    Notes: : A main water quality concern is accelerated eutrophication of fresh waters from nonpoint source pollution, particularly nutrient transport in surface runoff from agricultural areas and confined animal feeding operations. This study examined nutrient and β17-estradiol concentrations in runoff from small plots where six poultry litters were applied at a rate of about 67 kg/ha of total phosphorus (TP). The six poultry litter treatments included pelleted compost, pelleted litter, raw litter, alum (treated) litter, pelleted alum litter, and normal litter (no alum). Four replicates of the six poultry litter treatments and a control (plots without poultry litter application) were used in this study. Rainfall simulations at intensity of 50 mm/hr were conducted immediately following poultry litter application to the plots and again 30 days later. Composite runoff samples were analyzed for soluble reactive phosphorus (SRP), ammonia (NH4), nitrate (NO3), TP, total nitrogen (TN) and β17-estradiol concentrations. In general, poultry litter applications increased nutrient and β17-estradiol concentrations in runoff water. Ammonia and P concentrations in runoff water from the first simulation were correlated to application rates of water extractable NH4 (R2= 0.70) and P (R2= 0.68) in the manure. Results suggest that alum applications to poultry litter in houses in between flocks is an effective best management practice for reducing phosphorus (P) and β17-estradiol concentrations in runoff and that pelleted poultry litters may increase the potential for P and β17-estradiol loss in runoff water. Inferences regarding pelleted poultry litters should be viewed cautiously, because the environmental consequence of pelleting poultry litters needs additional investigation.

    Notes: Abstract Rapid Distortion Approximations (RDA) may be used to simplify the Reynolds stress equations in rapidly distorted flows, as suggested by Dussauge and Gaviglio (1987). These approximations neglect diffusive and dissipative terms while retaining the production and pressure terms. The retained terms are then modeled as functions of the Reynolds stress tensor and gradients of the mean flow. The models for the pressure-strain term as developed by Lumley (1978) and Shih and Lumley (1985) are evaluated by comparing the calculated results with experimental data for the case of a Mach 2.84 turbulent boundary layer in a 20° centered expansion. The agreement between computed and experimentally obtained Reynolds stresses was found to be encouraging.

    Notes: Abstract Two experiments were performed to study the response of a supersonic turbulent boundary layer to successive distortions. In the first experiment (Case 1), the flow passed over a forward-facing ramp formed by 20° compression corner followed by a 20° expansion corner located about 4δo downstream, where δo is the incoming boundary layer thickness. In the second experiment (Case 2), the forward-facing ramp was constructed of curved compression and expansion surfaces with the same turning angles and total step height as in Case 1. The radii of curvature for the compression and expansion surfaces were equal to 12δo. In both experiments, the flow relaxation was observed over a distance equal to 12δo. In this relaxation region, the mean and turbulent flow behavior of the boundary layer was measured. The mean velocity profile was found to be altered by the distortion. Recovery of the profile began near the wall and occurred rapidly, but in the outer part of the boundary layer, recovery proceeded slowly. Turbulence measurements revealed a dramatic reduction in the turbulence shear stress and a progressively decaying streamwise Reynolds stress profile.

    Notes: The facilitated transport of carbon monoxide through thin, immobilized films of cuprous chloride solutions is investigated, using a tracer C14O technique. Steady-state tracer flux measurements are used to determine the second-order forward rate constant and equilibrium constant for the reversible reaction of CO with cuprous ion. The enchanced results at low CO partial pressures can be interpreted in terms of a consecutive reaction model. The cuprous ion is remarkably effective as a carrier for CO. The CO permeation rate in the presence of cuprous ion can be increased by two orders of magnitude over the nonfacilitated flux. As such, the cuprous ion is an attractive facilitating agent for use in a membrane separation system.

    Notes: Summary Two clones from the Clarke-Carbon Escherichia coli colony bank were resistant to inhibition by trimethoprim, a potent inhibitor of dihydrofolate reductase. Both clones had elevated levels of dihydrofolate reductase. Furthermore, trimethoprim resistance and elevated enzyme levels were associated with ColE1 plasmids that carried DNA from the trkC ksgA pdxA region of the E. coli chromosome. Plasmid pLC1437a was shown by two criteria to carry the structural gene for dihydrofolate reductase: 1) A partial diploid containing plasmid pLC1437a produced a kinetically-recognizable dihydrofolate reductase that was not present in the parent haploid strain. 2) Plasmid pLC1437a coded for dihydrofolate reductase in vitro. A 1,000 base pair fragment of plasmid pLC1437a containing fol was used as a probe to measure fol mRNA in a mutant strain isolated by Sheldon and Brenner (Molec. gen. Genet. 147, 91–97, 1976). The mutation in this strain, which results in constitutively-high levels of dihydrofolate reductase and in the inability of the strain to grow at 42° C, is cis dominant (Sheldon and Brenner, 1976). The results of kinetic hybridization and pulse-labeling experiments indicated that the regulatory mutant produced elevated levels of dihydrofolate reductase in response to an increased rate of synthesis of fol mRNA.

    Notes: Summary We report the construction of recombinant plasmids containing the dihydrofolate reductase structural gene (fol) from several trimethoprim-resistant mutants of Escherichia coli. Strains carrying some of these plasmids produced approximately 6% of their soluble cell protein as dihydrofolate reductase and are therefore excellent sources of the purified enzyme for inhibitor binding or mechanistic studies. The nucleotide sequence of the fol region from each of the plasmids was determined. A plasmid derived from a Ki mutant which produced a dihydrofolate reductase with lowered affinity for trimethoprim contained a mutation in the structural gene that altered the sequence of the polypeptide in a conserved region which is adjacent to the dihydrofolate binding site. Two other independently-isolated mutants which overproduced dihydrofolate reductase had a mutation in the-35 region of the fol promoter. One of them, strain RS35, was also temperature-sensitve for growth in minimal medium. This phenotype was shown to be the result of an additional mutation in a locus unlinked to fol by P1 transduction. The fol regions from two temperature-independent revertants of strain RS35 were sequenced. One of these had a mutation within the dihydrofolate reductase structural gene which altered some properties of the enzyme. This confirmed some previous enzymological data which suggested that some revertants of strain RS35 had mutations in fol (Sheldon 1977). These results suggest that dihydrofolate reductase interacts physically with some other essential gene product in E. coli.
