    Notes: Summary The aim of the present study was to characterize theological and immunological features involved in the pathogenesis of vibration-induced white fingers (VWF). Plasma viscosity, at two shear rates (580 s−1 and 1164 s−1), levels of immunoglobulins (IgG, IgM, IgA), circulating immune complexes, rheumatoid factor, antinuclear antibodies, fibronectin, fibrinogen, hemoglobin and erythrocyte sedimentation rate were analysed in 30 male dockers with VWF and in 30 healthy male referents unexposed for hand-arm vibrations. Decreased plasma viscosity was observed among the men with VWF, although formal significance (P 〈 0.05) was only obtained at shear rate 580 s−1. The decrease was mainly seen among smokers. In the study there were no significant differences between the VWF group and the referents with regard to immunoglobulin levels, autoantibodies and other plasma proteins. From the study it is concluded that workers with VWF may have a decreased plasma viscosity. The biological relevance of this observation is uncertain and deserves further study.

    Notes: Abstract The local pattern of proinflammatory cytokine release was studied in Alzheimer disease (AD) and vascular dementia (VAD), by measuring intrathecal levels of IL-1β, IL-6, TNF-α, and its naturally occurring antagonists, soluble TNF receptors I and II. The cytokine levels were related to neuronal damage, as measured by the intrathecal tau concentration, to cerebral apoptosis assessed by levels of Fas/APO-1 and bcl-2, and to clinical variables. In vitro analysis was performed to study the effect of TNF-α on the production of bcl-2, an antiapoptotic factor, by human neuronal cells. Patients with both AD and VAD displayed significantly higher intrathecal levels of TNF-α compared to controls. In addition, patients with AD showed significantly negative correlations between the intrathecal levels of TNF-α and the levels of Fas/APO-1 as well as of tau protein. The level of bcl-2 in supernatants of TNF-α-exposed cultures of human neuronal cells was up to three times higher than in control supernatants. Our study demonstrates intrathecal production of TNF-α in patients with dementias, suggesting that this cytokine may have a neuroprotective role in these neurodegenerative conditions as evidenced by negative correlations between this cytokine and (i) levels of intrathecal Fas/APO-1 and (ii) levels of tau protein, both parameters closely related to brain damage. Our in vitro data suggest that TNF-α exerts its neuroprotective effect by stimulating neuronal cells to express bcl-2, a molecule which downregulates apoptosis.

    Notes: Our objective was to examine the ability of nucleate and anucleate fragments of artificially activated mouse eggs to transform sperm nucleus into male pronucleus. To this end, zona-free oocytes in metaphase II were activated by ethanol and bisected into halves (one with the spindle, the other anucleate) either within 10 to 20 min (series A) or 3 or 5 hr later (series B). In series A, the fragments were inseminated 3,5, and 8 h after activation, and in series B. 3 and 5 h after activation. Both nucleate and anucleate fragments lose the capability of transforming sperm nucleus into fully formed pronucleus sometime between 3 and 5 h after activation. In 8 h old parthenogenetic fragments, the majority of sperm nuclei remain unchanged or begin decondensation but never reach the stage of an early pronucleus. In over 1/3 of anucleate fragments of this age group, sperm nuclei develop defectively: chromatin decondenses inside the persisting nuclear envelope. In other experimental groups, the incidence of these abnormal sperm nuclei varies between 0 and 10%. In general, the anuclcate fragments retain the capability to transform sperm nuclei (fully or partially) longer than their nuclear counterparts. This difference may be accounted for by a different level of substances required for pronuclcar growth (extrachromosomal constituents of the germinal vesicle and nuclear lamins): high and constant in the cytoplasm of anucleate egg halves and low and progressively decreasing in the nucleate halves because of their putative uptake by the female pronucleus. However, the cytoplasmic factors responsible for the initial stages of transformation (nuclear envelope breakdown, chromatin decondensation) become eventually inactivated both in the presence and in the absence of a female pronucleus.
