    Notes: Matter produced by organisms is remarkable. Evolutionary optimized properties, e.g.regarding hydrodynamic, aerodynamic, wetting and adhesive behavior, can already be found in the“simplest” forms of organisms. Euglena gracilis, a single-celled algal species, performs tasks asdiverse as sensing the environment and reacting to it, converting and storing energy andmetabolizing nutrients, living as a plant or an animal, depending on the environmental constraints.We developed a preparation method for atomic force microscopy investigation of dried wholeEuglena cells in air and obtained data on whole cells as well as cell parts. Our studies corroborateTEM, SEM and optical microscopy results. Furthermore, we found new features on the pellicle, andset the ground for AFM force spectroscopy and viscoelastic studies on the nanoscale

    Description / Table of Contents: Zusammenfassung Der Nachweis von Blutplasma in erhitzten Fleischwaren wurde mit Hilfe der Ultradünn schicht-isoelektrischen Focussierung durchgeführt. Wegen der geringen Löslichkeit von denaturiertem Blutplasma in Wasser bzw. verdünnten Salzlösungen wurde der Extrakt durch Adsorption des Proteins an Hydroxylapatit und nachfolgende Elution konzentriert. Die für die isoelektrische Focussierung benötigte salzfreie Probe wurde durch Abtrennung der niedermolekularen Substanzen mittels Gelchromatographie durchgeführt. Wegen der hohen Trennschärfe sowie aus Zeit- und Kostengründen wurde die isoelektrische Focussierung in Gelen mit 150 μm Dicke durchgeführt. Mit dieser Methode können noch 0,2% Trockenblutplasma in Brühwürsten nachgewiesen werden.
    Notes: Summary The detection of blood plasma in heat-treated meat products was performed by means of ultrathinlayer isoelectric focusing. Because of the poor solubility of denatured blood plasma in water or diluted salt solutions the extracts were concentrated by adsorption of the protein on hydroxyl apatite and subsequent elution. Desalting of the samples was done by gel chromatography. Gels with a thickness of 150 μm were used for isoelectric focusing because of their exellent resolution as well as low expenditure of time and cost. 0.2% of dry blood plasma in sausages were still detectable by the method described.

    Notes: Abstract Investigations of the submicroscopic structure of gluten protein fractions by means of SEM have revealed significant changes in the structure of glutenins due to the removal of flour lipids by extraction with petroleum ether and chloroform-methanol resp. Defatting of the flour caused a rupture of the sheetlike structure observed in the glutenin of untreated flour and an increase of the amount of fibrillar components. The number of spherical particles arising from delipidation with petroleum ether decreases considerably in the glutenin from chloroform-methanol defatted flour. It has been concluded that these spherical particles may consist mainly of chloroform-methanol soluble lipoproteids originally existing in wheat flour. Furthermore it could be shown that the gliadin fraction also is very heterogenous and consists of sheetlike, fibrillar, and spherical aggregates.

Die hier beschriebenen Methoden (Lichtmikroskopie, mikrochemische Reaktionen, Emissionsspektroskopie, Chromatographie, Rasterelektronen-Analyse und Infrarotspektroskopie) sind hierfür von zunehmender Bedeutung. Sie ermöglichen ein besseres Verständnis neuerer kunsthistorischer Probleme nur durch die richtige Interpretation der Ergebnisse wissenschaftlicher Methoden.
    Notes: Summary The fundamental importance of scientific research for the preservation and restoration of works of art is uncontestable. The methods described in this paper, such as light microscopy, microchemical tests, emission spectroscopy, chromatography, scanning electron analysis, electron microprobe analysis and infrared spectroscopy, are of special significance in this filed. Nevertheless a better understanding of the problems that arise out of new special questions of art history can be obtained only by the right interpretation of the data obtained by scientific methods.

    Description / Table of Contents: Zusammenfassung Vergleichende Untersuchungen mit Hilfe der SDS-PAGE an reduzierten Gluteninen aus unbehandeltem und mit Petroläther bzw. Chloroform-Methanol entfettetem Weizenmehl haben ergeben, daß sowohl die Molekulargewichtsverteilung der Untereinheiten als auch das durch Densitometrie ermittelte Intensitätsverhältnis der elektrophoretischen Banden durch die Entfernung der Lipide stark beeinflußt wird. Es zeigte sich vor allem, daß durch die Entfettung die Ausbeute an hochmolekularen Komponenten bei Anwendung der AUC-Extraktion merklich vermindert wird. Aus der Untersuchung eines mittels reduzierender Extraktion (0,2% NaHSO3 in 60% Propanol-2) gewonnenen Gluteninpräparates haben sich Hinweise auf die Beteiligung Gliadin-ähnlicher Untereinheiten an der Gluteninstruktur ergeben.
    Notes: Summary Comparative investigations on the molecular weight distribution of the reduced glutenins from untreated and petroleum ether resp. chloroform-methanol defatted wheat flour by means of SDS-PAGE have shown that the molecular weight distribution and the intensity ratios of the electrophoretic bands are strongly influenced by the removal of lipids. It could be shown that defatting of flour causes a decrease in the yield of high molecular components in the course of AUC-extraction. The examination of a glutenin prepared by extraction under reducing conditions (0,2% NaHSO3 in 60% propanol-2) has revealed that there is evidence for the participation of gliadin-like subunits in the glutenin structure.
