Notizen: Abstract Posttranslational modification of proteins by phosphorylation is a universal mechanism for regulating diverse biological functions. Recognition that many cellular proteins are reversibly phosphorylated in response to external stimuli or intracellular signals has generated an ongoing interest in identifying and characterizing plant protein kinases and protein phosphatases that modulate the phosphorylation status of proteins. This review discusses recent advances in our understanding of the structure, regulation, and function of plant protein phosphatases. Three major classes of enzymes have been reported in plants that are homologues of the mammalian type-1, -2A, and -2C protein serine/threonine phosphatases. Molecular genetic and biochemical studies reveal a role for some of these enzymes in signal transduction, cell cycle progression, and hormonal regulation. Studies also point to the presence of additional phosphatases in plants that are unrelated to these major classes.

Notizen: The best available energy level data for the cubic Cs2NaLnCl6 and Cs2NaYCl6:Ln3+ (doped) systems are collected together and summarized. These data are analyzed in terms of a parametric model which assumes an octahedral (Oh) site symmetry for the Ln3+ ions and a 4 f N "free-ion'' Hamiltonian similar to that used previously in energy level analyses of the LaCl3:Ln3+ systems. The empirical energy level data are fit to the parametrized 4 f N Hamiltonian, and the resulting "best-fit'' parameter sets and calculated energy levels are presented and discussed.

Notizen: Knowledge is a resource that is valuable to an organization's ability to innovate and compete. It exists within the individual employees, and also in a composite sense within the organization. According to the resource-based view of the firm (RBV), strategic assets are the critical determinants of an organization's ability to maintain a sustainable competitive advantage. This paper will combine RBV theory with characteristics of knowledge to show that organizational knowledge is a strategic asset. Knowledge management is discussed frequently in the literature as a mechanism for capturing and disseminating the knowledge that exists within the organization. This paper will also explain practical considerations for implementation of knowledge management principles.

Notizen: Advances in computer technology facilitate innovative methods for delivering training in organizations. The Internet enables the delivery of computer-based training across time and distance. This medium, known as Web-based distance learning, provides opportunities to develop human resources to support creating a competitive advantage for an organization. This article discusses principles of WBDL design, measuring the effectiveness of training delivered through that design. Further, the relationship between WBDL and human resource development is provided. Suggestions for organizations planning to implement WBDL are included.

Notizen: Abstract Type 1 phosphoprotein Ser/Thr phosphatases (PP1) are highly conserved enzymes found in all eukaryotes. These enzymes have multiple functions in fungal and animal cells but little is known of their function and regulation in plants. Previous studies in our laboratory indicated that maize and Arabidopsis contain a family of PP1 genes and/or pseudogenes. In this study, we report the isolation of five distinct Arabidopsis cDNA clones (TOPP1, TOPP2, TOPP3, TOPP4 and TOPP5) which encode the catalytic subunit (PP1c) of type 1 protein phosphatases. Genomic Southern blot analyses indicate that these clones are the products of five distinct genes and that an additional 2–3 PP1c genes and/or pseudogenes may be present in the Arabidopsis genome. The derived amino acid sequences of the TOPP clones are very similar to published sequences of PP1c from animals, fungi and plants. Four of the TOPP amino acid sequences show unique structural features not observed in other PP1c sequences from fungi or animals. All of the TOPP genes are expressed in Arabidopsis roots, rosettes and flowers, although TOPP1, TOPP2 and TOPP3 appear to be expressed at higher levels in these tissues than TOPP4 and TOPP5.

Notizen: Abstract Type one serine/threonine protein phosphatases (PP1s) have been implicated in various processes of plant growth and development. In all plant species studied, PP1s are encoded by multigene families. Previous studies in our laboratory identified five Arabidopsis thaliana PP1 genes (TOPP1, TOPP2, TOPP3, TOPP4 and TOPP5). In the present study, we report the isolation of three additional PP1 genes (TOPP6, TOPP7 and TOPP8). Southern blot analyses indicate that these three newly isolated genes are single-copy genes in A. thaliana genome. All the three genes are expressed in roots, rosettes and flowers, although their expression levels appear to be lower than those of the five previously identified TOPP genes. Six of the eight TOPP genes were mapped to different positions on four of five A. thaliana chromosomes. Sequence comparison revealed that TOPP genes belong to different subgroups of plant PP1 genes, suggesting that they may encode proteins with distinct functions.

Notizen: An experimental program is described in which the capacitor-discharge initiation characteristics of a detonator containing a vacuum-deposited thin-film chromium bridge were studied. The objective of the effort was to define the conditions that would result in overall function times of 10 μs or less. The threshold initiation energy of the detonator was in the range of 11.5 mJ-20.0 mJ. Consistent performance was obtained from a firing energy of 54 mJ, which was achieved by a 5-μF capacitor charged to 147 V. Under these conditions, the average overall function time was 4.2 μs, with a standard deviation of 0.2 μs. Other tests showed that at higher capacitances and lower voltages the function time increased substantially and became more nonreproducible, even though the stored energy was considerably above the threshold value.
