    Notizen: We present the results of femtosecond optical heterodyn detected optical Kerr effect (OHD-OKE) spectroscopy and low frequency Raman scattering spectroscopy for three thiophene homologues (C4H4X, X=O, S, and Se). The observed OHD-OKE transients were analyzed both in time and frequency domains. Fourier transformed OHD-OKE data were directly compared with the corresponding spontaneous Raman scattering spectra. The reorientation lifetimes for furan (O), thiophene (S) and selenophene (Se) were determined to be 0.9, 2.0 and 3.2 ps, respectively, and were fairly explained in terms of the Stokes-Einstein-Debye relation. Their subpicosecond dynamics was analyzed with an antisymmetric Gaussian function and a skewed function developed by Bucaro and Litoviz, which gave good curve fits to observed data. Librational and translational dynamics of the molecules was discussed in terms of microscopic environments formed by neighboring molecules and the results of intermolecular interaction energy calculations by an ab initio molecular orbital method. © 1998 American Institute of Physics.

    Notizen: Abstract: In the present study, we compared bone regeneration ability in sinus floor elevation between a tissue engineering method using mesenchymal stem cells (MSCs) and platelet-rich plasma (PRP), and a promising new method using particulate cancellous bone and marrow (PCBM) and PRP. Bilateral sinus floor elevation procedures were performed in 18 adult Japanese white rabbits. MSCs/PRP or PCBM/PRP complexes were grafted to each maxillary sinus in the same rabbits. The MSCs were isolated from rabbit iliac crest marrow, and PRP was obtained from peripheral blood. PCBM were collected from the rabbit iliac crest and mixed with PRP. The animals were sacrificed at 2, 4, and 8 weeks after transplantation, and the bone formation ability of each implant was evaluated histologically and histometrically. According to the histological observations, both sites (MSCs/PRP and PCBM/PRP) showed well newly formed bone and neovascularization at 2 and 4 weeks. However, at 8 weeks, the lamellar bone was observed to be occupied by fatty marrow in large areas in both sites. There was no significant difference in bone volume or augmented height between MSCs/PRP and PCBM/PRP groups each week, but there were significant differences in bone volume and augmented height between 2 and 8 weeks in PCBM/PRP or MSCs/PRP groups and in bone volume between 4 and 8 weeks in the PCBM/PRP group (P〈0.05). These results suggest that the MSCs/PRP complex may well be used for bone regeneration in sinus floor elevation, compared with the PCBM/PRP complex.

    Notizen: Abstract: The present study investigated a correlation between osseointegration in dental implants and an injectable tissue-engineered bone, using mesenchymal stem cells (MSCs) and platelet-rich plasma (PRP). Initially, the teeth in the mandible region were extracted and the healing period was 1 month. Bone defects on both sides of the mandible were prepared with a trephine bar. The defects were implanted with graft materials as follows: PRP, dog MSCs (dMSCs), and PRP, autogenous particulate cancellous bone and marrow (PCBM), and control (defect only). Two months later, the animals were evaluated by histology, and at the same time dental implants were installed. Two months later, the animals were sacrificed and nondecalcified sections were evaluated histologically and histometrically. According to the histological observations, the dMSCs/PRP group had well-formed mature bone and neovascularization, compared with the control (defect only) and PRP groups, as was the same for the PCBM group. A higher marginal bone level was observed around implants with PRP, PCBM, and dMSCs/PRP compared with the control. Furthermore, the values describing the amount of bone–implant contact (BIC) at the bone/implant interface were significantly different between the PRP, PCBM, dMSCs/PRP, and control groups. Significant differences were also found between the dMSCs/PRP and control groups in bone density. The findings of this experimental study indicate that the use of a mixture of dMSCs/PRP results in good results such as the amount of BIC and bone density comparable with that achieved by PCBM.

    Notizen: This study aimed to investigate the effect of hyperbaric oxygen (HBO) therapy on the tissue reactions to commercially pure (c.p.) titanium implants placed in free autogenous bone graft by a 1-stage procedure. Eighty c.p. titanium implants were placed in the bone grafted from iliac crest to bilateral mandible of 40 Japanese white rabbits without tapping. Twenty rabbits underwent daily a HBO treatment for 60 min under 2.4 ATA during 20 consecutive days and the other untreated 20 rabbits served as controls. The implants with surrounding bone tissue were retrieved 20, 30, 60, 90 and 120 days after surgery, fixated, dehydrated and embedded in resin. About 20 μm thick ground sections were prepared prior to microscopical observations. The bone area and the bone-to-implant contact inside the threads were calculated separately in the grafted bone and in the host bone for each implant. After 30 and up to 120 days, the HBO treated group showed more bone-to-implant contacts in the grafted bone as compared to the non-HBO treated group. In the host bone there were no differences observed between HBO and non-HBO treated groups. This study indicated that HBO treatment was beneficial for the tissue incorporation of c.p. titanium implants when placed immediately in free autogenous bone grafts.

    Notizen: Abstract Background A comparison of chemotherapy with PP (cisplatin, CDDP; and peplomycin, PEP) and TPP (4′-O-tetrahydropyranyladriamycin: THP-ADM, CDDP and PEP) in the treatment of oral cancer. Methods This study included 159 out of 199 patients who had visited the collaborating institutions' hospitals, and who had been registered in the Examination Meeting of Chemotherapy for Oral Cancer during the 2-year period from June 1990 to May 1992. Ninety-seven patients underwent PP therapy and 62 underwent TPP therapy. In PP intravenous infusion therapy, 60–80 mg of CDDP per m2 was administered on day 1, followed by infusion of 5–7.5 mg of PEP per m2 from day 2 until day 6. In the PP intra-arterial infusion therapy, in which the drugs were infused in a retrograde fashion into the superficial temporal artery, 50–70 mg of CDDP per m2 was infused on day 1, followed by infusion of 5 mg of PEP per m2 from day 2 until day 6. In both the intravenous and intra-arterial TPP therapy regimens, 20 mg of THP-ADM per m2 was infused on day 1, followed by infusion of 50 mg of CDDP per m2 on day 2, and infusion of 5 mg of PEP per m2 from day 3 until day 7. Results The response rate of TPP therapy was 64.5%, which was not significantly different from the 53.6% obtained with PP therapy. However, the complete response (CR) rate with TPP therapy was 22.6%, significantly higher than the 9.3% with PP therapy. The response rate of TPP intra-arterial infusion therapy was particularly high (92.3%), and significantly higher than that of PP therapy. The response rate of TPP intra-arterial infusion therapy was very high (100.0%) in stage I and II patients and in primary cancers of the tongue, gingiva, buccal mucosa, and hard palate. The toxicity of TPP and PP therapies resulted in a high incidence of nausea/vomiting and anorexia. Skin disorders and epilation were observed at high rates with TPP therapy, and incidences were particularly high (100.0%) in the TPP intra-arterial infusion therapy group. The incidence of leukopenia was high in patients treated with TPP therapy but was not severe. Conclusion The results of this study suggest that TPP therapy is more effective than PP therapy in the treatment of oral cavity cancer. Although TPP therapy was associated with skin abnormalities, alopecia, and leukopenia as side effects, there was no influence on subsequent treatment. TPP intra-arterial infusion therapy appears promising in the treatment of oral cancer.

    Notizen: Abstract In acinar cells freshly dispersed from rat parotid glands, the effects of ammonium chloride (NH4Cl) on membrane currents were studied using the whole-cell clamp method. When membrane currents were recorded with command pulses to 0 mV, applied at 2-s intervals from a holding potential of −70 mV, NH4Cl (5–20 mM) transiently decreased outward currents and then slowly increased both outward and inward currents. After reaching a peak in about 40–50 s, both outward and inward currents gradually decreased in the presence of NH4Cl and, on its wash-out, the currents returned to the control level. Butyrate (5–20 mM) had little effect on the resting membrane currents, but markedly inhibited the response to NH4Cl. Tetraethylammonium (5 mM) strongly reduced both the resting and NH4Cl-induced outward currents, whereas it slightly potentiated the NH4Cl-induced inward current without affecting the membrane current at the holding potential. Without ATP in the patch pipettes, carbachol-induced membrane currents were relatively resistant to Ca2+ removal from the external medium, but NH4Cl-induced currents were quickly abolished in the absence of Ca2+. We conclude that intracellular alkalinization with NH4Cl increases Ca2+ influx and activates Ca2+-dependent outward K+ and inward Cl− currents.

    Notizen: Abstract In freshly dispersed rat parotid acinar cells, 10 μM carbachol increased outward currents at 0 mV and also inward currents at −70 mV recorded with the whole-cell clamp method using patch pipettes containing 1 mM EGTA. When EGTA in the pipette was increased to 2.4 mM, carbachol increased only outward currents and a further increase of EGTA to 4 mM blocked the carbachol response. Effects of changes in external K+ and Cl− concentrations suggested that outward currents were carried by K+ and inward by Cl−. Effects of Ca2+ removal from the medium differed between experiments with 0 and 5 mM ATP in the patch pipettes. When pipettes contained no ATP, responses evoked by repeated applications of 10 μM carbachol (0.5–1 min) at 1.5–4 min intervals decreased only slowly after Ca2+ removal, outward currents being reduced to 90±6% and inward currents to 47±11% (n=6) in 10 min. On the other hand, when 5 mM ATP was included in the electrodes, Ca2+ removal abolished the carbachol responses in about 5 min (n=4). It was also found that tetraethylammonium (5 mM) strongly reduced both currents, by blocking muscarinic receptors, while Ba2+ (2.4 mM) inhibited only the outward K+ current.
