    Notes: Summary Lectin histochemical methods and immunohistochemical techniques have been utilized to investigate and partially characterize glycoconjugates in the developing eye. Peanut-lectin-binding sites associated with radial glial cells were found in the diencephalon. In the optic primordia, binding sites associated with radial glia were masked by terminal sialic acid, and only reacted with peanut lectin when pretreated with sialidase. This finding indicates that glycoconjugates associated with diencephalic radial glia contain terminal galactose-β-(1→3)N-acetyl galactosamine, but glycoconjugates associated with radial glia in the optic primordia contain sialic acid→galactose-β(1→3)N-acetyl galactosamine. The selective distribution of galactose, N-acetyl galactosamine and fucose associated with radial glial cells has also been demonstrated. We postulate that these distributions mediate the shaping of the developing eye.

    Notes: Summary Fine structural localization of acid phosphatase and acid mucosubstance in the human blood platelet has shown that these two components coexist in the platelet α granules. Within the α granules acid phosphatase activity and acid mucosubstance both appear confined to the nucleoid region. Additionally, heavy acid phosphatase activity filled occasional small cytoplasmic bodies as well as elongated irregular cell organelles which latter appeared to correspond with atypical granules in morphologic preparations. A complex of mucosubstance with nonenzymatic protein, sequestering the lysosomal enzyme, or a complex of mucosubstance with the enzyme reversibly inactivating it, might explain the cytochemical latency of acid phosphatase in certain lysosomes and the masked dialyzed iron affinity in these sites after glutaraldehyde fixation.

    Notes: Summary Fine structural localization of arylsulfatase in the rabbit blood platelets has been investigated in this study. Among many cell organellae, reaction products were exclusively observed in the alpha granules of the platelets. Whithin the alpha granules, arylsulfatase activity appeared to localize in variable patterns, i.e. reaction products confined mainly at the peripheral region in many granules, while they deposited heavily throughout the granule matrices in some others. In a blood platelets, each alpha granule showed the different staining pattern which indicated more variable functional heterogeneity in the granules.

    Notes: Summary Senile plaque amyloid of both isolated and perivascular (dyshoric) types was compared histochemically to other major types of amyloid. In contrast to most amyloids tested, senile plaque amyloid contained less tryptophan and tyrosine reactivity and more alcianophilia. These histochemical features indicate that senile plaque amyloid is similar to apudamyloid, and suggest that local factors are more important than systemic circulating factors in its genesis. The implications of these histochemical observations are discussed.

    Notes: Summary Four cases of elastofibroma located in the subscapular region of 3 men aged 66, 74, and 83 years, and a woman 70 years old are reported. A correlated light and electron microscopic study including ultrastructural examination of Verhoeff's iron-hematoxylin (VIH)-stained sections was performed. Light microscopically, the elastofibromas were characterized by connective tissue built up by collagen fibers and sclerotic masses mingled with numerous fibers and globules of elastin material. In one micron thick Epon sections these elastin fibers often revealed a central axis surrounded by a mantle composed of periodic segments giving them a necklace-like appearance. The ultrastructural findings of these elastin structures, stained with VIH, and the appearance of the stroma cells and their relation to the elastin indicate that elastofibroma is a non-neoplastic reactive lesion in which elastin material is synthesized by the stromal cells and predominantly laid down around preexisting elastic fibers.

    Notes: Summary Copper-zinc superoxide dismutase (CuZn-SOD) has been localized in formalin-fixed, paraffin-embedded sections of both canine and rat brains. Staining with an immunoenzyme bridge sequence revealed CuZn-SOD in all regions of the brains examined. Specific sites of localization included cerebral cortical pyramidal cells, cerebellar Purkinje cells, neurons in ‘subcortical nuclei’, and oligodendrocytes throughout the brain. Similar sites of CuZn-SOD localization were identified in both species. These results are compared with reports by various investigators of SOD bioactivity in the brain.

    Notes: Summary Sections of fixed, paraffin-embedded brain from mice and rats were stained with agglutinin fromVicia villosa conjugated to horseradish peroxidase (VVA-HRP) to localize glycoconjugate containing terminalN-acetylgalactosamine (GalNAc). VVA-HRP binding sites were localized in periodic foci at the surface of a selective population of non-pyramidal interneurons in layers II through VI of the rodent cerebral cortex. These multipolar interneurons were shown to utilize γ-aminobutyric acid (GABA) as a transmitter and thus to be GABAergic by their immunostaining for glutamic acid decarboxylase (GAD) throughout the cytoplasm in serial sections. Pyramidal and other non-pyramidal cortical neurons received GABAergic input as evidenced by punctate immunostaining for GAD on their soma and proximal dendritic aborizations, but these cells failed to show VVA affinity or cytosolic GAD reactivity. Most neurons in the thalamic reticular nucleus stained for the presence of glycoconjugate with terminal GalNAc on their surface and for GAD in the cytosol. In contrast, cerebellar Purkinje cells showed strong cytosolic reactivity with anti-GAD but lacked surface staining with VVA-HRP. These observations show that some but not other populations of GABAergic neurons possess binding capacity for VVA on their surface. The surface of neurons in the deep cerebellar nucleus stained heavily with VVA but failed to show clear cytosolic reactivity for GAD. Some neurons with surface glycoconjugate containing terminal GalNAc are therefore not GABAergic.

    Notes: Summary Bridging between the two bodies of cell science, histochemistry has brought together knowledge of morphological structures and biochemical constituents and provided insight into the function of one or the other. Where it has localized an enzyme, hormone or other entity of known biological activity to a cell type, histochemistry has contributed insight into the cell's function. By detecting heterogeneity in the content of an enzyme or glycoconjugate within a presumed uniform population of cells, the histochemical approach has differentiated among these cells subtypes with demonstrated or presumed differences in function. On the other hand, in instances where it has located an isozyme, antigen, glycoconjugate or other entity of uncertain significance in a cell or organelle of known function, histochemistry has suggested a possible role for the constituent related to that of the structure. Histochemical examination provides the observer with a different view of body tissues and their composition from that obtained by strictly morphological or chemical techniques. Information acquired through this advantage is cited wherein an immunohistochemical method disclosed previously undiscovered neural organs and carbohydrate histochemistry detected previously unrecognized glycoconjugates.

    Notes: Summary Ultrastructural immunostaining of carbonic anhydrase in gastric parietal cells was accomplished with the immunoglobulin-peroxidase bridge procedure applied to cryostat sections of fixed guinea-pig stomach prior to dehydration and embedment. Of a variety of fixatives tested, only freshly prepared paraformaldehyde buffered with calcium acetate provided both immunostaining and adequate preservation of ultrastructural morphology. Delipidization or exposure of specimens to detergent prior to staining enhanced the intensity of the immunostaining and increased the sensitivity of the method. Increased diaminobenzidine concentration in the peroxidase substrate appeared also to intensify the densification at the reactive site. Carbonic anhydrase was localized ultrastructurally with this pre-embedment immunobridge procedure in the hyaloplasm of gastric parietal cells and less consistently in the superficial surface epithelium. The basal portion of the parietal cells stained more intensely than the apical region and immunoreactivity appeared concentrated at the plasmalemma and around mitochondria.
