    Notes: Abstract We were intrigued by reports of the inhibition of phospholipase A2 (PLA2) by indomethacin. In order to increase the potency of the indomethacin system as an inhibitor of PLA2, it was decided to make more lipophilic analogs. Indeed, covalent attachment of a quinoline ring to the methoxy substituent of indomethacin affords WAY-122,220 which is almost an order of magnitude more potent than indomethacin in inhibiting human synovial fluid PLA2 (IC50=15 and 145 μM, respectively). TheN−p-chloro-benzyl analog of this compound, WAY-121,520, was an even more potent inhibitor of PLA2 (IC50=4 μM). Structural analyses and molecular modeling suggest that these compounds may inhibit PLA2 by mimicking arachidonic acid. WAY-121,520 is also a potent leukotriene biosynthesis inhibitor both in the rat PMN and mouse macrophage assays (IC50=10 and 4 nM, respectively), possibly acting via a 5-LO (5-lipoxygenase) translocation inhibition mechanism. The multiple actions of WAY-121,520 may contribute to its favorable anti-inflammatory profile.

    Notes: Summary Compared to poly(methyl methacrylate) (PMMA) or other poly(n-alkyl methacrylates), the thermal degradation of poly(2-hydroxyethyl methacrylate) (PHEMA) is rather complex, whereby the generated monomer (HEMA) is accompanied by a variety of other pyrolysis products. Besides HEMA, the dominating compounds were methacrylic acid and acetaldehyde, which could be established by gas chromatography.

    Notes: Summary The effectiveness of gene probe methods for tracking genetically engineered microorganisms (GEMs) in the environment was tested by inoculating nutrient-supplemented freshwater microcosms withAlcaligenes A5 (a naturally occurring 4-chlorobiphenyl degrader) orPseudomonas cepacia AC1100 (a genetically engineered 2, 4, 5 T-degrader) and following the fates of the introduced bacterial populations. Colony hybridization of the viable heterotrophic bacterial populations and dot blot hybridization of DNA recovered from the total microcosm microbial communities showed persistence of bothAlcaligenes A5 andP. cepacia AC1100 in the microcosms in the presence and absence of the xenobiotic substrates that these organisms biodegrade. Although there was a gradual decline in the added populations, both of the bacterial populatins were still detected in the microcosms two months after their introduction into the microcosms. Addition of 2, 4, 5-T enhanced the survival ofP. cepacia AC1100 — and 4-chlorobiphenyl addition resulted in increased levels ofAlcaligenes A5. The results indicate that both organisms may persist for very long periods in freshwater habitats.

    Description / Table of Contents: In addition to pyrolysis and degradation with superheated steam the hydrogenolytic degradation of cured phenolic resins induced by hydrogen-donors has been investigated. Especially with 1,2,3,4-tetrahydronaphthalene (tetralin) a considerable amount of transferred hydrogen to the dispersed resin particles, involved with formation of naphthalene, has been observed at 400°C. With total maximal yields of 98 wt.-% (ref. to the initial amount of phenolic resin) of phenol, methyl phenols, and oligomerous phenolic degradation products a recovering of industrial phenolic resin wastes should be possible. Kinetic and scanning electron microscopic investigations are employed to give some mechanistic explanations involving the hydrogenolytic liquefaction of cured phenolic resins.
    Notes: Neben der Pyrolyse und dem Abbau mit überhitztem Wasserdampf wird insbesondere die hydrogenolytische Spaltung ausgehärteter Phenol-Formaldehyd-Harze in Gegenwart von Wasserstoff-Donatoren untersucht. Als H-Donor wird hierbei 1,2,3,4-Tetrahydronaphthalin (Tetralin) eingesetzt, das bei Temperaturen um 400°C unter Wasserstoff-Transfer an die dispergierten Harzpartikel zu Naphthalin rearomatisiert. Bei maximalen Ausbeuten von insgesamt 98 Gew.-% (bez. auf die eingesetzte Harzmenge) Phenol, Methylphenolen und oligomeren phenolischen bruchstücken wird eine Nutzung technischer Phenolharzabfälle unter Gewinnung verwertbarer Spaltprodukte diskutiert. Anhand von Abbau-Kurven und rasterelektronenmikroskopischen Untersuchungen wird auf den Mechanismus der extraktiven hydrogenolytischen Phenolharzverflüssigung eingegangen.

    Description / Table of Contents: Zusammenfassung Zur chromatographischen Charakterisierung von Abbauprodukten ausgehärteter Phenol-Formaldehyd-Harze mit DC, GC, GPC und HPLC wurden die optimalen chromatographischen Bedingungen erarbeitet. Neben der Pyrolyse wird insbesondere der hydrogenolytische Abbau der vernetzten Kondensate näher untersucht, wobei eine Vielzahl von phenolischen, nichtphenolischen aromatischen und gasförmigen Spaltprodukten gebildet werden. Niedermolekulare phenolische Abbauprodukte können dünnschichtchromatographisch sowohl an formamid-imprägniertem Kieselgel HF254 mit dem Laufmittel Benzol/Methanol (95∶5 Vol.-T.), als auch an Kieselgel 60 F254-Fertigplatten mit Benzol/Diethylamin (85∶15 Vol.-T.) als Eluens befriedigend getrennt werden. Die qualitativen und quantitativen gas-chromatographischen Bestimmungen von Phenol, Kresolen, Xylenolen und anderen nichtphenolischen Aromaten wurden mit einer 2m×1/8 Zoll-Säule, gepackt mit 10 Gew.-% UCON LB-550X auf Chromosorb W NAW (60–80 mesh), durchgeführt; für die Gasanalysen wurde eine 2m×1/4 Zoll-Fertigsäule, gepackt mit einem 5Å Linde-Molekularsieb, eingesetzt. Die Molekulargewichtsverteilungen der Hydrogenolyse-Produkte wurden gel-chromatographisch an Merckogel 6000, einem mit Butandiol-1,4-divinylether vernetzten Poly(vinylacetat), und mit DMF als Eluens ermittelt. Eine alternative Analysenmethode zur chromatographischen Charakterisierung löslicher Phenol-Formaldehyd-Kondensate und niedermolekularer phenolischer Verbindungen ist die reversed phase Hochleistungs-Flüssigkeits-Chromatographie, wobei mit Spherisorb ODS als stationäre Phase und THF/H2O-Mischungen als Eluens eine weitgehende Trennung von Phenol, Methylphenolen und oligomeren phenolischen Spaltprodukten erreicht wird.
    Notes: Summary With chromatographic methods like TLC, GC, GPC, and HPLC it is possible to characterize relatively complex mixtures of phenolic, non-phenolic aromatic, and gaseous degradation products of cured phenolic resins, which have been generated by pyrolytic and hydrogenolytic cleavage. The low molecular weight phenolic compounds were separated with good resolution by thin-layer chromatography on formamideimpregnated silica gel HF254 with the eluent benzene/ methanol (95∶5; vol.), or silica gel 60 F254 precoated aluminium sheets, using the eluent benzene/diethylamine (85∶15, vol.). The qualitative and quantitative gas chromatographic determinations of phenol, cresols, xylenols, and other non-phenolic aromatic compounds were accomplished with a 2 m×1/8 inch column, packed with 10wt.-% UCON LB-550X on Chromosorb W NAW (60–80 mesh), and a 2m×1/4 inch ready packed column with a 5Å Linde molecular sieve for gas analysis. Gel permeation chromatographic investigations with Merckogel 6000, a poly (vinyl acetate) cross-linked with butanediol-1,4-divinylether, as stationary phase and dimethylformamide as eluent, gave information on the molecular weight distributions of the hydrogenolysis products. In addition, good separations of oligomer phenolic compounds, methyl phenols, and phenol are achieved by high performance liquid chromatography in the reversed phase mode, using the stationary phase Spherisorb ODS and THF/H2O-mixtures as eluent.
