    Notes: Abstract The budding yeast Saccharomyces cerevisiae divides asymmetrically, giving rise to a mother cell and a smaller daughter cell. Individual mother cells produce a finite number of daughter cells before senescing, undergoing characteristic changes as they age such as a slower cell cycle and sterility. The average life span is fixed for a given strain, implying that yeast aging has a strong genetic component. Genes that determine yeast longevity have highlighted the importance of such processes as cAMP metabolism, epigenetic silencing, and genome stability. The recent finding that yeast aging is caused, in part, by the accumulation of circular rDNA molecules has unified many seemingly disparate observations.

    Notes: Abstract The Labour government elected in the United Kingdom in May 1997 has embarked on the country's biggest ever program of constitutional change. This chapter sets out the main constitutional measures adopted so far and identifies further changes that will need to follow in order to accommodate these early changes. The new constitutional architecture in the United Kingdom is a form of quasifederalism in which new institutions will have to be created to rebalance the political center if the Union is to hold together.

    Notes: A postal survey was conducted into the use of performancemeasurement in companies using total quality management. A total of 89responses were obtained from 22 companies. The results from the analysisof the data show that even in companies using TQM, there is a gapbetween the factors of performance perceived by employees as beingimportant, and the factors which are emphasized by their organizations.However, some of the key goals of TQM - customer service andsatisfaction, and quality improvements - are among the highestrated factors in terms of both importance and emphasis.

    Notes: This article outlines the impact of significant constitutional reform in the UK. It uses scenarios of minimum and maximum outcomes of the process of devolving government to Wales, Scotland and Northern Ireland. Domestic issues could include financial tensions, human rights, parliamentary reform, reorganization of the civil service and moves towards a written constitution for the UK. Internationally, changes to the UK's relations with other countries may result. The article concludes that currently the minimalist scenario is the closest representation of the UK's constitutional future.

    Notes: Sustainable competitiveness can only come through buildingrobustness into processes and their effective management and control.Through a culture of continuous improvement and using the Deming Cycleof plan-do-check-act, complacency can be prevented and value optimizedfor the end customer. Measurement is the trigger for process improvementand the achievement of superior competitive standards. In the first ofthree articles, following a review of the literature, studies theapplications of total quality-based performance measurement through twocase studies. The type of practices highlighted suggest that performancemeasurement forms a broad, integrated concept in organizations whichhave implemented TQM. Few organizations have developed a separate"performance measurement system"; performance measurementinstead forms an integral part of the management processes and systemswithin the organization. In the second article, develops a comparison of15 case studies. From this analysis, introduces and tests a model oftotal quality-based performance measurement in the third article of theseries.

    Notes: The glycine decarboxylase complex (GDC) is composed of four subunits referred to as H-, L-, P-, and T-proteins. The Saccharomyces cerevisiaeGCV2 gene, encoding the P-protein has been cloned by complementation of the gsd2 mutation which prevents cells converting glycine to serine or using glycine as the sole nitrogen source. The gene, located on the right arm of chromosome XIII adjacent to TPS1, encodes a product with a Mr of 114 385. Expression of GCV2 was induced by the addition of more than 200 μM glycine in the medium, and a maximal sixfold induction occurred above 1 mM. This response was specific to glycine and was not observed for any other amino acid. Under the same conditions, the intracellular level of glycine increased up to 30-fold. The levels of P- and L-protein transcripts and GDC activity were also elevated in cells grown in the presence of glycine. Deletion analysis of the GCV2 promoter delimited the control region which contains putative regulatory sites for GCN4 and GLN3 transcription factors.

    Notes: Barring genetic manipulation, the diet known as calorie restriction (CR) is currently the only way to slow down ageing in mammals. The fact that CR works on most species, even microorganisms, implies a conserved underlying mechanism. Recent findings in the yeast Saccharomyces cerevisiae indicate that CR extends lifespan because it is a mild biological stressor that activates Sir2, a key component of yeast longevity and the founding member of the sirtuin family of deacetylases. The sirtuin family appears to have first arisen in primordial eukaryotes, possibly to help them cope with adverse conditions. Today they are found in plants, yeast, and animals and may underlie the remarkable health benefits of CR. Interestingly, a class of polyphenolic molecules produced by plants in response to stress can activate the sirtuins from yeast and metazoans. At least in the case of yeast, these molecules greatly extend lifespan by mimicking CR. One explanation for this surprising observation is the ‘xenohormesis hypothesis’, the idea that organisms have evolved to respond to stress signalling molecules produced by other species in their environment. In this way, organisms can prepare in advance for a deteriorating environment and/or loss of food supply.
