Notizen: [Auszug] Laying queens were immobilized at -20 °C. Mandibular glands were excised, macerated and extracted twice in HPLC-grade methanol to give a combined total extract of 100 JJL! per queen. A 3-queen equivalent (Qeq) HQMC extract was chromatographed on a micro-column of acid-treated silica gel. The ...

Notizen: Abstract  Disruptive selection for responsiveness to queen mandibular gland pheromone (QMP) in the retinue bioassay resulted in the production of high and low QMP responding strains of honeybees (Apis mellifera L.). Strains differed significantly in their retinue response to QMP after one generation of selection. By the third generation the high strain was on average at least nine times more responsive than the low strain. The strains showed seasonal phenotypic plasticity such that both strains were more responsive to the pheromone in the spring than in the fall. Directional selection for low seasonal variation indicated that phenotypic plasticity was an additional genetic component to retinue response to QMP. Selection for high and low retinue responsiveness to QMP was not an artifact of the synthetic blend because both strains were equally responsive or non-responsive to whole mandibular gland extracts compared with QMP. The use of these strains clearly pointed to an extra-mandibular source of retinue pheromones (Pankiw et al. 1995; Slessor et al. 1998; Keeling et al. 1999).

Notizen: Abstract Quantitative laboratory biosassays measuring short-range attraction of worker honeybees to pheromone-treated pipets or glass pseudo-queens are described. Each replicate involves only 15 workers, allowing a single colony to provide sufficient individuals for a complete study as well as for between-colony comparisons. The more sensitive pseudo-queen bioassay provides a measurable response at levels of queen mandibular extract of 10−7 than that of an average individual mated queen. Formation and maintenance of the pseudo-queen's retinue, as well as more detailed behavior, can be evaluated by following the actions of individual workers replayed on video tape. Using this sensitive bioassay, the response of workers to queen mandibular pheromone has been shown to be sensitive to daily variation and colony source of the workers, but independent of worker age.

Notizen: Abstract The ontogeny of the five queen mandibular gland semiochemicals that initiate and maintain the retinue behavior of worker honeybees was investigated by quantitative splitless capillary gas chromatography. No detectable pheromone is present at the time of eclosion, but decenoic acid levels build up rapidly during the first week of the queen's life. Two aromatic components attain detectable levels later, with the more plentiful methylp-hydroxybenzoate preceding the 4-hydroxy-3-methoxyphenylethanol. Pheromone levels are maximal in mature, mated, laying queens. The ratio of (R,E)-(−)-9-hydroxy-2-decenoic acid to the (S,E)-(+) enantiomer increases with the age of the queen. Pheromone levels in queen mandibular glands are largely unaffected by queen banking, restraint with workers in mailing cages, and limited storage on dry ice. All major body parts of typical queens, especially the head and legs, have sufficient mandibular exudate to be highly attractive to worker bees.

Notizen: Abstract Ipsenol was identified from the frass of male, but not female,Ips latidens from British Columbia, feeding in phloem tissue of lodgepole pine,Pinus contorta var.latifolia. The responses ofJ. latidens to sources of ips-enol andcis-verbenol were determined with multiple-funnel traps in stands of lodgepole pine in British Columbia. Ipsenol attracted both male and femaleI. latidens, verifying that it is a pheromone for this species. MaleI. latidens showed a slight preference for (S)-(−)-ipsenol.cis-Verbenol was not produced by beetles of either sex and, in contrast to an earlier report, both enantiomers inhibited attraction to ipsenol-baited traps. The predators,Enoclerus sphegeus andThanasimus undatulus (Cleridae), were attracted to traps baited withcis-verbenol and ipsenol.

Notizen: Abstract The sex pheromone of the western hemlock looper (WHL),Lambdina fiscellaria lugubrosa (Hulst), comprises three methylated hydrocarbons: 5,11-dimethylheptadecane (5,11), 2,5-dimethylheptadecane (2,5), and 7-methylheptadecane (7). Compounds extracted from female pheromone glands were identified by coupled gas chromatographic-electroantennographic (GC-EAD) analysis and coupled GC-mass spectroscopy in selected ion monitoring mode. In trapping experiments, (5,11) alone attracted male moths, but addition of either (7) or (2,5) significantly enhanced attraction. (5,11) combined with both (7) and (2,5) was significantly most attractive. (5,11) and (2,5) are also sex pheromone components of the eastern hemlock looper (EHL),Lambdina fiscellaria fiscellaria (Guen.). Although (7) is produced by the EHL, it is a pheromone component only in the WHL. It constitutes the first behaviorally active monomethyl-branched hydrocarbon to be found in a geometrid and is a novel lepidopteran sex pheromone component. The different 2- versus 3-component sex pheromone supports taxonomic division of EHL and WHL.

Notizen: Abstract Three candidate sex pheromone components, (Z)6,(Z)9-, (Z)6,(E)8-, and (Z)6,(E)9-heneicosadien-11-one (Z6Z9, Z6E8, and Z6E9) were identified in pheromone gland extracts of female Douglas-fir tussock moths (DFTM), Orgyia pseudotsugata (McDunnough). Their occurrence in subnanogram quantities in extracts and structural conversion during analytical procedures and bioassays complicated chemical identifications. Complete identification required comparative analyses of stereoselectively synthesized and female-produced dienones by coupled gas chromatographic–electroantennographic detection (GC-EAD), high-performance liquid chromatography (HPLC) and coupled GC–mass spectrometry (MS). Determination of the pheromone component was contingent upon an experimental design that minimized structural rearrangement of dienones before and during the field test. In a 40-min field experiment, acetonitrile solutions of each of the above dienones were carried on Dry Ice to traps and were syringed onto cotton release devices below trap lids. In combination with the previously known sex pheromone component of DFTM, (Z)6-heneicosen-11-one (Z6), Z6E8 was the only synergistic dienone and the mixture was highly attractive. Because Z6 by itself attracts seven species of tussock moths (two sympatric with DFTM), a blend of Z6 and Z6E8 may impart specificity to DFTM pheromone communication. In commercial lures, this binary blend may facilitate species-specific, sensitive monitoring and efficacious control by mating disruption of this important forest defoliator.
