    A rapid method for the isolation of L-cell surface membranes using an aqueous two-phase polymer system (1971)

    Notes: Summary A dextran-polythylene glycol aqueous two-phase system has been used to separate cell surface membranes from other cellular organelles. The surface membranes have been identified on the basis of morphology, content of Na+, K+-ATPase, and presence of surface antigen as detected by a51Cr release method. Contamination of the surface membrane preparations by smooth endoplasmic reticulum, mitochondria, and nuclei has been found to be minimal. An average of 6.5% of the total protein was found in the membrane fraction. Less than two hours is required to isolate the membrane fraction after preparation of a Dounce homogenate. Fractionation by aqueous two-phase polymer systems appears to be a rapid and effective method for the isolation of surface membranes.

    Notes: Mice made tolerant to allogeneic tissues in neonatal life have been examined at different times for their ability to respond to the tolerizing determinants in a variety of assays (in vitro CML, MCL and in vivo GvH assays). All animals were tolerant in terms of their inability to produce CTL to the relevant determinants, and to induce GvH in lethally irradiated F1 recipients. Nevertheless, some mice also showed a normal MLC proliferative response and contained antigen-specific serum inhibitory factors, while other mice contained apparently antigen-specific suppressor cells. The pool of the latter, furthermore, was expanded considerably upon adoptive transfer of tolerant cells (with tolerizing antigens) to lethally irradiated syngeneic recipients. The data are compatible with the notion that suppression of clonal expansion represents the primary mechanism of tolerance maintenance (induction), and that the infrequently observed serum reactivity in such tolerant mice represents a vestige of the means whereby cell-mediated suppression was induced.

    Notes: Abstract The purpose of this paper is to examine uses of quality of life (QOL) measurements at different levels of decision making within the health care system, ranging from the micro (clinical) level, through the meso (agency, institutional or regional) level to the macro and meta (governmental) levels. We use individualized, group and population-based QOL and preference assessments as illustrative examples of ways in which QOL information and decision making level interact. We conclude that the meso and macro levels pose particularly challenging problems, and suggest that, if the primary emphasis is placed on applications of QOL assessments at the micro (clinical) level of decision making, a research agenda that is much too limited may be adopted.

    Notes: Abstract The relationship between quality of life (QOL) assessments and decision making, in relation to the delivery of health services, is subjected to critical appraisal. Three levels of decision making in the health care system are taken into account in the analysis. Criticisms of opinion polling provide the basis for the appraisal. Examples of criticisms considered are: Might the use of QOL information be manipulative? Could the interviews or questionnaires used to obtain QOL data influence personal opinions? Are the methods used sometimes defective and/or superficial? Will QOL information always be used in decision making in ways that are ascertainable and justifiable? It is concluded that the time has come for the main focus of critical appraisal in QOL research to shift, from an emphasis on evaluation of the quality of methods used for assessments of QOL, toward an emphasis on the practical usefulness of QOL data.

    Notes: Abstract Stategles and processes for the effective dissemination and diffusion of research findings on healthrelated quallty of life (HQL) into practical applications were explored at a workshop held in Montreal in April, 1994. A conceptual model for dissemination and diffusion of evidence about HQL was used to identify five different target groups: HQL assessors, policy makers, planners of clinical trials, developers of clinical practice guidelines, and those at the level of patient-practitioner clinical decision making. A framework designed to facilitate the translation of fundamental research knowledge into effective action was then used as a starting point for discussions about priorities for research on HQL. The most important problem identified at the workshop was how best to strike an appropriate balance between priorities deemed to be important to those directly involved in HQL research, and priorities deemed to be important to potential users of the results of HQL research?

For better or worse, the Internet has arrived as a force in our lives, and medical information and support constitute two of its more important uses. Both authors agree that the novel medium of the Internet provides a unique opportunity to create a true partnership of health care providers and consumers.
