    Notes: Abstract Human melanocytes established in MCDB-153 culture medium do not express α1-, β1-, β-2 adrenoceptors without extracellular stimulation. The addition of 50×10−9 M norepinephrine to the medium causes a time-dependent induction of α-1-adrenoceptors with 4.278 receptors/melanocyte after 24 h. Under the same experimental conditions, the dendricity of melanocytes as well as melanogenesis was unaffected over 60 h. Since keralinocytes hold the full capacity for catecholamine biosynthesis but melanocytes lack this system, the secretion of catecholamines from keratinocytes appears to be of critical importance to the α-1-adrenoceptor in melanocytes, underlining the symbiosis of both cells in the epidermal unit.

    Notes: Abstract Melanocytes were successfully established from involved and uninvolved skin of a patient with acute acrofacial vitiligo. Cells from involved epidermis showed a fivefold decrease in the rate of radiolabelled 45 Ca uptake compared with uninvolved and control cells. These results are similar to previous findings in keratinocytes from involved skin in patients with vitiligo. Since 6-biopterin is cytotoxic to melanocytes and calcium controls the redox status of the 6-biopterin /(6 R )5, 6, 7, 8-tetrahydrobiopterin equilibrium via the thioredoxin reductase/thioredoxin system, these results underline the importance of this electron transfer system for both melanocyte function and survival.

    Notes: Abstract Patients with atopic eczema have significantly higher norepinephrine levels in plasma than healthy controls. In addition, significantly higher levels of the essential cofactor (6R)-l-erythro-5,6,7,8-tetrahydrobiopterin (6BH4) were found in this patient group. Cell extracts from epidermal suction blister roofs revealed only half the normal activity of phenylethanolamine-N-methyl transferase (PNMT) together with a threefold induction of the norepinephrine-degrading enzyme monoamine oxidase A (MAO-A). Taken together, these results support earlier observations of a defective catecholamine/adrenoceptor signal in patients with atopic eczema.
