University of Birmingham, been reassessing the reliable yields of its existing chalk sources in order to make maximum use of the available groundwater resources. An approach to yield assessment was developed which took into account the hydrogeology and flow processes in the vicinity of pumping stations, and within the contributing area. The yield of many sources was found to be very dependent on regional water level conditions. Initial estimates of potential yields were made but future monitoring should aim to clarify the interrelationship between source output and aquifer state.

    Notes: Using a polyclonal antiserum raised against the C-terminal heptapeptide of pro-enkephalin A, we have isolated the opioid heptapeptide Tyr-Gly-Gly-Phe-Met-Arg-Phe (MERF) from ovine median eminence and mapped its distribution in that structure. MERF-immunoreactivity was confined to the pars externa (neurosecretory zone) where it colocalized with corticotrophin-releasing factor in the majority of terminals. No larger, N-terminally extended forms of MERF were detected in median eminence extracts suggesting that pro-enkephalin is fully processed to its constituent enkephalin congeners, and that the bioactive products, including MERF, act at the level of the hypothalamus in regulating anterior pituitary function.

    Notes: Transgenic Trifolium subterraneum expressing a phytase gene (phyA) from Aspergillus niger were generated. Five independently transformed lines showed an average 77-fold increase in exuded phytase activity in comparison with null segregant and wild-type controls. Unlike other phosphatases, exuded phytase activity was unaffected by P supply, verifying the constitutive expression of phyA. Transgenic T. subterraneum grown in agar with P supplied as phytate, took up 1.3- to 3.6-fold more P than controls and had equivalent P uptake to plants supplied with orthophosphate. This unique phenotype was compromised when the plants were grown in soil. None of the five lines showed increased shoot biomass or total P uptake in an unfertilized, low-P soil taken from under permanent pasture. With addition of P, one of the five transgenic lines had consistently greater P nutrition compared with control plants. Despite variable growth and P nutrition responses, P uptake per root length was on average greater for transgenic lines. Exudation of phytase by transgenic T. subterraneum allowed utilization of P from phytate in non-sorbing, sterile laboratory media, but was less effective when plants were grown in soil. Release of extracellular phytase is therefore not the only requirement for the acquisition of P from endogenous soil phytate by plants.

    Notes: A chemical hybridizing agent (CHA: WL 84811, azetidine-carboxylic acid. Shell Chemical) was applied to annual ryegrass (Lolium rigidum Gaud. cv. Wimmera) at four levels: 0, 1·6, 2·4 and 3·2 kg active ingredient (a.i.) ha−1, at booting and at anthesis with a view to reducing seed set and increasing the proportion of cell solubles in the senescing and dead plants. The CHA treatments reduced stem elongation by up to 40% and reduced the proportion of pollinated florets progressively according to the concentration of CHA. At 3·2 kg (a.i.) ha−1 only 14% of florets were fertilized compared with 86% on the zero (control) treatment.The CHA treatments also resulted in progressive increases in the number of secondary tillers. Since there was no increase in the amount of neutral-detergent solubles associated with increased CHA application, it was concluded that storage carbohydrates, which might otherwise have been directed towards seed development, were partitioned towards increased tillering. There was no effect, therefore, on the proportion of neutral-detergent soluble material in senescing and dead plants.

    Notes: A part of the mechanism of the gastrointestinal toxicity exhibited by non-steroidal anti-inflammatory drugs is believed to involve the uncoupling of mitochondrial oxidative phosphorylation. Most previous uncoupling studies have used rat liver mitochondria. There is little information on the effects of the drugs on mitochondria from other species.〈section xml:id="abs1-2"〉〈title type="main"〉Aim:To study the effect of indometacin on isolated liver mitochondria from rats, mice and humans.〈section xml:id="abs1-3"〉〈title type="main"〉Methods:We studied the effects of indometacin on respiration and adenosine triphosphate synthesis by isolated liver mitochondria from rats, mice and humans. Its effects were compared with those of dinitrophenol, a classical uncoupler.〈section xml:id="abs1-4"〉〈title type="main"〉Results:Indometacin uncoupled oxidative phosphorylation at low concentrations (P 〈 0.05) and inhibited respiration at high concentrations (P 〈 0.01) in all three species. Adenosine triphosphate synthesis was, however, more sensitive to dinitrophenol or indometacin at lower concentrations in mouse and human compared to rat liver mitochondria (P 〈 0.05).〈section xml:id="abs1-5"〉〈title type="main"〉Conclusions:The current study shows that indometacin acts as an inhibitory uncoupler in human mitochondria. It also demonstrates that the responses of rat, mouse and human mitochondria to indometacin are broadly similar.
