    Notes: Summary Copper-zinc Superoxide dismutase is known to protect pancreatic β-cell function from the damage of alloxan. A morphological investigation of copper-zinc Superoxide dismutase prophylaxis against alloxan was undertaken in rats to investigate the mechanism of this protective action. Exogenous copper-zinc Superoxide dismutase reproducibly protected the morphological features of pancreatic β cells against damage by alloxan as determined by light microscopic immunostaining for insulin and by ultrastructural examination. By the same criteria, α and δ cells appeared unaffected by administration of a combination of alloxan and copper-zinc superoxide dismutase or either agent alone. Autoradiography after injection of 125I labelled copper-zinc superoxide dismutase into normal rats showed no evidence that the enzyme enters viable islet cells, suggesting an extracellular site of protection against alloxan.

    Notes: Plasmodium berghei infection was more severe in pregnant than in nonpregnant mice. Infection initiated on gestation day 7 resulted in rapidly increasing parasitemia and deaths of all pregnant mice within 12 days, while some nonpregnant mice survived until day 21 postinfection. When mice were infected on gestation day 12 or 14, a proportion of mice died before parturition; but some animals survived to deliver living pups. Reduced birthweights and increased spleen weight to body weight ratios were seen in pups from infected mice as compared with pups from uninfected animals. Histopathological abnormalities of placentae from infected animals included degeneration of the normal labyrinthine architecture and thickening of the trophobast separating maternal and fetal blood vessels.

    Notes: Abstract– 14CO2 production and 14C incorporation into proteins was studied in isolated rat sciatic nerves during incubation with 0.1 mM-[1-14C]leucine. Rats were made diabetic with streptozotocin. Nerves from diabetic rats incubated with glucose oxidized more [14C]leucine than controls. This difference was abolished in the presence of insulin (1 mU/ml). The effects of diabetes and insulin on leucine oxidation could not be demonstrated in the absence of glucose. Insulin stimulated the incorporation of [14C] from leucine into proteins by nerves from controls and diabetic rats.Nerves undergoing Wallerian degeneration showed a marked increase in DNA content and stimulated incorporation of [14C]leucine into proteins. 14CO2 production from leucine proceeded at 75% of the rate observed in intact nerves. Neither insulin nor diabetes affected leucine metabolism in degenerating nerves.Neither the extracellular space nor the concentration of free amino acids were significantly different in nerves obtained from control and diabetic rats, except for lower glutamine content in the latter. In vitro leucine metabolism of nerves is affected by diabetes, insulin and the integrity of the axon. The Schwann cell is suggested as a possible site of the observed changes in leucine metabolism.

    Notes: Histochemical methods specific for carbohydrate constituents distinguish a number of widely different entities in several types of gastric epithelial cells in the dog.The surface gastric epithelium reveals basophilia, indicating mucosubstances rich in both carboxyl groups (presumably sialic acid) and sulfate esters. The carboxyl-type basophilia predominates in the superficial convolutions, and that attributed to sulfate is found in the foveolar walls. The strong reactivity with both the periodic acid-Schiff (PAS) and the periodic acid-para diamine (PAD) methods for vicinal-hydroxyls indicates presence of hexose- or deoxyhexose-rich mucosubstances in both of these areas. Susceptibility of basophilia to blockage by initial periodate-meta diamine treatment points to presence of vicinal-glycols in proximity of the acid groups in surface epithelium mucin. The surface epithelium in the pylorus differs slightly in that its secretion is less strongly basophilic.The mucous neck cells secrete an apparently neutral mucosubstance which differs from the surface epithelium in its lack of both PAD reactivity and basophilia. The parietal cells also form a diastase-resistant, neutral, carbohydrate-rich substance. The zymogen cells are abundant, deep in the glands of the cardiac portion and the body of the stomach, but less numerous in the intermediate zone. They contain apical granular secretion apparently rich in sulfated mucosubstances. Lack of reactivity toward vic-glycol methods, resistance of alcianophilia or azurophilia to a previous periodate-meta diamine sequence, and resistance of mixed diamine staining to previous periodate oxidation indicate absence of residues with unsubstituted adjacent hydroxyls (e.g., hexose or deoxyhexose), in this acid mucosubstance.The pyloric glands resemble mucous neck cells in their strong PAS staining and lack of PAD reactivity, but differ in that they show a weak staining indicative of sulfomucin.
